How to calculate concentration of 100uM ACETOSYRINGONE?
I need to prepare 100uM solution of acetosyringone for my co cultivation experimentsbut I'm confused in calculation of concentration, can someone please verify my calculations for 100uM acetosyringone? what amount of AS should be used to make 100ml of 100uM AS solution?

I have just started working with Acetosyringone. I would suggest you prepare 100mM stock solution by adding 1.920gm in 100ml of DMSO. Then you can add say like 10ul in 100ml Media to make your final concentration of AS to be 100uM.

1. Prepared 0.1M Actosyringone solution: (see link below)
2. To prepare 100 mL of 100 uM AS solution:
(1) 0.1 M stock = 100 mM = 100, 000 uM
(2) (100,000 uM) x (Y/(100 mL))= 100 uM
(3) Therefore, Y =0.1 mL = 100 uL.
(4) So, add 100 uL of stock solution to your 100 mL solution and you will get around 100 uM AS

I'm suggest you following the isntruction by Yuan-Yew Yau. Preparing 100 mL of 100 uM AS Solution is very hard. You need to weigh 1.96202 mg of AS powder on analytical balances. That's why you need to make 0.1 M stock solution.
100uM =0,0001 M
M=m/ma*v 0,0001=m/196,19*0,1 so it makes 0,0019619 gr
congr. Dont forget forget to solve it with DMSO.

I have 3 groups. 1. Control 2. Disease 3. Treatment. I want to lookup the gene expression btw these groups, compared with control (whether is upregulated or downregulated).
I did real-time qPCR and have ct values. I calculated ∆Ct = Ct[Target]-Ct[Housekeeping] ... and ∆∆Ct = (∆Exp.)-(∆Control) and got the -∆∆Ct log-fold-change. It looks all the values are almost same and not much different between the groups.
