How to select the upper and lower limit for a variable while carrying out DOE ( Design of Experiments) ?
Is it advice able to choose a large range (i.e. upper limit -lower limit) for variables while performing DOE ( Design of Experiments) ? Or is it like always low range is preferred to avoid the non-linearity ? Because i am not sure whether the effect while moving from lower to upper limit is linear or not.

Another aspect of the answer from the Linux distribution perspective is that. The Debian based distribution like Ubuntu more focuses on day to day usage of the PC. Like we use the windows so its more focus is to provide tools that make your life easy. Lot of development is going on to provide ready to use packages instead of compiling and setting up by your own. Thats the reason the new distributions are some time not stable Mark mentioned. However, if you use the stable release like 12.04, 13.04, 14.04 and so on it would provide the stability as well.
On the flip side RedHat based distributions focus on server side services. Like Application server, Web server, DNS server, VoIP server and so on. So for this reason their focus is to provide stability and security instead of providing new tools that makes your life easy.
In summary if are good with Linux and can tackle with errors you will get while setting up your video services then go for Redhat based distribution like CentOS which is freely available. Otherwise better to install the stable release of Ubuntu. The current stable release 15.04.

I modelled my concrete using the discrete model for my reinforcement. In Ansys workbench. However, in Ansys workbench I can define my material due to 2 methods using engineered material or by adding a command. Now I try to model using the smeared method. Can anyone suggest a robust algorithm for large systems treated with TD-DFT and with a diffuse basis set in Gaussian?
I'm currently trying to use Gaussian to obtain ECD spectra of a fairly large molecule in a mono- and dimeric state (roughly 150/300 atoms). I'm using TD-DFT with a DZV Dunning cc basis set, the CAM-B3LYP hybrid XC functional to better account for long-range interactions and the PCM to simulate the aqueous environment. With this setup, the calculations (20 representative structures of each state) converge quite nicely, but fail to give satisfactory results when comparing with experimental measurements.
I thought this problem might be caused by the lack of diffuse basis functions, which are crucial to describe non-covalent interactions in large systems, but with the augmented Dunning set, the calculations fail miserably in terms of convergence (which AFAIK is a common issue with diffuse sets). I've already tried several "robust" approaches to the SCF procedure, with keywords Big, YQC and CDIIS, but none of them helped -- even for the smaller systems with no PCM and very few excitations. I know this is a lot to ask for anyway, but maybe there is a way to improve convergence in such difficult cases? I'd be very grateful for any piece of advice.
Indeed, there are many hydroxyl groups in my molecule -- it's a polyene antibiotic with a polyol part; however, the orientations should be more or less random in my 20 frames (they are at least several ns apart), and I do not see that much variation in the band positions. There appears to a blurring of the lines regarding grounded theory as a methodology for analysing data and as a framework for generating theory. Can grounded theory be used as a research technique or methodology to analyse data only without generating theory?
One more comment if you will. Once you have analyzed your data, you have what? What exactly are your results telling you? Why is it NOT a theory? Perhaps, its the use of that word that is in question. Then we go back to what is the meaning of theory and what is a theory?

Does anyone encounter these contaminants in cell culture before?
Recently I sub-cultured one prostate cell line into two T25 flasks, and observed some contaminants (as shown in the figure) in one of the flask after 5-day growth. Cells seem to grow normally with these contaminants when I observed on the 5th day. However, today (which is 6th day) I observed again, all the cells died and were floating in the media. Cells in the other flask grew normally. I suppose this is individual contamination, that is, not caused by contamination in media or equipment used. Does anyone know what are these contaminants? Are they fungi?

Cell functions are maintained under normal physiological homeostasis, of which the redox (reduction-oxidation) status is of importance. Which subsets of genes are specifically (or coordinarily) responsible for redox homeostasis or are responsive to redox signalling? More interestingly, the endoplasmic reticulum (ER) enables the more oxidating status to be maintained for protein synthesis, as well as some lipid synthesis and processing. By contrast, the ER-continuous membrane-enclosed nucleoplasm keeps it in more reducing conditions facilitating gene transcriptional expression. This raises another question of which subsets of genes are involved in the reduction (or oxidation) controls or are responsive to changes in the reducing (or oxidative) conditions?
I can tell you that the GST family plays a large role in oxidizing substrates for biosynthesis, handling ROS, and xenobiotic defense. Maybe you could focus on the GST family members that are specific to dealing with ROS. I also suspect some key players of redox homeostasis to be heavily affiliated with the mitochondria- does the mtDNA give any clues?
As for genes responsive to redox changes? In theory, any gene product that requires disulfide bonds for proper functioning would be responsive. More specifically, ROS activates JNK signaling through Ask1 (depending on who you "ask"). Translocalization of cytochromeC to the cytoplasm is thought to also be in response to oxidative stress.
