Introduction
I am an Assistant Professor at the Institute of Crop Science, University of the Philippines Los Baños, and the laboratory head of the Crop Breeding and Genetic Resources Laboratory. My research interests involve plant genetic resources conservation and management, cultivated plant systematics, crop breeding and other allied fields.

I am currently having conduct a study regarding the total phenolic content of a plant extract; however, I cannot determine if a 42.7 mg/g gallic acid equivalent phenolic content is high or not.

It is because I have very low RNA yield. I was thinking if it is because I use 1-week old freeze dried samples stored in the freezer at -80 degree celsius.
Amybe it is better to use fresh samples and then freeze dry it proceeding directly to RNA extraction.
Any tips in incresing RNA yield would be appreciated.
I am currently using  ReliaPrep™ RNA Miniprep Systems - Promega kit.

My hypothesis: Since nucleic acids has a peak absorbance at 260nm wavelength and proteins at 280nm, the ratio of greater than 2.0 is considered relatively pure RNA.
Twist: However, literature indicates that 260/280 ratio for relatively pure RNA is approximately 2.0. (What about having 2.4 or 2.5?)
Other concern: could a > or = to 2.0 also indicate purity from DNA contamination?
