Hello, Dose any one has an idea about the DNA concentration of induced phage lysate to be able to see a clear band in PFGE?

I amplified single clone of phage and extracted the DNA from it. However, the DNA concentration is very low (around 2 ng/ul). Does anyone know how I can increase the DNA concentration?

After the RNA isolation from fresh colorectal cancer biopsies, I dose my RNA's concentration, and I find about 3000µg/ml. In RT, I take 5µl of my RNA at 1ug :

Ci= 3000 µg/ml

Cf= 200 µg/ml (1µg/5µl)

Vf= 50ul

then I found Ci= 3,3 µl from my RNA + 46,7 H2O

I would ask if someone knows if I can use a concentration of RNA higher than 1µg??

Polyvalency of phage

I am unable to store marine bacteriophages for a long time with an SM buffer at -4 C°, -20 C° and -80 C°. Please share the correct protocol for this.

I understood that to generate a standard curve for a certain bacteria, the Ct values should be plotted relative to corresponding serial 10-fold dilutions of template DNA of known concentration extracted from representative cultures. But, how can I then convert the DNA concentration I'll get (ng/ul) into CFU/gram of stool?

I have PCR product for sequencing and its too low concentration. How do I get more yield of PCR products. 

I am wondering if the QPCR method is sensitive to RNA? Do my samples need to be treated with RNAse? How sensitive is telomere QPCR to DNA concentrations; may differences across samples be negligible since I am standardizing with a control gene?

I think RNAse treatments may be more important for accurate quantification, though I am assuming there will not be telomere-like sequences that are transcribed. I am working with folks that use real time PCR for gene expression and some of their cautions that one has to consider in setting these experiments up.

Does anyone have experiences working with Human Domain Antibody Phagemid library constructed by Dr Daniel Christ (formally of the MRC Laboratory of Molecular Biology, Cambridge UK and currently based at the Garvan Institute, Sydney, Australia) which is provided by Source BioScience Inc.(united Kingdom) to generate specific monoclonal antibody in form of scFv. I’ve recently used this phage antibody repertoire and all steps carried out according to the protocol, but I didn’t get the correct and acceptable result in screening of scFv by ELISA.I want to know, does it need further modification in protocol and I would be so grateful if someone suggest me the person who has experience in this field to get in touch with him/her.
