I'm synthesizing some supported catalysts batches with silica and alumina support. Support particles are in granular shape. The final catalysts should be ground into powder form prior to loading into the reactor. I was wondering if there is a significant difference or any preference between 1) First grinding the granular support, followed by impregnating and drying and 2) Impregnating and drying, followed by grinding.

I´m using as secondary antibody: HRP Conjugated anti chicken IgY (http://www.genetex.com/Chicken-IgY-HL-antibody-HRP-GTX26753.html) and TMB substrate (http://www.mpbio.com/product.php?pid=02190277).
I´m having problems preparing the substrate. I´m adding 30mg TMB + 5ml DMSO to prepare stock solution. The working solution is prepared as described: 100ul stock solution TMB + Then add 9.9ml of acetate buffer 0.1M and 30ul of H2O2 (30%) for a final concentration of 0.01% H2O2.
Please if some one has any suggestions I would really appreciate it. I´m not very experience with this technique.

I have done an EDC Coupling reaction of 3-amino-N-Benzyl pipiperidine with acid chloride. The reaction went neat and clean and the product was isolated. While in H1 NMR I saw a geminal coupling of N- benzylic methylene splitting with 13.2Hz at 3.2 and 3.8 delta scale. I didn't understand how this happened.

One of our students would like to solve antenna placement (or radio network design) problem, using evolutionary computing methods. We have to formulate an optimization problem for her. As I am not expert in antennas, it is difficult for me to point out the most important antenna parameters for such an optimization problem.
The formulation of the optimization problem should be as simple as possible (with reduced set of antenna’s parameters), because the focus will be on developing heuristic algorithms.
The simplest idea emerging is to place antennas in a set of given sites, so that maximum radio coverage with minimum costs is achieved. In this case the fitness function of an individual will be calculated by radio coverage and costs (for placing or for rental).
Would you, please, advise me if such a problem formulation is meaningful?

I'm trying to use the assay described in this protocol to assess bone marrow-derived macrophage migration. Rather than using 35mm dishes, I'm using 6-well plates, which have wells with the same area as a 35mm dish. I find that the medium I've loaded into the wells cut into the gel disappears in an hour or less. I've tried filling the inter-well spaces of the plate with water, but this doesn't prevent the loss. Are my wells drying out or can the medium leak under the agarose? How can I keep this from happening?

I took 200 sugarcane leaf samples preserved in silica gel abroad for DNA extraction and SSR analysis. After the DNA is extracted and checked in 0.8% agarose gel, the DNA quantity and quality seems good but it failed to amplify using SSR markers. What could be the possible reason?

I wish to analyze the lipid content in a solution I created. The surface lipids of lizards were extracted by soaking previously deceased lizard in hexane for 4 hours. I then removed lizards, added ethanol and evaporated hexane from the solution. The resultant mixture was surface lipids with ethanol. I wish to confirm the extraction was successful by measuring lipid content.

I am trying to mutate a residue in my gene of interest that is cloned into a pET28a vector. After transformation, I get no colonies from my LB/Kan plates. I re-designed the primers and repeated the experiment but it still failed. As a control, I tried to transform the template DNA into the XL1 Blue cells and it gave me >100 colonies. This protocol has always worked for me in the past. I was wondering what could be a possible explanation for this.

I would like to come up with a more effective heat trasfer from the device to the cooling air. Heatpipes, and copper spreaders are but a few examples - I'm looking for more and different ideas.

I am making a coupled temperature-electrical-structural model for spark plasma sintering. I have used Abaqus standard.
The model worked for simple geometry involving only 3 parts (2 punch and die), but it is not converging for my original model which has 2 punch, sample, die, 4 spacers.
The error that I am getting is -
Too many attempts made for this increment
There are few warning messages aswell
1. Displacement increment for contact is too big
2. SURFACE INTERACTION "interaction name", ASSOCIATED WITH ONE OR MORE CONTACT PAIRS WITH SURFACES HAVING UNDERLYING ELEMENTS WITH ELECTRICAL, TEMPERATURE, AND DISPLACEMENT DEGREES OF FREEDOM, DOES NOT HAVE ANY ELECTRICAL INTERACTION DEFINED. NO ELECTRICAL INTERACTION WILL OCCUR.
I used to SI units and applied a pressure of 5 MPa at the top of upper spacer.
