I order DNA from IDT company.
They mentioned the Tm at datasheet.
But when I measure by software or check the Tm from reference, I got different number. Which one do I have to follow?

I check the humidity in my lab, mostly the hygrometer shows 60-80 percent with temperature 20-30 C.
I have 100 uM of DNAzyme.
When I heat it up and cool it down, I still have the rest of volume inside PCR tube.
Then, may I reheat it for using in the next another day??
Previously, I made Pb(NO3) stock in miliQ water and diluted into buffer.
But I tried different way yesterday. I prepared 100 uM of Pb(NO3) in buffer 10 mM TA containing 50 mM NaCl, pH: 7.0-7.4
The lead powder is difficult to dissolve, then it was precipitated.
How to solve it??
If we want to fabricate LFA or uPaD, we need to prepare AuNPs-DNA reporter probe or using AuNPs-Antibody as a label (colorimetri).
Other solution that we have to prepare is Bovin Serum Albumin (BSA) for blocking step.
The question:
How long the stabilization all of them when we keep at 4 degrees Celcius?

Tube 1 --> Tris-HCl EDTA (TE 1X)
Tube 2 --> 10 mM Tris-acetate containing 50 mM NaCl
The color is change to purple (the parameter of aggregation).
How it becomes like that and how to prevent it??
Yesterday I synthesized 13 nm colloidal gold nanoparticles using hotplate stirrer (T= 300 C, s= 600 rpm), the solution boiled for a long time (until bubles out). Even it had already ~ more than 1 hours.
Do you have experience what is the temperature and the correct length of time for synthesis gold nanoparticles?
