Prior to starting my PhD in Ecology, I talked with multiple trusted friends who have been or are close to being awarded their doctorates. I asked them how they found the PhD process hoping to get an idea as to whether a DTP was the right thing for me. Their words of wisdom were that a PhD is an extremely long, arduous process that will push your limits to and beyond the bitter edge. I thought to myself, if this is so, why would anyone embark on such a journey. For me, it stems all the way back to childhood. Exploring the world through the starey eyes of a child I was always engrossed by nature. I am about to start a lab-based experiment where some of the treatments of leaf litter require either bacterial or fungal exclusion and am wondering what the best (defined by most effective vs cost) bacteriocide/ fungicides are and why?
Looking through the literature, there are many slightly different techniques for fixing biological specimens using different pre-post fixatives, mainly paraformaldehyde, gluteraldehyde, cacodylate and osmium tetroxide followed by critical point drying but all of the papers use slightly different concentrations with little justification as to why. If anyone has access to a solid protocol for this it would be greatly appreciated.
As a part of my MSc by Research we will be utilising high throughput microplate assays following Jackson et al. (2013), a technique that utilises 4-methylumbelliferone (MUF) labled substrates.
In our lab we have some of the necessary model substrates ( 4-MUF-β-D-glucopyranoside and 4-MUF-N-acetyl-β-D-glucosaminide) but their condition is unknown.
Does anybody know if there is a way to determine if they are still fit for purpose as they are expensive to purchase and it seems like a waste of money to purchase more if they are still good to use.

The study aimed to determine what effect the Waterboxx had on both germination and survival compared to other, more regimented watering techniques and to assess the costs and benefits associated with each technique. The treatments were: the Groasis Waterboxx, initial deluge and regular watering with a control which received no supplementary water.
Develop a more detailed understanding of the underlying mechanisms of photodecomposition, focusing on the relationship between biotic and abiotic decomposition throughout the dry season and subsequent wet season.
