Red ginseng grades are divided into four grades. (A) Heaven-grade product should not have cracks and scratches. For rootlets, one or more should be well developed and the rootlet length should be three quarters or less of the main root. On the rootlet, there must be a diameter of ≤0.5 mm of whitening and pitting and an affected length of ≤10 mm. The color of the textures must be maroon, brown, or dark brown. (B) Earth-grade product has cracks and scratches on the body of the total surface area of less than a quarter. Earth-grade rootlet development is the same as that of heaven-grade. There must be a diameter of ≤2.0 mm of whitening and pitting and the affected portion length of a quarter. The earth-grade textures/colors are similar to heaven-grade. (C) The yang-grade standard is not limited to the body. Rootlets are unbalanced. Whitening of the body length is one third or less, and the pitting of the body length is one half or less. Like the other two grades, the Yang-grade color is shiny, but the color is not uniform. (D) The jab-grade has no standard and grade. The heaven-, earth-, yang-, and jab-grades are given based on the parameters provided in the Korea Ginseng Industry Act.

Abstract: The present study examined the volatile profiles of Panax ginseng (Korean ginseng) and Panax quinquefolium (American ginseng) grown for different cultivation years by using HS-SPME/GC-MS and determined the key discriminant volatile compounds by chemometric analysis including principal component analysis (PCA), hierarchical cluster analysis (HCA), and partial least squares-discrimination analysis (PLS-DA). Fifty-six compounds, including forty terpenes, eight alcohols, one alkane, one ketone, and one furan, were identified in the ginseng roots. The chemometric results identified two major clusters of American ginseng and Korean ginseng cultivars with distinct volatile compositions. The volatile compounds in fresh white ginseng roots were affected by the species, but the influence of different cultivation ages was ambiguous. The major volatile components of ginseng roots are terpenes, including monoterpenes and sesquiterpenes. In particular, panaginsene, ginsinsene, α-isocomene, and caryophyllene were predominant in Korean ginseng cultivars, whereas β-farnesene levels were higher in American ginseng. The difference in volatile patterns between Panax ginseng and Panax quinquefolium could be attributed to the composition of sesquiterpenes such as β-panaginsene, ginsinsene, caryophyllene, and β-farnesene.

Ginseng (Panax ginseng) has a low genetic diversity and a narrow pool of genetic resources. Mutagenesis is one of the most powerful methods for inducing genetic variation in this species, but little research has been performed in ginseng. In this study, various tissues, including dehiscent seeds, 1-year-old roots, and somatic embryos were irradiated at different doses of gamma rays (20–400 Gy and zero dose as a control) to determine the most optimal concentration and tissues for the successful use of mutagenesis in ginseng breeding. The results revealed that high gamma doses (> 100 Gy) were detrimental to all irradiated tissues. A gradual and significant reduction in germination, emergence, and seedling growth were found as gamma irradiation dose increased. The reduction in survival rates and seedling growth by irradiation at serial doses showed that the LD50 of ginseng was 20–80 Gy, although the irradiation doses were tissue dependent. Based on our results, the optimal doses of gamma rays for inducing mutation in ginseng are < 20, 40, and 60–80 Gy for 1-year-old roots, dehiscent seeds, and somatic embryos, respectively. Given the fact that ginseng somatic embryos are less sensitive to gamma rays than other tissues, the combination of in vitro culture and mutagenesis could be more effective than the conventional method for mutation breeding in ginseng. These results provide a good basis for radiation sensitivity of ginseng and are useful as a guideline for ginseng mutation breeding.

