Does FAK +/+ mice osteoblasts behave similar to wild mice osteoblasts?

Does anyone have good experience in this regard?

I want to know in vitro and in vivo.

Hello, everyone. One thing I just want to ask you, because I am a freshman in bone engineering. 

We all know that Mesenchymal stem cell could differentiate into osteocyte. In this process, Mesenchymal stem cell may first differentiate into osteoprogenitor, and then into pre-osteoblast, then into osteoblast, finally into osteocyte.

My question may state as following, "what is the important time point in Mesenchymal stem cell differentiation in every differentiation stage??" For instance, when Mesenchymal stem cell differentiate into osteoprogenitor or pre-osteoblast or osteoblast or osteocyte??

To assess the antiosteoporotic activity in vitro, In literature, many manuscripts explain the use of hFOB cells. We enquired at NCCS, they do not have.

The hFOB cells requires 32 degree for sub-culturing and maintaining. We have cell lab, but we grow cancer cells and normal cells which grow at 37 degree.  Is there any alternative to hFOB?

I'm looking to understand if there is a similar type of cell that is responsible for formation of ectoskeleton, or has a similar role to human osteoblasts in Drosophila.

I am carrying out in vitro digestion methods and using PEG as a liquid marker. However, I am unable to find a good relaible method for its determination in detail. Can someone please help me out. Thanks

I got PGD2 powder from Sigma-Aldrich ( P5172-1MG).

I have read works diluting it in 100% EtOH and DMSO. In my design the amount of any diluent will be far below toxic levels in vitro.

Still wanted to respectfully ask this Forum which one would be the better diluent considering that further 1:10 dilution with PBS will be done.

Should i compare the results of in vitro literature results with in vivo results in my discussion chapter?

I am planning in vitro experiment for Cas12 (cpf1). Can someone please suggest to me the procedure to design crRNA to target a gene in vitro. In silico methods available for the design can be shared.

When I feed a particular substance to pea plant, I see conversion to unknown compound.

I don't know, what is the product, I don't know (obviously), what is the reaction and I do not know required conditions (like cofactor(s) etc.).

I intend to identify the product, as that is the easiest thing to do right now, I guess. But I'm afraid that won't help me much, as I won't be able to specify the conditions.

Because any in vitro measurement/detection is ruled out because of the lack of knowledge about the reaction conditions, it seems to me like the only option is to go in planta. What I could come up with is either random mutagenesis or finding two genotypes with different activities and then trying to cross them.

However, for that I would need to have either reliable sensor to monitor amount of my compound in planta, or feed it to the plant and then isolate and measure on HPLC/MS (not very high-throughput).

I was also thinking about utilizing cDNA library, but so far, I did not come up with simple way to do so.
