The suggestions from Aditi and maria are good. Another possibility is to precipitate the whole extract with 10% TCA (Trichloroacetic acid) or do cold Acetone perecipitation,. Precipitation will remove salt and other detergents, and concentrate protein-but yield of protein may decrease. If you do want to try-pl. check web for protocols. As a test, you can precipitate a known amount of BSA, and see how well it works for you. Hope this helps!

That's a vast question whose answer depends mainly on the methods you want to use for docking and scoring. Some protocols require proper charges and strict parametrization of atoms (e.g. Autodock, Glide). Others are much more permissive on parametrization (e.g. Gold, SwissDock). Regarding geometry of mutated residues, though some automated procedures exists (e.g. PDB_Hydro, FoldX) I still think it is wise to have a clear visualization of the local microenvironment (protonation, hindrance, secondary structure, concavity, etc...) before optimizing locally. Rotamer libraries help a lot as well ( e.g. SCRWL, PREPARE). Personally I like to start within Meastro environment which mutates residues in a fashion that is a good starting point. Yes, cell density should be lowered. Passage no. also should be below 5. If cells are detaching, its worth checking with other sources of gelatin. Unless cells adhere well in the beginning of the assay, you cannot measure survival or proliferation...you will have to account for detachment which leads to death. Perhaps, the final steps in HUVEC prep or the washes make a difference in how well the cells attach? Hope this helps!

I have been having more advice from my University professors, and it seems like the inter-cultural and the inter-disciplinary writing styles are a big academic problem that very few people is aware of... For instance, yesterday I just knew that United States style uses (" ") as quotation marks and (' ') as a quotation within a quotation. The British style it the opposite.
