Rock varnish is the skinny outer layer of rock surfaces in arid and semiarid regions, it is composed of clay minerals cemented together by oxides and hydroxides of manganese and iron. Al-Jafer basin is located in the south Jordan in a region that classified as a arid and semi-arid region with mean annual rainfall of about 50 mm . The basin has a total area of 13500 km2, scattered with a plenty of rock varnishes. It is well known that microbial communities associated with desert environments play pivotal roles in various biogeochemical cycles.
