Introduction
I am working as a professor of Food Science and Technology at the department of Food Science and Nutrition, King Saud University. Our research group is focus on the development of nutritious food products through fortification with natural bioactive from plant sources. We also focus on assessing the effect of domestic and modern processing methods on the quality of food products. In addition, our research is also directed toward food biochemistry and biotechnology.

We keep working for long time on antioxidant and bioactive properties of plant extracts however we some time miscalculate the accurate values my question is as follow:
We dissolved 2 g sample in 200 mL solvent, then we freeze-dried the extract
1 g of the extract was dissolved in 100 mL
0.2 mL extract was used for the assay of TPC and the absorbance was 0.602.
The standard curve equation was y=0.0005x + 0.039
How to accurately calculate the TPC of the extract and the original sample?
For flavonoids (TFC) we used 0.1 mL extract and the absorbance was 0.111
The equation was y=0.0004x + 0.0281
How to calculate the TFC?
I know that there were many posts about this issue but I need step by step analysis.
