In the freshman-level "Introduction to Signals" course that I teach, I notice that some students have a hard time in grasping the notion of negative frequencies. - How to measure self-made protein with his-tag after purification with imidazole-containing buffers?
As mentioned I am currently producing 2 different types of recombinant his-tagged proteins. One is intracellular and one is secreted into the supernatant - and them I am using a Ni-NTA Resin to purify these. Herefore I made buffers (equilibration buffer, wash buffer and elution buffer) that contain different concentrations of imidazole. This usually works for some point - but when I measure I get barely anything if not negative values - if I measure in the nano drop but I see in the peaks on the machine that the imidazole makes problems in measurements. I tried a BCA which did not really work. But if I load just very few on a western - I get a huge and correct band, detecting with my monoclonal specific antibody. Now I was wondering - is there anyone who can help in terms of " how to measure these proteins" ? This is of course essential for establishing protocols for ELISAs or anything...(I already tried the low-UV absorbance imidazole from SEMBA - this did NOT change anything). There are 63 sites in total. With Chm25, Wetnes25 and Light25 = my environmental covariates.
Here, there are 7 years of sampling but sampling effort (the number of time a site was surveyed) are not equal between years and between 3 groups of sites.
How can I introduce this sampling effort in the "occu" function ?

This is Pomacentrus proteus. The species is common in Sri Lanka; it is only found in Sri Lanka and the eastern Andaman Sea. The lower half of the body in the juvenile is yellow; the adult is mostly bluish grey, including the caudal fin.
Pomacentrus trilineatus is restricted to the Western Indian Ocean (Red Sea, East Africa, Seychelles, Madagascar and Mascarenes); it has a pale caudal fin and posterior dorsal and anal fin in adults, and a much more yellow body in juveniles. The two species are illustrated in Allen (1991):

I am in the process of developing a proposal which hinges on developing a risk assessment model. I am proposing to use multi criteria decision analysis tools for this especially the AHP by Prof Saaty. However, one of the Professors commented as follows: "The research method suggested looks rather simple and is using a very outdated method".
I note that much as there have been newer versions as alternatives to the AHP ie fuzzy methods and TOPSIS, the AHP appears to remain dominant both in practice and academia. Based on this, i feel the AHP remains a dominant legend in multi criteria decision analysis. Any thoughts for tried and tested alternative methods for me to develop a risk assessment model?
I have applied AHP and (several versionsof) fuzzy AHP before. AHP suffers from some very major issues. Fuzzy AHP (not all, the one proposed by Mikhailov, 2003) is much better than AHP. It is true that AHP is still the dominant MCDM method, however it does not mean it is a good method.

I used Denver II screening developmental test to measure the developmental outcomes, for children from 3 months to 5 years. For the data analysis, I used step-wise multiple regression.

- Can we automatically classify only the rivers from satellite images using any programming or different methods? Suppose we have temporal data of a particular river from the past 10 years; it would take a long time outlining the rivers and than perform river change detection. So can we automatically classify those river using a programming platform?
In addition to these described above, I suggest you to use images of the dry season in order to differentiate the water body in the river from other land covers provided that the target rivers flow throughout the year. Again, if the rivers are wide enough to be seen on the satellite image according to the resolution, you can easily interpret the rivers by displaying the image in the NIR band or you can also calculate the NDVI to easily identify water bodies and you can use supervised classification followed by image difference to detect the changes over time. However, if the rivers are too narrow which cannot be seen on the image, i suggest you to make field survey to digitize using GPS and then overlay on the image.Following
