I am working on establishing a 30-marker panel for human FFPE skin tissue for Imaging mass cytometry (IMC). It is hard to find antibodies that are already tested on IMC. Most antibody providers do not test their antibodies for this application. I can look for antibodies that are described for Immunohistochemistry-paraffin (IHC-P) and try my luck after self-conjugating the metal isotope with antibodies. But the conjugation does not necessarily work for all antibodies. The cost can rise up quickly this way. Previous IMC studies would help but they are rare and the antibodies that worked for them are sometimes discontinued by the provider.
I am here to ask if somebody else is also currently working on IMC for human FFPE tissues and have tested some of the antibodies for the markers that are also my targets. T-bet is one of them. Others include, ROR gamma T, CRTH2, IL-17F, CCR7, CXCR6, CD56, CD94.
