Endophytic fungi have garnered attention for their ability to synthesize chemical compounds similar to those of their host plants. This study evaluated the bioactivities of BwKpRt-1, an endophytic fungus associated with Kaempferia parviflora rhizome. Bioactivity analyses included antioxidant assays, antibacterial, and antiproliferative toward cell lines, identified a fungal species and its chemical compounds using liquid chromatography high resolution mass spectrometry (LC-HRMS). Docking study for equisetin was also carried out in this research. The extract of BwKpRt-1 demonstrated antioxidant activity with an IC50 value of 57.36 μg/ml (DPPH) or AAI value of 0.5 (moderate). Its α-amylase and α-glucosidase inhibitory activities were 0.121 ± 0.02 nmole/minute/ml and 335.38 unit/l, respectively. Antibacterial activity is strong against Staphylococcus aureus with a minimum inhibitory concentration value of 64 μg/ml. The anticancer activity of this extract against MCF-7 and A549 showed IC50, which were 58.61 and 42.64 μg/ml. Chemical compound analysis using LC-HRMS identified equisetin as a main compound (42.18%). Docking study showed that Equisetin (EQ) exhibits stronger antioxidant and anticancer potential than standard drugs, showing lower energy affinities than vitamin C (VitC) for HsKEAP1 (−8.5 vs. −6.1 kcal/mol) and doxorubicin (DXR) for HsEGFR (−8.5 vs. −7.8 kcal/mol). Phylogenetic analysis revealed that BwKpRt-1 had a 99.08% similarity to Fusarium equiseti. These findings highlight the potential of F. equiseti BwKpRt-1 as a source of bioactive compounds for pharmaceutical applications.

Bacteria are one of the organisms that cause infectious diseases. One of the bacteria that can cause infection is Staphylococcus aureus. Treatment of bacterial infections is by using antibiotics, but using the antibiotics for a long term can cause resistance. It is necessary to look for alternative compounds that can inhibit the growth of Staphylococcus aureus. In the bioprospecting process of medicinal plants, it is not only necessary to rely on empirical information but also to have scientific evidence proven through scientific testing. One of the plants with potential as a medicine is black ginger (Kaempferia parviflora). The aim of this study is to determine the effectiveness of Kaempferia parviflora rhizome in inhibiting the growth of Staphylococcus aureus bacteria. The concentrations of the Kaempferia parviflora extract tested were 25%, 50%, 75%, and 100%. Based on the results, it can be concluded that the ethanol extract of Kaempferia parviflora rhizome has antibacterial activity against the growth of Staphylococcus aureus ATCC 25923 at concentrations of 25%, 50%, 75%, and 100%, with strong antibacterial inhibition. This study is expected to contribute to the field of health in general and pharmacy in particular by developing the potential of Indonesian medicinal plants. The findings of this study will be directed toward the development of formulations with antibacterial activity.

Kaempferia parviflora (KP), also known as Black ginger or Kra-chi-dum in Thailand, is a Zingiberaceae plant native to Thailand and Malaysia. This plant is commonly used by the community as a spice in cooking and healthy drinks for disease prevention and treatment. According to research, KP extracts have broad bioactivity against infectious diseases caused by viruses and bacteria, as well as diseases related to metabolic disorders such as obesity, diabetes, aging, and gastrointestinal disorders. KP is also effective as an anticancer agent. This review article focuses on KP's anticancer activity and possible mechanisms of action. Furthermore, the botanical aspect and content of active compounds are investigated in this article. Considering KP's high potential as a medicinal plant, the potential interactions of KP with synthetic drugs and its potential formulation are investigated. According to the findings of this review, KP has anticancer and antimetastatic activity, and the active ingredient responsible for this activity is methoxyflavone.

