I obtained 2 melting temperatures (75, and 82 C) for the same sample with specific confirmed primers (forward, and reverse) by RT-PCR. But, I have single Ct for each. Do you think there is a mutation or polymorphism within the detected gene?, or it may be a contamination knowing that, I applied the same samples with another primer pairs and I obtained single melting peak.

I have a substance that dissolve in at least 5% ethanol. I want to prepare a stock solution of this substance for further serial dilutions in 96 well plate for calculation of IC50 by MTT assay. knowing that, the conditin for ethanol concentration per well shouldn't exceed 0.4%.
