Introduction
In the Castro Lab, we are interested in understanding how microbial communities respond to disturbances, how they maintain diversity, and their role in ecosystem processes. To approach this, we apply molecular microbiology, high-throughput sequencing, computational biology, and field work on microbial communities in situ or in the lab.

Streptomyces sp. H-KF8 is a fjord-derived marine actinobacterium capable of producing antimicrobial activity. Streptomyces sp. H-KF8 was isolated from sediments of the Comau fjord, located in the northern Chilean Patagonia. I'd like to get plasmid DNA from a Sterivex unit containing bacterial cells from ocean samples. The purpose of this is to do a shotgun sequencing run but ideally on a DNA extract enriched for plasmid DNA. I could go brute force and get total DNA and then computationally identify DNA reads of plasmid origin but that would cost more money and analysis time than simply starting from a plasmid DNA extraction.
Any advice is appreciated

I've been trying unsuccessfully to clone a gene from one plasmid to another, and I'm starting to think maybe a company can do it for me. I have two plasmids that contain full HiV-1 genomes (infectious molecular clones). Plasmid B has an RT gene with drug resistance mutations that I'd like to replace into plasmid B. So far I know both plasmids have the necessary restriction sites to directionally insert the RT gene into the other. I can also see the vector and insert on both plasmids after double digestion. In fact, I presume something is wrong at the ligation step. I know it's a trivial problem but so far no success. Any help is appreciated.
