Immunodeficient mice reconstituted with a human immune system represent a promising tool for translational research as they may allow modeling and therapy of human diseases in vivo. However, insufficient development and function of human natural killer (NK) cells and T cell subsets limit the applicability of humanized mice for studying cancer biolo...

Protective immunity against pathogens depends on the efficient generation of functionally diverse effector and memory T lymphocytes. However, whether plasticity during effector-to-memory CD8+T cell differentiation affects memory lineage specification and functional versatility remains unclear. Using genetic fate mapping analysis of highly cytotoxic...

Despite scientific evidence supporting the fact that vaccines are fundamental tools for preventing infectious diseases, a percentage of the population still refuses some or all of them. Vaccine hesitancy has become a widespread issue, and its complexity lies in the great variety of factors that can influence decisions about immunization, which are...

Question
Hello,
Does anybody here have experience with staining IL-10 receptor on T cells? Preferably for flow cytometry analysis.
I tried on splenocytes from a steady state mouse for 40min at 37°C or 4°C, but it doesn't show me any proper staining.
Any recommendations would be highly appreciated!
Best,
Valerie
Question
Hello,
I am looking for any anti-mouse clones of either CD4 or Thy1.1 antibodies that I could use for sorting cells. I plan on transferring the cells right afterwards into an infected recipient, therefore the antibody should not cause depletion or block T cell responses.
According to my internet research, it doesn't seem promising for CD4 ABs ( https://www.bio-rad-antibodies.com/cd4-marker-antibody.html ), but maybe somebody here has different experiences.
Thanks,
Valerie
Question
Hello,
does anybody have recommendations for staining CD122 (IL-2Rb) on T cells for FACs?
I tried staining it for 30min at 4°C (SF staining) on cultured CD4 T cells using an aCD122 antibody from biolegend, but the signal barely differed from my FMO.
Could it help to starve the cells for IL-2 a couple of hours prior to the staining?
Would appreciate any input!
Thanks,
Valerie
