Saksagliptin, hipoglisemik (bir anti-diyabetik madde arasında) dipeptidil peptidaz-4 (DPP-4) inhibitörü olan ve ülkemizde de kullanılan oral bir ilaçtır. Bu çalışmada Saksagliptin’in insan lenfositleri üzerinde meydana getirdiği DNA hasarı ve sitotoksik etkisi incelenmiştir. Bu amaçla, günlük kullanım dozları (2,5 ve 5 mg) baz alınarak 0,017, 0,035, 0,07, 0,14 µg/mL’lik konsantrasyonlarda ökaryotik hücrelerdeki DNA hasarını hücre bazında saptayabilen duyarlı test metodu olan Komet Assay (KA); yine genotoksik ve anojenik etkinin değerlendirilmesi için Mikronükleus Testi (MN); sitotoksisitenin belirlenmesi için de hem 24 hem de 48 saatlik uygulamayla Mitotik İndeks (MI) testi yapılmıştır. Çalışmalarda kontrol grubu olarak pozitif ve negatif kontroller de dahil edilmiştir.
24 saatlik Saxagliptin maruziyeti ile mitotik indeks saatlik uygulamada sadece en yüksek konsantasyonlarda kontrole göre anlamlı farklılık göstermiştir. MN testi ile elde edilen sonuçlara göre MN frekansı sadece 0,14 µg/mL’lik konsantrasyonda kontrole göre istatistiksel olarak anlamlı fark oluşturduğu tespit edilmiştir. KA testinde ise kuyruk uzunluğunun istatistiksel değerlendirilmesi yapıldığında 0,035 ve 0,14 µg/mL’lik konsantrasyonlar kontrole göre istatistiksel olarak anlamlı bulunmuştur. Saxagliptin maruziyeti ile Komet kuyruk momenti ve kuyruk yoğunluğu sadece en yüksek konsantrasyon olan 0,14 µg/mL’de anlamlı bir şekilde artmıştır.
Sonuç olarak, çalışmada Saxagliptin Komet ve Mikronükleus testlerinde sadece piyasada kullanımı olmayan en yüksek konsantrasyonlarda istatistiksel olarak kontrole göre farklılıklar oluşturmuştur (kuyruk uzunluğu, 0,035 µg/mL hariç). Mitotik indeks testinde de benzer şekilde sadece yüksek dozlarda sitotoksik etki gösterdiği tespit edilmiştir.
