Hello: I am trying to measure NADPH oxidase activity in wheat roots and leaves and I cannot do it adequately. I measure activity y the supernatant from 15000rpm extracts, but I cannot detect any activity in plant microsomes that come from a 45000rpm centrifugation step, according to several published protocols. Can anyone suggest me the best method to measure this enzyme activity?
