Introduction
I am both a biologist and an aquaculturist. I studies the cryobiology of aquatic organisms and the impact of cryopreservation protocols on cell ultrastructure and DNA integrity. I had years of experience in cryopreservation of semen, oocytes, embryos and embryonic stem cells from aquatic organisms. And actually apply the technique to Taiwan aquaculture industry. I examined the genetic structure of Formosa landlocked salmon and is trying to save the endangered species.

The assessment of morphology and immunology of specimens at different maturity stages is essential for the determination of spawning strategy and reducing the risk of mortality. In this paper, female and male Pacific abalones (Haliotis discus hannai) at different statuses (pre-spawning, spawning and post-spawning) were collected to estimate the eff...
Oncorhynchus masou formosanus (Formosa land-locked salmon) is a critically endangered salmonid fish endemic to Taiwan. To begin to understand how its drastic change in lifestyle from anadromous to exclusively river-dwelling is reflected in its immune genes, we characterized the genes encoding six cytokines (IL-2A, IL-2B, IL-4A, IL-4B1, IL-4B2, and...

We investigated the possible conditions to cryopreserve six marine microalgae that are frequently used in Taiwanese aquaculture: Amphora exigua, Isochrysis galbana, Nannochloropsis oculata, Seminavis gracilenta, Skeletonema costatum, and Tetraselmis elliptica. The approaches for microalgae cryopreser-vation with cooling from room temperature to À40...
We investigated the effects of cryopreservation on the quality of Portuguese oyster (Crassostrea angulata) sperm, which were examined before and after freezing; sperm motility, fertilizing capacity, and ultrastructural morphology were analyzed. The motility percentage and fertilizing capacity of the cryopreserved sperm (mean ± standard error) were...
In the present study, we have investigated the ultrastructures of the mature gonadal spermatozoa of R. variegata and T. literatus and presented comparisons with the Manila clam, R. philippinarum, sperm ultrastructure examined. Spermatozoa of R. variegata consist of (in anterior to posterior sequence): an elongate conical, deeply invaginated, acroso...
Spermatozoan ultrastructure and complete mitochondrial genome in the marine bivalve mollusk Meretrix sp. (Taiwan) from Taiwan are described and contrasted with other bivalves, especially within Meretrix. We have examined the features of the mature gonadal spermatozoa of Meretrix sp. (Taiwan) and provided comparisons with the other four Meretrix spe...
Climate oscillations during the Pleistocene had profound effects on the evolutionary history of freshwater fishes now distributed across northern temperate regions. The extent of continental glaciation on the western side of the North Pacific, including areas of East Asia, was more limited as compared with regions of North America, Europe and high‐...
Small abalone (Haliotis diversicolor) is a commercially valuable species for both fisheries and aquaculture. The production of annual farmed small abalone in Taiwan, once the highest in the world, has dramatically decreased in the past 15 years, and currently, the industry is close to collapse. Understanding the genetic diversity of small abalone a...

