i am currently doing in-vitro proliferation studies using peptides from different sources. So far, i am sticking to the MACS sorted CD4 or CD8 t-cells (purity about 85%).
Is it really necessary to add irradiated feeder cells or can one consider the remaining 15% as enough to present peptides to the t-cells?

the CD3/28 activation is unspecific and always works without extra-presentation by APCs. But i am trying to get peptides to work, and they need to be presented via the MHC molecules to the T-cells.

I need help on suitable equipment to isolate various flavonoids in pure form so as to test it on plasmodium SPP. Which one is the best? Is it TLC or HPLC? I also need a protocol to start with if it TLC or HPLC.
You can isolate flavonoids by HPLC or open chromatographic column (silica gel 60 F254 sds) for Flavonoids apolar, and the silica gel RP18 for polar flavoinoides.

What is causing the low fill factor of MAPbI3-spiro based solar cell, when current is around 16 mA/cm2 and Voc is 0.9V?
I had made some Perovskite solar cells, current up to 16mA/cm2 and Voc of 0.9V was achieved but FF was around 26 which was killing the efficiency, and in different cells that is consistent? can you suggest the possible cause.
Dear Kenneth, thanks for the feedback. Electron transport layer was based on TiO2, and in order to complete the doping of spiro it was left in dry oxygen for overnight (well reported method now). The devices IV curve do have S-Shape curve which can be seen in attached file. I did made the ETL in different ways, though Voc was in the range of 0.85-0.9V for all devices with, Jsc in the range of 14-17mA/cm2, however FF was in the range of 20-25, i am thinking to do metal evaporation in a different chamber just to see if that is causing it.Following

First, test whether the first-order factors are correlated. If this is the case, include the second-order structure. Second, test whether the second-order factors are correlated. If this is the case, include the third-order structure.You would need at least three second-order factors as indicators of the third-order factor. If you only have two second-order factors, fix both loadings on the third-order factor to one. If you have three or more second-order factors, fix the variance of the third-order factor or one of the loadings to one.
When I am running into estimation problems, I usually fix all factor loadings to one - which will of course result in model that does not fit well. Then, these constraints should be relaxed one by one in order to trace the problem.
If you provided the LISREL syntax or the output, I could try to detect the origin of your problem.

I spent quite a lot of time trying to understand why the simulated variability (like El Niño and La Niña events) by coupled Atmosphere-Ocean Global climate models do not occur temporal in phase with reality, but still couldn't understand it. Do you mean the atmosphere runs forced by the observed SSTs? As they are forced by the observed SSTs, the real El Ninos and La Ninas are in your model output. But for coupled models, the atmosphere and ocean are initialized and then the model is freely run, so the initial atmosphere and ocean conditions evolve according to the model physics and generate the models El Nino/La Ninas.
If your high quality atmosphere-ocean initial conditions contain realistic information to produce El Nino/La Nina in the coming months to seasons, CGCM with good physics will produce that event (or maybe one more event after that) close to the reality , but because your model errors will keep grow as Adam said, eventually model will do its own thing.Following

Any suggestions in Western Blot for band extraction after SDS - PAGE and analysis with MALDI - TOF?
I try to prove the presence of the LDL - receptor.
I performed my Western Blot several times, with different antibodies and I always received the same bands.
One of these bands (between 25 and 35 kDa) is very strong and after quite a long research, I found out, that this band is most likely a splice variant of the LDL - receptor.
I would like to verify this assumption by a MALDI - TOF MS analysis. My problem is, that I have no idea, how to extract this specific band from my gele after SDS - PAGE, as there will be many other bands, which will overlap my band of interest. Or is there any fast and easy way to extract the band after incubating the membrane with the AB´s ?
Bastian, what does the gel look like when you stain it with silver and/or Coomassie? If you see the same discrete band there, it is worthwhile to perform an in-gel digest with trypsin and analyze with MALDI-TOF using Peptide Mass Fingerprinting and (instrumentation allowing) confirm some of the assigned peptides by MS-MS or at least PSD.
If the band pattern from a global staining looks completely different then your protein of interest is very probably a minority compound and as such not amenable to PMF or MALDI-TOF. Try and find somebody who runs high-quality nanoLC-MS/MS instead which can resolve up to 150 different proteins in a gel band in many cases. Am not sure who would be able to do this in Bielefeld - the Niehaus group might be able to help.Following
