Kindly identify the symptom observed in Capsicum leaves?
- John Momo
Please help identify the possible symptom observed in Capsicum annuum leaves. From my observation, it doesn't look like it is pathological (my guess is a physiological symptom but i'm not sure of which).

How to dissolve precipitated total proteins (from fruits)?
- John Momo
Kindly assist me with suggestions on how to dissolve total proteins (from fruits) easily using appropriate buffers. I used resuspension buffer containing 0.5% SDS, 50 mM Tris-Cl and 100mM DTT but i still find it difficult to dissolve the pellets i obtained after precipitation, and washing with 80% acetone, resulting in major loss of proteins. I would be grateful. Thank you!

What happens if DTT is stored at room temperature?
- Martina Boncaldo
I used DTT in protein extraction for WB, but unfortunately it was stored at RT when in the label the storage temperature was 2-6°C. What are the consequences? What is DTT's role in protein extraction before WB? I got only one band after electrophoresis, do you think it's DTT's fault?
Now I have put DTT in the fridge for next extractions but I guess that is too late

How to prepare 5% Formic Acid in acetonitrile/water 1:1 v:v HPLC Mobile Phase?
- Shashikant Kotwal
Hello to whomever is reading this.
I am working on HPLC analysis of anthocyanins in sweet potato leaves and tubers.
One of the methods I have found requires a mobile phase solvent of "5% formic acid in acetonitrile/water 1:1 v:v".
Do they mean 5% formic acid in acetonitrile 1:1 v:v?
Also, how do I prepare 5% formic acid from 98-100% formic acid? Do I just use C1V1 = C2V2?
Thank you for your help!
Here is the article I am referring to:

Why random_state in train_test_split is equal 42
- Mohammed Y. Kamil
Whenever used Scikit-learn algorithm (sklearn.model_selection.train_test_split), is recommended to used the parameter ( random_state=42) to produce the same results across a different run.
why we used the integer (42)?
can we use another number?
Peptide desalting is important before doing inhibition assay for bacteria ?
- Marwa Nowir
i injected immunized hemolymph of larvae into FPLC (fast protein liquid chromatography)
to separate peptides from each other.
every peak of protein separated in a falcon tube in 5 ml of Sodium chloride buffer.
next step is lypholization , after samples were lypholized , protein powder plus sodium chloride crystal is precipitated .
should i do desalting for protein to have a pure protein or can i use it as a whole for inhibition assay fro bacteria??
