Do you have experiance on VCG ?

I want plant Pathology practical manual related to fungi, like isolation, identification, different pathogenecity test etc. Can anyone please suggest it.

I tried to identify a fungus using the ITS region, results came back inconclusive. I now have the full genome sequenced, how can I use the results of the whole genome to determine what fungus it is and how can I find the ITS region within the genome in order to use it in future identification?

I have been trying to isolate fungi from soil and water. I have a challenge identifying them.

Our research work involves identification of Ganoderma spp. isolates using ITS region. We purified the PCR product and then sent for sequencing. Prior to PCR amplification, we cultured the fungus on PDA and extracted the mycelia DNAs using conventional method or available kits. However, we are now facing multiple signals problem in the acquired sequencing results. One way to counter this problem is cloning and this work is currently undertaken. Another primers for identification such as transcription elongation factor (tef) etc will also be used in the next PCR amplification.

Nevertheless, we would like to get any suggestion, recommendation or advice regarding this multiple signals problem from your experience and expertise.

Vegetative compatibility assay in fungi is a time consuming process for huge numbers of isolates, so is there any alternative molecular method for these assays? I think the best method may be use of hybridization with probe. What is your opinions?
