Hi
Currently i'm using post-hoc tukey test to analyze the significant terms of each factor i used.
Temperature : 30, 40. 50, 60
Time : 5, 10, 15, 20, 25
To analyse the tukey test, for each factor, i fixed the parameter and varied another condition
(in e.g :
1: To analyse the significant of time (5-25 min) as a fixed temperature of 30
2: to analyse the significant of temperature (30-60) as a fixed time of 5 min
(for each different factor i change the value of responses)
ANOVA give significant (p<0.05) as a whole temperature, while insignificant (p>0.05) for time factor
However, for tukey-test expression the letter was different for each other.

The explaination of tukey test as followed.
for instance:
1) For the effect of extraction time at 30 and 60ºC, extraction time were not significantly different (p > 0.05) from each other.
For the effect of extraction temperature on TFC using UAE through tukey’s test analysis, its shows that at 5 min sonication, extraction temperature at 30ºC were significantly different (p < 0.05). At 15 min sonication, TFC at 30ºC and 50ºC were significantly different (p < 0.05). Meanwhile, at 25 min sonication, low temperature (30, 40ºC) were significantly different (p < 0.05)
my question.
1) does the way of my reporting data was suitable/right?
2) How to report if there is combination of two letter?.(e.g ab, abc, bc, d)
3) should i present the tukey test in separated graph of fixed parameter or tabulated data as attached?
I am planning to extract plant samples and analyse without evaporate the solvent.
as for DPPH analysis, it is needed to be in different concentration to get IC50. Thus, I am wondering on how to determine the concentration without evaporating the solvent.
for plant/herb sample preparation, can i re-dry the sample before extraction process to reduce the moisture content? (i have dried those before, unfortunately some of them are still 10-15% moisture content) (some of them are been grounded)
and is it ok to natural drying (for re-dry process), does it will affect the bioactive compound inside?
In deproteinization process of fish scale, the procedure said to have 1% of enzyme to be added into 10% (w/v) of fish scale, and I am confusing on how to prepare 1% of enzyme either is related into fish scale or volume of enzyme.

I would like to know, how I can study the effectiveness of an extraction procedure?
Is the solubility of material (analyte) to the solvent one of the criteria?
Also, either mass transfer coefficient and kinetic study, which one is most important to be studied for optimal design of the process?
