Can any one please assist in the identification of these fungi?

I need at least 5 articles on how to identify fungi on a petri dish at least up to the genus level.

I'm wondering if the mold is sufficient enough or you need to scrape off some parts(?) of it and place it on a slide and stain then observe it under a microscope.

If some require some identification kits please provide some links or manufacturers name o where to buy these

I want to prepare lactophenol cotton blue for fungi staining. But I have solid lactic acid. The protocol said to use 20 ml of lactic acid solution. question is -

1. What should be the percentage of the lactic acid in solution?

2. How to make it?

How can we calculate the exact number of the dead cells in a specific population of Candida albicans cells, with or without staining ? If a dye were used, which one is better ?

I am looking for a well performing red nuclear stain for microscopy in Candida albicans. Often dyes suitable for Saccharomyces work in Candida albicans...

I am specifically looking for a red nuclear stain because I am using other FP's and dyes in the blue and green channel...

Syto dyes did not work, they showed only a mitochondrial staining pattern....

Our research work involves identification of Ganoderma spp. isolates using ITS region. We purified the PCR product and then sent for sequencing. Prior to PCR amplification, we cultured the fungus on PDA and extracted the mycelia DNAs using conventional method or available kits. However, we are now facing multiple signals problem in the acquired sequencing results. One way to counter this problem is cloning and this work is currently undertaken. Another primers for identification such as transcription elongation factor (tef) etc will also be used in the next PCR amplification.

Nevertheless, we would like to get any suggestion, recommendation or advice regarding this multiple signals problem from your experience and expertise.

In 2019, in the city of Turin, Italy, I participated in the 4th international Molecular Mycorrhiza Meeting. In the exhibition of works, I saw a summary that the author used a method of root clarification with a red dye. This method seemed very interesting to me but I forgot to take note. Could someone tell me which one?

I would like to stain Mycobacterium tuberculosis using a fixable live/dead probe. I have some Ghost Dyes to evaluate its viability. Has anyone done this before? If so, could you share a protocol? Thank you!
