この論文は国立情報学研究所の電子図書館事業により電子化されました。 3価陽イオン(スペルミジン)により凝縮した1分子DNAの力学応答を測定するとともに,蛍光色素(YOYO-1)でDNAを可視化し伸張時の脱凝縮過程を観測した.凝縮状態ではDNA上に蛍光強度が高い部位が観測され,その強度は伸張にともない減少する.蛍光強度の減少は,伸張による凝縮部位の脱凝縮を示唆している.また,色素の結合によりDNAの全長は増加するが,スペルミジン濃度の増加にともない再び減少する.これは,1分子DNA上で色素-スペルミジンの対イオン交換が生じていることを示している.