Panax ginseng C.A. Meyer is a traditional medicinal herb that produces bioactive compounds such as ginsenosides. Here, we investigated the diversity of ginsenosides and related genes among five genetically fixed inbred ginseng cultivars (Chunpoong [CP], Cheongsun [CS], Gopoong [GO], Sunhyang [SH], and Sunun [SU]). To focus on the genetic diversity related to ginsenoside biosynthesis, we utilized in vitro cultured adventitious roots from the five cultivars grown under controlled environmental conditions. PCA loading plots based on secondary metabolite composition classified the five cultivars into three groups. We selected three cultivars (CS, SH, and SU) to represent the three groups and conducted further transcriptome and gas chromatography-mass spectrometry analyses to identify genes and intermediates corresponding to the variation in ginsenosides among cultivars. We quantified ginsenoside contents from the three cultivars. SH had more than 12 times the total ginsenoside content of CS, with especially large differences in the levels of panaxadiol-type ginsenosides. The expression levels of genes encoding squalene epoxidase (SQE) and dammarenediol synthase (DDS) were also significantly lower in CS than SH and SU, which is consistent with the low levels of ginsenoside produced in this cultivar. Methyl jasmonate (MeJA) treatment increased the levels of panaxadiol-type ginsenosides up to 4-, 13-, and 31-fold in SH, SU, and CS, respectively. MeJA treatment also greatly increased the quantity of major intermediates and the expression of the underlying genes in the ginsenoside biosynthesis pathway; these intermediates included squalene, 2,3-oxidosqualene, and dammarenediol II, especially in CS, which had the lowest ginsenoside content under normal culture conditions. We conclude that SQE and DDS are the most important genetic factors for ginsenoside biosynthesis with diversity among ginseng cultivars.

Panax ginseng is a medicinal plant belonging to the Araliaceae family. Ginseng is known as the king of oriental medicine, which has been practiced since ancient times in East Asian countries and globally in the modern era. Ginseng is used as an adaptogen, and research shows that it has several pharmacological benefits for various ailments such as cancer, inflammation, diabetes, and neurological symptoms. The pharmacological benefits of ginseng are attributed to the triterpenoid saponin ginsenosides found throughout the Panax ginseng species, which are abundant in its root and are found exclusively in P. ginseng and Panax quinquefolius. Recently, with the completion of the entire ginseng genome sequencing and the construction of the ginseng genome database, it has become possible to access information about many genes newly predicted to be involved in ginsenoside biosynthesis. This review briefly summarizes the current progress in ginseng genome analysis and genes involved in ginsenoside biosynthesis, proposing directions for functional studies of the predicted genes related to ginsenoside production and its regulation.

The purpose of this study was to analyze the correlation between the external, internal and chemical quality characteristics of 6-year-old red ginseng produced in punggi area. Red ginseng samples are graded as cheon (1st grade), ji (2nd grade), yang (3rd grade), gi (out of grade) by trained inspectors. External characteristics of red ginseng depend on weight, volume, density, length and color. Internal characteristics were expressed by length and occurrence ratio of inner white and inner cavity. Chemical characteristics were evaluated by crude saponin, ginsenoside (Rg1, Rb1, Rc), total sugar, acidic polysaccharide, and 50% ethanol extracts contents. Overall, this study showed that quality of external and internal characteristics was matched with grade, but chemical quality was not matched.

There is a total of 32 ginseng cultivating varieties (cultivars) in Korea which have unique agronomic and morphological traits. Each variety was registered as an official cultivar and protected for breeder’s rights permitted by the Korean government. All 32 elite cultivars have been developed by pure-line selection from local landraces and show diverse agricultural performances and characteristics such as growth, floral and fruit shape, root growth, shapes and processing traits, characteristics for biotic or abiotic stress, and ginsenoside compositions. This chapter will discuss the breeding method and characteristics, breeding history in Korea, and characteristics of officially registered ginseng varieties.

Chinese materia medica (CMM) is indispensable component of Traditional Chinese Medicine (TCM) therapy. With the widespread of TCM around the world, the quality control and safe use of CMM become a major concern. This paper introduces the role of ISO standards for industrial development and current development status of CMM standards in ISO/TC 249. Through the comparison of similarities and differences between CMM standards in ISO/TC 249 and pharmacopoeias of main stakeholders, this paper suggests strengthening standard formulation in the following areas to provide more appropriate documents to facilitate the international trade and promote the industrial development of CMM: (1) Develop standards to fill the blanks among the whole industry chain of CMM; (2) Develop standards for new forms of CMM and services; (3) Develop specification and grade standards for CMM with large quantity and high value.