This study focuses on the strategy for the conservation of masu salmon, Oncorhynchus masou, in the northern part of the species range (via the masu populations in Sakhalin Oblast), based on data of its population structure. It is shown that masu populations that inhabit different rivers genetically differ from each other in allele frequencies at mi...
In the winter of 2007–2008, a large number of fish were killed by chilling damage that occurred in the Pescadores (Penghu Archipelago). Such extreme weather events can cause rapid population changes, which provide an opportunity to understand the possible restoration mechanism of coral reef fishes in the Pescadores. The fine-scale genetic structure...
The small abalone, Haliotis diversicolor, is an economically important shellfish species in East Asia. The wild stocks have been highly threatened due to overfishing in the past several decades. Taking into account conservation and recovery of wild resources, long preserved samples were used to analyze population structure. However, extreme fragmen...
The residual estrogenic pollutant alkyl phenols, 4-nonylphenol (NP), 4-tert-octylphenol (OP), and the sex ratio of Formosa landlocked salmon Oncorhynchus formosanus were surveyed in the Formosa landlocked salmon ecological reservation area. This species survives as a remnant, landlocked population in the headwaters of the Chichiawan Stream, a tribu...
The small abalone ( Haliotis diversicolor) is an important aquaculture species in southern China. Traditional breeding programs have been implemented for this species. But due to long reproduction cycle of the small abalone, the traditional method takes too long. There is therefore an urgent need to accelerate molecular breeding in H. diversicolor....
The use of hatchery-reared fish to replenish existing threatened wild populations has been shown to reduce or change the natural genetic diversity of the wild populations. In this study, the genetic diversity of wild Formosa landlocked salmon Oncorhynchus formosanus in its main habitat of the Chichiawan Stream in Taiwan was examined after a large-s...
This study sampled six times of river water, sediment, and tilapia (Oreochromis niloticus) in the Dan-Shui River, Taipei, Taiwan; 10 feminizing compounds were analyzed using ultra-performance liquid chromatography-tandem mass spectrometry. Bisphenol A (508 ± 634 ng/L, geometric mean (GM) 303 ng/L) and nonylphenol (491 ± 570 ng/L, GM 328 ng/L) were...
Single nucleotide polymorphisms (SNPs) are the most common type of genetic variation among individuals of a species. Recently, in spite of the development of high-throughput genotyping technologies, SNPs have been applied as markers for population genetic and high-density genetic mapping. However, the high costs of SNPs discovery and genotyping ass...
Peanut worm (Sipunculus nudus) is a cosmopolitan species mainly distributed in tropical and subtropical coastal waters. Analysis of the mitochondrial cytochrome c oxidase subunit I (COI) gene sequences among S. nudus from GenBank revealed high genetic variation (p-distance, 0.115-0.235; k2p, 0.128-0.297) and paraphyletic relationships. These indica...
Nuclear-encoded microsatellites and mitochondrial DNA (mtDNA) sequences were assayed from Cobias Rachycentron canadum sampled in waters offshore of Virginia (U.S. Atlantic), Mississippi and Louisiana (Gulf of Mexico), and Taiwan (Southeast Asia). Global exact tests and analysis of molecular variance revealed that Cobias from U.S. waters were homoge...
PTEN (phosphatase and tensin homolog on chromosome 10) is phosphatase with dual activities on different substrates are involved in tumor suppression and embryonic development. PTEN seems more prominent in a swift biological process (tumorigenesis, angiogenesis, folliculogenesis), PTEN has well-known roles as a negative regulator of the PI3K/AKT pat...
Papaya (Carica papaya L.) is established as a cash crop throughout the tropical and subtropical regions due to its easy adaptation to diverse agricultural conditions, high yields, and prompt returns. The sex types of papaya plants are hermaphrodite, male, and female. Among them, hermaphroditic plants are the major type in papaya production, because...

In this study, DNA analyses were employed to verify the identity of six morphologically similar species that occur in the coastal waters of Taiwan: the black seabream complex (Acanthopagrus latus, Acanthopagrus schlegelii, Acanthopagrus sivicolus, Acanthopagrus taiwanensis, Acanthopagrus chinshira and Acanthopagrus pacificus). Amplified fragment-le...
Mottled spinefoot Siganus fuscescens and white-spotted spinefoot Siganus canaliculatus are two similar species that differ subtly in colouration and morphology. Three major mtDNA clades were identified for these species, but individuals were clustered by amplified fragment length polymorphism (AFLP) according to geography rather than morphology, su...
In mammals, androgens appear to enhance the development of primary ovarian follicles, but PI3K (phosphoinositide 3-kinases) pathway is well recognized as one of the critical pathways in early follicular development. Roles of the PI3K were revealed by deletion of PTEN (phosphatase and tensin homolog on chromosome 10). PTEN is demonstrated to play an...
A few studies have evaluated demographic and genetic consequences of population subdivision by damming on the population as
a whole. Formosa landlocked salmon Oncorhynchus masou formosanus have persisted as a relatively large population that has recently been subdivided into seven subpopulations by erosion-control
dams and a natural waterfall. The...
Species and sex identification are among the most important parameters for conservation management. However, it is extremely difficult to perform such identification in Formosa landlocked salmon (Oncorhynchus masou formosanus). Both sexual dimorphism in landlocked dwarf form Formosa landlocked salmon and morphological difference among cherry salmon...
