Does anyone here know how to calculate the R2 statistic in DnaSP software? Or rather a reason for why I am not able to calculate the R2 statistic. It calculates all the other tests like Tajima D, Fu's F, Achaz's Y, Strobeck's S but it never calculates R2 for my dataset. Is there a bug in the software or have the developers removed this test? Any help is appreciated.

I am extracting host DNA from fecal samples. However, feces contains lot of microbes and their DNA too gets extracted during DNA extraction procedure.
I want to get rid/minimise the microbial DNA. It is causing a lot of issues in my PCRs (lots of misamplification). Is there an inexpensive method to do so?

I have nexus file (*.nxs) which contains a lot of taxa and the sequence length is also very long, so I cannot keep copy pasting it as it will take a lot of time. I want to know if there is any other way in which I can import or open this nexus file into MEGA 5.2 which can save a lot of time for me?
