To check for protein degradation pathways how to make a solution of chemical inhibitors?
I want to make a solution of 3-methyladenine and MG-132 that can be infiltrated into Nicotiana benthamiana. So, I tried dissolving in DMSO as per product guidelines and then diluted in PBS that resulted in inconsistent results. I would like to ask if anybody uses some other kind of solvents that resulted in reproducible results? P.S I am always making fresh solutions before using it.

Dear Sayanta,
As 3-MA is soluble in water at a relatively high temperature (>40C for 10mM solution)(https://www.sigmaaldrich.com/catalog/product/sigma/m9281?lang=de®ion=DE), I would suggest preparing a stock solution of around 0.1M concentration in water by warming to around 60C, then aliquoting and storing at -20C. Each aliquot will need to be redissolved at around 40C when required. Another Possible approach is to use cell culture medium like RPMI to dissolve it (at around 37 degree).
I found this suggestions very helpful and got them from this thread:
I hope that helped a little bit. All the best,
Babak

Background:
Alpha-synuclein aggregation is considered one of the main causes of Parkinson's Disease (PD). Malfunction of autophagy-lysosomal pathways is believed to be an underlying mechanism of α-synuclein aggregation. Although such malfunction has been observed in PD brains, it is unclear whether it may also occur in extraneuronal tissues.
