Each part of your theoretical model should be justified according to analogous research, or alternatively you can explain why you think they are wrong and testing for something different.
If you don't enter with a strong theoretical model, justified using existing analogous research, then you run a very high risk of trying 20 models and getting a result that is 95% likely to pass statistical threshold by random chance.
Anyone can construct a dataset and pick it apart until you find a p < 0.05 somewhere, somehow. That's why you need to justify it theoretically first. Justify the theory before crunching numbers so you don't find yourself in a situation of mining the data and arriving at results with fake statistical certainty. imoFollowing

In short I am getting a bit over whelmed with the difference in nomenclature used by the Gaussian output, text books, and people's suggestions.
I am trying to pin point the most energetically favourable conformation between two geometry optimised structures (A and B). Both structures are in a minumum with no imaginary frequencies. What 'value' would be the best to report. Here is an example of where I am looking in a Guassian log file after a frequency calculation.

I want to use Clauser method to predict zero pressure gradient boundary layer characteristics in our wind tunnel to be used for a baseline in adverse pressure gradient turbulent boundary layer experiment. I tried to used wall skin friction for pipe as suggested in many books for an assumption but the result seems to have a margin of error.
If the aim is to determine the skin friction from available data, there are a number of methods besides the Clauser chart method that could be used. Few are referenced here:

- Do you have any tips for hMSC culturing and subculturing?
I have recently started working with hMSC's (Bone Marrow-Derived). My issue is when I culture from stock they seem okay, but once I sub-culture it I see lot of cell death and debris. Also several of the live cells have vacuoles because my experiment requires a study on actin cytoskeleton of hMSC's. I feel the presence of vacuoles will not help my aim.
Any suggestions are welcome.

ISSUE: Validity of an instrument & reliability of an instrument
The issue of this question requires a careful reading. The questioner is not asking about the validity or reliability of the “data used in the audits.” She asks about validity and reliability of the “instrument for the audit reports.” It is the “audit reports” that are the secondary data, not the financial statements, but the reports themselves. The instrument is the “what produce the audit report”---thus, the instrument is the audit methodology.
INSTRUMENT: The instrument of this case is the “audit methodology.” The data that the questioner deals with are the reports generated or produced by the auditors. The Audit report is the data. The income statements used in the audit report are not at issue. To rephrase the question: (i) Is the instrument valid, i.e. does the audit methodology produced what it intended to produced? and (ii) Is the instrument reliable, i.e. if an independent auditor engages the audit using the same method, would they produce the same result?
STANDARD OF PRACTICE: If the accounting or financial statements used for the audit were in compliance with IFRS, IAS or GAAP, then the issue of “accounting data reliability” becomes moot. Assume that that IFRS, IAS or GAAP was used. Accounting data reliability is not an issue. Are the questions of validity and reliability test still an issue in this case? The question presented is about “validity and reliability tests of the instrument.” What is the instrument? The data is the audit report. The instrument is the audit methodology used to produce the audit report (data). This different language points to different points of analysis.
SUBJECT MATER: The subject matter for the analysis is not the original accounting data or financial statements generated by the companies. The focal of the analysis is the “accounting reports” themselves. Note the use of the term “reports”---plural. This means that the audits may have come from many auditing firms. Different auditing firms may use different auditing methodology in completing the audit. Therefore, the issue of validity and reliability of the audit result become legitimate questions.
VALIDITY: Validity is the test for precision. Does the data (observation) hit the target as predicted or hypothesized? In this case: does each accounting report confirm the asserted statements? If the audits had been signed off as “passed” audit, then this issue becomes moot because the observed data (reports made by the firm) and the theoretical data (standard of reviewed imposed by auditor) matched---hence the auditor signed it off as passed, i.e. confirmed the asserted statements in the financial statements by using the generally accepted principles and methods in auditing.
RELIABILITY: Reliability is a test for reproduction. The issue is whether if other auditors using the same accepted principles and methods in auditing would produce the same result? If subsequent review by independent auditors under generally accepted principles disagree that the result should have been different then the audit reports does not pass reliability test. Hence, the “audit reports” are the “data” in this case, it is then said that the data does not pass reliability test. Note it is the audit report that is the data, not the financial statement. Then the researcher must look into the sources of disagreement, i.e. auditing method or different treatment of certain categories of event in the accounting at different time period that would have produced different result.
- Have you seen a quenching effect of zinc ionophore on fluorescent intensity?
I am using the ionophore (sodium pyrithione) from Sigma to do several experiments either with zinc indicator or with another fluorescent tagged protein. I am looking at both soma and small cellular structures like presynaptic boutons or postsynaptic spines from dissociated hippocampal culture from rat embryo. When the ionophore was used in soma imaging using FluoZin-3, we did not observe any quenching effect. Instead, there was a proportional increase in fluorescent intensity with increasing zinc concentration. We used 10x objective in this experiment. That was exciting. However, over the past few weeks, it has occurred to me that the application of 2 uM ionophore plus either 10 uM or 300 uM ZnCl2 seemed to quench the intensity of both the dye and the fluorescent protein when looking with high magnification objective (63x). Did any of you see this before? Would you have any suggestion for another ionophore?
