Everytime I try I lose most of the sample and am left with bits to analyse. I use 4% paraformaldehyde fixative, wash steps are done with 0.05% tween 20 in PBS, blocking with 1% BSA for 1 hour. Previously I have gotten ok staining once but never again, especially in the thicker cell cultures.
