We want to evaluate monocytes supernatant after 24-48 hrs of stimulation with LPS. The main question should we use PMBC or firstly remove lymphocytes and other cells by plastic adhesion (2-3 hrs and almost all monocytes be on plastic) and only then stimulate them by LPS?

We need to stimulate monocytes by LPS (1 ug/ml) with brefeldin A (10 ug/ml)  for 24 hrs. Can we use the whole blood protocol (http://www.bdbiosciences.com/documents/Detecting_Cytokines_Monocytes.pdf) only with 24 hrs incubation  or we need to use PMBC and so on?
