Hello Every one, I am wondering whether southern blot can determine the mutated genes while they will be used as probes. Will it show result if the DNA fragment used as Probe has mismatches at intervals. Secondly, If the probe is 1KB long, but 700 bp is same, but 300 bp are not same on either of the sides. Will southern blot give the result or not? If yes, what is the possible solution to get a accurate band. Thank you.

Hello Everyone, I need to go through a question. From the theoried and laws, all the organisms having 2n number have a pair of genes, named as Allels of each other, here are 3 questions. 1. Do both of them having same sequesce (probably not)? 2. If we mask the effect of 1 gene, by silencing or knockout or gene manipulation techniques, what will happen to its allel, will it work or not? 3. Do allels work independent of each other, work dependently or in cooperative manner?

Hello,
I am working on some gene manipulation experiments; many times I faced a problem, When i t-clone my GOI(Gene of Interest) if give many colonies. When i run Colony PCR from the middle portion it gives exact sized positive result, but when I try to amplify full fragment, it gives nothing. When I mini-prep it and double digest it, I dont get any band of vector size nor fragment size. I did this about 10 times, What may be the problem? It is happened when I am using LA taq Takara.
