About the lab

Persian Gulf Institute established at 1997 in Persian Gulf University which named as Persian Gulf Research Center. At July 2013 Iranian council on the extension of higher education upgraded it to institute and named Persian Gulf Institute.

ساختار جمعيتي Epinephelus coioides، هامور معمولي، در خليج فارس از نظر 6 جايگاه ريزماهواره پلي مورف مورد بررسي قرار گرفت. 5-3 گرم از بافت نرمِ هر کدام از 120 نمونه صيد شده از 4 ايستگاه تهيه گرديد. استخراج DNA با استفاده از روش استاندارد استات آمونيوم انجام و کميت و کيفيت آن با روش هاي اسپکتروفتومتري و الکتروفورز بر روي ژل آگارز 1 درصد بررسي شد. قطعات DNA در دستگاه PCR تکثير شده سپس بر روي ژل پلي اکريلاميد رانده شد و رنگ آميزي با کمک نيترات نقره صورت گرفت. ميانگين پارامترهاي ژنتيکي شامل آلل هاي واقعي و موثر براي تمامي لوکوس ها و جمعيت ها، بترتيب 458/5 و 793/3، همچنين هتروزيگوسيتي مشاهده شده و قابل انتظار بترتيب 500/0 و 649/0 مي باشد. آزمون AMOVA بيشترين ميزان Fst، (086/0) و کمترين ميزان جريان ژني (652/2=Nm) را بين دو جمعيت خوزستان و بوشهر، کمترين ميزان Fst، (034/0) و بيشرين ميزان جريان ژني (070/7=Nm) را بين دو جمعيت دير و بندرعباس نشان داد. جمعيت خوزستان جدايي نسبي را در قياس با ساير جمعيت هاي مورد مطالعه نشان مي دهد، بنابر اين اعمال مديريت شيلاتي جداگانه توصيه مي گردد.
Population structure of Epinephelus coioides (Hamilton, 1822), Orange-spotted grouper, was studied based on six polymorphic microsatellite loci in the Persian Gulf waters. 3-5 gr of each individual's soft tissue were prepared through 120 hunted individuals of 4 sampling stations. DNA extraction was performed using a standard ammonium/acetate method. Quantification and quality control of DNA were done using spectrophotometery method and 1% agarose gel electrophoresis. DNA fragments was amplified via PCR and the products were run on polyacrylamide gel. Staining of the gels was performed through silver nitrate. Statistical analysis represented mean of genetic parameters include real and effective alleles, observed and expected heterozygosity for over loci in all populations, 5.458, 3.793, 0.500 and 0.649 respectively. The AMOVA test showed maximum rate of F st (0.086) and minimum rate of N m (2.652) between Khuzestan and Bushehr populations; Also minimum rate of F st (0.034) and maximum rate of N m (7.070) were between Dayyer and Bandar-Abbas populations. Khuzestan was the ultimate population and results were showed an intermediate separation from other studied populations; thereby separated fisheries management is recommended.
