On that point Roy, assuming that I understand you, I would say that we diverge drastically. If you were not in your turn pulling my trick of letting bad proofing speak for my opinions, then please clarify what specific mechanisms or principles you had in mind. Considering the volume of tissue and the volume of solution in the slicing chamber, the % of fixative is going to be at best at trace levels, especially after rincing the chamber. What was your approximate beam size? Do you have the ability to conduct rocking curve analyses? What is the smallest angular step possible? Do you have Phi rotation capabilities? Have you determined the sample surface orientation or do you have a guess? If so then you may map both the "relative dislocation density" & the "relative orientation" over the wafer surface by adjusting the sampling size (beam size @ sample surface). Assuming the detector window is "slit-less".

Different temperatures are used for blunt end vs sticky end ligations. For blunt end ligations, there is no Watson-Crick base pairing to promote DNA fragment association. Dear Andras, probably time perspective psychology might be of interest to you. The concept of a so called 'balanced time perspective' might represent one aspect of a 'mature personality'. Instruments for assessing (balanced) time perspective are the ZTPI (Zimbardo) and the BTPS (Webster). Another concept that might be interesting for you is 'wisdom'. Hi Sven, I work for Blue Ventures a marine conservation NGO based in Madagascar which has been working to involve communities in fishery management since 2004. We believe our model is working and communities are now involved in planning temporary (2-3 month) octopus and crab fishery closures with little input from us as the support NGO. Fishery catches are also relatively stable considereing natural variations in abundance. Recently they also began paying a voluntary contribution into a community held central management pot so that the costs of organising future closures could be covered by themselve.