Ngải đen (Kaempferia parviflora Wall. ex. Baker) là một dược liệu có nhiều tiềm năng. Theo dân gian, đây là loài có tính vị cay, nóng, ấm. Ngải đen được biết đến là vị thuốc phá huyết hành khí cực kì hiệu quả. Ngoài ra, thân rễ Ngải đen còn nhiều tác dụng dược lí như: chống loét dạ dày, loãng xương, kiểm soát cholesterol, kháng khuẩn... Nghiên cứu này được thực hiện với mục tiêu khảo sát độc tính cấp đường uống và tác động kháng viêm của cao chiết thân rễ cây Ngải đen trên mô hình thực nghiệm nhằm cung cấp thêm nguồn dữ liệu khoa học về tác dụng dược lí của dược liệu này. Thân rễ tươi của cây Ngải đen dùng phương pháp ngâm lạnh bột dược liệu bằng ethanol 70% để thu được cao đặc thân rễ cây Ngải đen. Hoạt tính kháng viêm của cao chiết thân rễ Ngải đen được xác định ở các liều 250 mg/kg và 500 mg/kg trên mô hình gây viêm bàn chân chuột nhắt bằng cách tiêm dưới da carrageenan 1%, chất đối chứng được sử dụng trong mô hình là diclofenac 5 mg/kg. This study aimed to develop a Kaempferia parviflora (KP) extract based on electrospun shellac fibers capable of transporting methoxyflavones. This study used a Box–Behnken design to determine the optimal production parameters that influence the fiber diameter and bead-to-fiber ratio responses. The optimization step produced fibers with a small diameter (574 nm) and a lower bead-to-fiber ratio (0.48 beads per fiber) by combining 37.25% w/w shellac and 1.50% w/w KP extract with a solution feed rate of 0.8 mL/h and an electrical voltage of 18 kV. The KP extract was found to be dispersed throughout the electrospun shellac fibers during the characterization study. The results were highly correlated with the theoretical values, indicating that the regression models used to predict the response variables were adequate. A study of in vitro dissolution confirmed that KP extract-loaded electrospun shellac fibers could produce a sustained-release profile within 10 h. Additionally, KP-infused shellac fibers demonstrated antibacterial activity against Staphylococcus aureus. This KP loading method combined with shellac properties provided a new delivery system and could be used to explore novel biomedical materials.

Background: Kaempferia parviflora (KP) has been used in traditional Thai medicine to cure gastrointestinal disorders since ancient times. Helicobacter pylori is an initiating factor in gastric pathogenesis via activation of massive inflammation, the cumulative effect of which leads to gastric disease progression, including gastric carcinogenesis. Accordingly, the effect of a crude ethyl acetate extract of KP (CEAE-KP) on proinflammatory cytokine production and cell chemotaxis was the focus of this study. Methods: The cytotoxicity of CEAE-KP (8-128 μg/ml) on AGS (gastric adenocarcinoma) cells was determined at 6, 12 and 24 h using an MTT assay. The effect of CEAE-KP on H. pylori-induced interleukin (IL)-8 production by AGS cells was evaluated by ELISA and RT-PCR. The effect of CEAE-KP on monocyte and neutrophil chemotaxis to H. pylori soluble protein (sHP) and IL-8, respectively, was determined using a Boyden chamber assay with THP-1 or HL-60 cells. Results: CEAE-KP reduced AGS cell viability in a concentration- and time-dependent manner, but at 8-16 μg/ml, it was not cytotoxic after 6-24 h of exposure. Coculture of AGS cells with CEAE-KP at a noncytotoxic concentration of 16 μg/ml and H. pylori reduced IL-8 secretion by ~ 60% at 12 h, which was consistent with the decreased level of mRNA expression, and inhibited neutrophil chemotaxis to IL-8. sHP (100 ng/ml) induced marked monocyte chemoattraction, and this was decreased by ~ 60% by CEAE-KP. Conclusion: CEAE-KP might serve as a potent alternative medicine to ameliorate the inflammation mediated by H. pylori infection.
