I would like to measure the cell sedimentation velocity in different fluids and I would like to find a simple a fast method to calculate this velocity.
I have two plasmid with compatible origin of replication but the same antibiotic resistance. Could it be a problem?
We work with a special plasmid in our group. But for a concrete experiment we need this plasmid with another origin of replication. Is it possible to change the BR322 origin by the SC101 origin? We will use Gibson assembly for that purpose.
I am unable to transform BLRs with a pSC101 origin plasmid. However, I am able to transform these bacterias with the same plasmid but with Ori as origen of replication.
