Fish suffer, from bacteria, fungi, virus and parasites or by physical ailments. Gurami (Osphronemus gouramy), nila (Oreochromis niloticus), carp (Cyprinus carpio), catfish (Clarias sp.) were the most reported infections caused by Aeromonas are bacterial hemorrhagic septicemia or Motile Aeromonas Septicemia (MAS). Antibiotics are drugs of natural or synthetic origin that have the capacity to kill or to inhibit the growth of micro-organisms included MAS. However, the use of antibiotics in the long term can cause negative impacts, among others, feared the occurrence of bacterial resistance in certain antibiotics. The results showed five of isolates were sensitive to antibiotics of chloramphenicol, gentamycin, oxytetracycline, cefradoxil and nalidixic acid but resistant to vancomycin colistin sulphate, rifampisin, cephalosporin and novobiocin.
High market demand of mud crab (Scylla serrata) encourages farmers to increase the production of mud crab. However, mud crab can not synthesize essential amino acids, so it is necessary to supply essential amino acids such as lysine and methionine in the diet. This study aims to determine the effect of lysine and methionine on feeds to increase growth and retention of mud crabs (Scylla serrata). In this study the amount of lysine amino acid and methionine added to the trash fish diet were: P0 (0: 0%); P1 (0.75: 0.75%); P2 (1: 1%); P3 (1.25: 1.25%); P4 (1.5: 1.5%) with the ratio of lysine and methionine 1: 1. The parameters observed in this study were Survival Rate (SR), Specific Growth Rate (SGR), Feed Conversion Ratio (FCR), Efficiency Feed (EF), protein retention and energy retention. The results of the 35-day maintenance study showed significant differences (P <0.05) against Specific Growth Rate (SGR), Feed Conversion Ratio (FCR), Efficiency Feed (EF), protein retention and no significant effect (P> 0.05) on energy retention and Survival Rate (SR) on mud crab. The best results in this study were found in P4 treatment with addition of lysine amino acids and methionine (1.5: 1.5%).
Ecopath model approach was used to describe trophic interaction, energy flows and ecosystem condition of Tangerang coastal waters. This model consists of 42 ecological groups, of which 41 are living groups and one is a detritus group. Trophic levels of these groups vary between 1.0 (for primary producers and detritus) to 4.03 (for tetraodontidae). Groups with trophic levels 2≤TL<3 and 3≤TL<4 have a range of ecotropic efficiency from 0 to 0.9719 and 0 to 0.7520 respectively.The Mean transfer efficiency is 9.43% for phytoplankton and 3.39% for detritus. The Mixed trophic impact analysis indicates that phytoplankton havea positive impact on the majority of pelagic fish, while detritus has a positive impact on the majority of demersal fish. Leiognathidae havea negative impact on phytoplankton, zooplankton and several other groups. System omnivory index for this ecosystem is 0.151. System primary production/respiration (P/R) ratio of Tangerang coastal waters is 1.505. This coastal ecosystem is an immatureecosystem because it hasdegraded. Pedigree index for this model is 0.57. This model describes ecosystem condition affected by overfishing and antropogenic activities. Therefore, through Ecopath model we provide some suggestions about the ecosystem-based fisheries management.
This study aims to determine the bacterial pathogens that cause disease of the gourami in Blitar (East Java) and Yogyakarta (Central Java), Indonesia. A total of 50 fish samples taken randomly gourami in pond farmers in seventh different locations. There were 18 isolates were isolated and then test Koch's postulates were injected 0.1 ml/fish intraperitoneally to gourami. Characterization is done by using the biochemical tests. Pathogenicity test carried out on 3 isolates of Aeromonas spp. with intraperitoneal injection at a dose of 10⁴-10⁸ CFU/fish, the value of Lethal Dosage 50 (LD50) using the method Dragstedt Behrens. After the treatment, spleen and kidney samples were processed for histopathological analysis. The all of identified bacteria were 5 isolates Aeromonas hydrophila. Isolates of A. hydrophila in a row AH3 was virulen to gourami with LD50 (4.53 x 10⁶ CFU/fish), while isolate AH4 and AH5 (2.903 x 10⁸, 1.319 x 10⁹ CFU/fish) not be avirulen. Koch's postulates; 3 isolates are pathogenic with mortality of 40-100 % and 2 are non-pathogenic isolates with a mortality of 0 %. Clinically; ulcers, haemorhagic at the base of the fins, body, mouth and exophthalmia. Histopathologically indecated spleen necrosis, piknosis, necrosis and inflammatory cells in kidney.
