I use MBP tag to promote the soluble expression of our interesting protein which cannot expressed in E.coli fused with His6 and GST tag. Now, we can obtain the soluble MBP fusion proteins, but it is difficult to perform further purification. There were always some contaminated proteins which were GroEL and single MBP and maybe other proteins identified by MS. So in my opinion, this means the fusion protein maybe form soluble aggregate or at least they didn't fold perfectly. My questions are as following:
1, How can I confirm this assumption? I use sepharose 200 column to determine the molecular size of fusion protein. The apparent size is obviously larger than estimated size, but the estimated size is premised on global protein. so are there some other methods which can determine its aggregated state?
2, if the fusion protein did not fold properly, are there some more efficient strategies to optimize its expression and folding. we have tried different induction conditions such as different temperature and different IPTG concentration and induction time, we use DH5a and Bl21 to perform the expression, they all doesn't work.
Others, the main function of this protein is to perform protein-protein interaction, so the activity is hard to confirm.

This may be tricky. I know, that the typical approach is to make a crystalline derivative of an enantiopure compound and then using the typical crystallographic methods determine the absolute configuration. Yet, in my case, when working with specific non-proteinogenic amino acid derivatives this poses a problem, as getting a crystalline derivative seems impossible. I heard, that there is a possibility of establishing the absolute configuration of such molecules via CD or other spectra, via their comparison with modelled data. This would suit my needs, yet there seems to be not much data on the topic. Can anyone help me with that? What kind of equipment is needed, in what software should I build the model?

We are observing two different species populations recovering from disturbance. Both populations were reduced by about 80% with significant changes to their size structure. Following the disturbance, one population's size structure immediately returned while abundance remained low. The second population's abundance immediately returned while the size structure was unexpectedly different (shifted from a bi-modal to a uni-modal distribution). I am searching for some explanations for these different recovery patterns. Any help would be very much appreciated.

I'm working in GRASS-GIS and need some help
I have to analyze 10 years of MODIS images by using the product -MOD13Q1 NDVI. I understand this band have all pixel already with values of NDVI calculated. So, to get this band I used MRT to reproject the files into Geo-Tiff format.
a.- Is it possible to use these Geo-tiff to analyze NDVI for every month in a full 10 years?
Also, the region/area downloaded is just one tile, and the area to be analyzed is smaller (crop-land). I guess I will have to specify the geographic area.
b.- would someone guide me in this process? by choosing pixels? or selecting a square inside the tile?

I'm synthesizing some supported catalysts batches with silica and alumina support. Support particles are in granular shape. The final catalysts should be ground into powder form prior to loading into the reactor. I was wondering if there is a significant difference or any preference between 1) First grinding the granular support, followed by impregnating and drying and 2) Impregnating and drying, followed by grinding.

One of the many ways to synthesize silver nanoparticles is to reduce AgNO3 with NaBH4. However, the final product we obtain from this reduction process is a yellow colloidal silver. Is the yellow colloidal silver itself ok for direct analysis of silver nanoparticles (UV-vis , SEM, etc) or should I actually further process the yellow colloidal silver by centrifugation / drying in oven to filter out other chemicals present in the yellow colloidal silver in order to get pure silver nanoparticles (and then resuspend it in DI water again)?
Does anyone have the exact protocol of the above mentioned steps? I am quite clueless on the sample preparation part for instrumental analysis.
The chemical equation for this reduction process is :
AgNO3 + NaBH4 ----> Ag + 1/2H2 + 1/2B2H6 + NaNO3
So is it correct to say that centrifugation and drying are needed to remove 1/2H2 + 1/2B2H6 + NaNO3 ?
Will centrifugation and the drying process actually lead to the agglomeration of the silver nanoparticles? If yes, is there any way to avoid it?
So far, in most of the journals that I've read, not many actually mentioned about the further processing steps that i mentioned above.

I have a cytidine•HCl analog, and would like to do some chemistry on it, but the HCl is limiting my solubility in organic solvents. I would like to remove the HCl, but the kicker is the subsequent reaction must be done in the absence of water. NaOH will give me NaCl, but then I would need to remove the salt from the solution. AgNO3 precipitates AgCl, but leaves the NO3 anion.
Please note, this is a cytidine analog that is not commercially available, so purchasing it without the HCl is not an option.
