There is a clear indication to perform a biopsy if MRI and CT are not conclusive (or by missing of clear signs such as "wash out") and AFP is within the reference. I agree that there is no need for biopsy if the indication for surgery (open or laparoscopic) is already given. In cases of transplant candidates (inside Eurotransplant) it makes sense to perform a biopsy in order to have a definite diagnosis, especially in presence of multiple liver nodules. We must also consider the risks of a biopsy, especially in patients with advanced liver cirrhosis with clear impairment of the liver function or by presence of ascites.

Thanks for enlighten about various aspects of organic farming and food production and in most of the answers very simply an equation is generated that is food production = food consumption and that is partly true because there are several factors ( social, climatic, political etc.) that decided who will get how much food. Till date 30 % of the world rich population is eating equal to 70 % of the poor population. So it is the matter of distribution first. Second is the losses during transport, storage etc. Third is the commercial crops like floriculture ,sugarcane, cotton, biofuel, plantations etc and even construction of cement forests- is rapidly encroaching the fertile fields of food grain crops.Now we always worry about growing population but on the other side no effort to preserve/ reserve fertile lands for food grain crops. Now come to organic vs. chemicals take the example of India plateau in food grain crops is observed since last two decades despite of sharp increase in fertiliser and pesticide consumption than how there is chemical system gives guarantee for feeding the population in 2050 or after , at that time petroleum and phosphate reserves will also be exhausted- the major source for fertilisers and pesticides. And the pollution level will reach to an alarming level. In most of the seminars now a days a ray of hope is carbon sequestration, low emission of green house gasses - which farming fits in that . Obviously organic farming but it has to be taken in wider perspective and not keeping limited to the certification, high price in market , safe food BUT a holistic approach for efficient utilisation/recycling of natural resources. AND it has been proved at many places that this approach is successful to feed the population till the civilisation exist. Learning from such examples may help to make strategy for planning to feed growing population. In conclusion, The challenge is multidimensional therefore, solution will have to be multidirectional with near to nature approach as the agriculture is still a nature's gift and may not be long lasting if we go away to nature.

As mentioned early in this thread, podcasts should be viewed as a form of media, rather by a pre-defined conception of its worth as a source of information. Loads of "scientific books" are worthless and devious. Concerning the primacy, the validity of a source, I'm of the opinion that thoughts are build, ideas structured on former conception and further generating new ones, all inspired and transferred by each-other. The interaction is undeniable. It's not so much about the media form in which information is presented, but rather by what way and by what method it is distributed. With the multitude of developing means of distribution of information and ideas, we just have to adapt. THIS SAID, there are complication as each method might enforce an ad hoc approach and criteria when examined - just as others have already mentioned. Orality, for example in a podcast, might be difficult to transcript or digitally embed in a work. A spontaneous dialogue might have its complications but also it's advantages. Interesting interactions between talkers, new ideas, metaphoric references or new formulations might occur. It might also be very precise and well formulated if not a direct reading of a text. Like an audiobook. And some lectures might be like that too; spontaneous and free-flowing or strict and precise. Then again, a text too could be bad written, poorly structured and generally confusing and yet well appreciated. In fact, i find this to be the case within many fields. Few researcher are good writers - both in humanities and natural sciences. In general, my opinion is that, we should focus more on constructive criticism upon the content rather than the media. This seems obvious, but is rarely the case. We are creatures of structure and habits - we like to stick with what works and what we know; safety. But more over, we are creatures of logic. No organism, have been so inventive as humans. Adapting to new circumstances and changing our environment to our advantage is what we do. Media is communication. Better communication strengthens our development and survivability. Don't reject new media. Embrace it. Modulate it.

Hi, I can only think that there was a high concentration of proteins in the samples that interfered with the assay. The protocol suggests filtering through a 10kDa MW cut-off filter. Also make sure that all reagents are at the right temperature. Also the protocol does not say whether to use flat or V shaped plates. I would recommend flat or try both and that the plate is recommended for your type of application. Good luck and let me know!

I did western blot expression of HIF alpha in animal tissue homeganates, In my case i used PVDF membrane and results were good. Nitrocellulose membrane is good when the bands you get are strong, for light bands its difficult to detect. PVDF is better in those cases. Try to activate your membrane with methanol before transfer. Hi Daniel, I've found that tet-inducible expression in T brucei is usually maximal at 10 to 20 ng/ml, however most people use 1 ug/ml as standard. If you're dealing with toxic expression (significant loss of function RNAi or deleterious over-expression) you can dial the expression over a reasonable dynamic range by using 1 to 10 ng/ml. You also need to be aware that integration of expression cassettes at different rDNA spacer loci leads to different expression levels, especially in bloodstream form parasites (see Alsford et al, 2005, Mol Biochem Parasitology, for details on position effect in T brucei). This will obviously influence the amount of tet you need for each cell line. Hope this helps.