The purpose of this research was to discover the process of enrichment of dissolved oxygen in fish cultivation media using nanobubble technology. This study was conducted with two treatments, namely a cultivation media without fish and a cultivation media containing 8 fish with an average body length of 24.5 cm. The results showed that the concentration of dissolved oxygen increased from 6.5 mg/L to 25 mg/L. The rate of increase in dissolved oxygen concentration for 30 minutes is 0.61 pp/minute. The rate of decrease in dissolved oxygen concentration in treatment 1 is 3.08 ppm/day and in treatment 2 is 0.23 ppm/minute. It was concluded that nanobubble is able to increase dissolved oxygen.
The purpose of this research was to analys immunogenic membrane protein as immunostimulant development material to control the mortality of white shrimp in traditional plus pond. This research was designed to use explorative experiment and experimental laboratory methods which used completed random sampling design. Collected data was analyzed with analysis of variance for examination of survival rate (SR), total haemocyte count (THC) and differensial haemocyte Count (DHC). The research divided into 2 part of riset: (1) Identification, cultivation Zoothamnium penaei, analysed of membrane protein by SDS-PAGE, (2) Field test protein membran on Survival Rate level, immune response (THC and/or DHC level) and infestation of Zoothamnium penaei in traditional plus pond. The result showed that there were seven bands membrane protein of Zoothamnium penaei with molecular weight 38 kDa, 48 kDa, 67 kDa, 71 kDa, 77 kDa, 98 kDa dan 104 kDa by using SDS-PAGE. Immunogenicity tested decrease by using ELISA and western blotting there are only found three bands with molecular weight 38 kDa, 48 kDa dan 67 kDa. The membrane protein could increase the immun respons and decrease the mortality, by subsequenly, it could increase the survival rate from 17% until 68% and pressured the parasite infestation of white shrimp.
Eel requires unsaturated fatty acids of linolenic acid for growth. Which can be supplied from earthworms. In this study, addition of earthworm in formulation feed aimsed to improve the fatty acid profile eel meat. This research used experimental method and randomized complete design method with five treatments. Each treatment was repeated four times. The use of earthworms in feeding treatment formulation was done for 21 days with different level i.e: 0 % (P0), 25 % (P1), 50 % (P2), 75 % (P3) and 100 % (P4). The result showed that the addition of eartworm significantly influenced the omega 3 contents (EPA & DHA) of eel meat.
The goal of this study was to detect of Aeromonas hydrophila carrying the hlyA gene in guramy by PCR assay. A total of 5 A. hydrophila strains were isolated from gouramy with different location and furthermore genotypic of all A. hydrophila strains havedetected by PCR assay for 16S rRNA gene. The primers used in the PCR targeted a 592-bp fragment of the hlyA gene coding for the hemolysin gene. Particularly hlyA genes are responsible for haemolysin toxins production in this genus. After gel electrophoresis, the amplicons from representative strains of the A. hydrophila were purified using extraction kit and were subjected to the DNA sequencing analysis. The results showed that: (i) the 592bp amplicon of the hlyA gene was detected in 5/6 of the A. hydrophila; (ii) the nucleotide blast results of hemolysin gene sequences of the strains of A. hydrophila revealed a high homology of 90-97 % with published sequences, and;(iii) the protein blast showed 95-98 % homology when compared to the published sequences. The PCR clearly identified the haemolysin-producing strains of A. hydrophila by detection in hlyA genes and may have application as a rapid species-specific virulence test.
Abundance of plankton in a waters is influenced by environmental parameters and physiological characteristics. Complex factors support phytoplankton growth. They are light intensity, dissolved oxygen, temperature stratification, salinity and availability of nitrogen and phosphorus. Nitrogen is an important element in the formation of chlorophyll, protoplasm, protein and nucleic acids. Observation of the three ponds in North Banyuwangi (Wongsorejo, Bangsring and Bulusan) and three ponds area of South Banyuwangi (Bomo, Jatisari and Badean). The observed parameters were NH4, NO2, NO3, PO4. Hidrogen peroxide effectively improve water quality, however it has a different effect in each different area.
