Can anybody help me on AAS instrument?
we are analyzing Sodium in steam water which OEM spec fall in ppb range. calibration graph is plotted using method of standard addition. please kindly explain me how sample concentration is calculated with using absorbance values for standards and reagent blank and how we can decide whether its times to plot a new calibration graph that mean what happens to a graph when we use it for long period of time.
Method of Standard Addition is a very popular technique in AAS. This is a very valuable technique especially if you observe interference due to matrix. The procedure of standard addition is already discussed by Mr. Najib and hence I am not repeating it. Once you plot the graph, you see whether it is passing through origin(0,0). If the plotted line is passing through origin, then your sample does not have that target element. If your sample contains the target element, then the plotted line will not pass through origin and it will have a Y-intercept. You can then extrapolate the graph and it will touch the X-axis on the negative side. Since the X-axis is the concentration axis, you can easily read the concentration of the element in your sample. I am working on anaerobic baffled reactor to treat paper mill wastewater. I have noticed that the concentration of Total Dissolved Solids (TDS) in the effluent (wastewater after treatment) has increased in compare to the influent (wastewater before treatment).
Hence, I want the understand what do TDS's mean in the anaerobic system? whats the source? what are the key factors which control the concentration?

How can I purify protein with 27 KDa from GST ?
Dear colleagues, i am trying to purify a protein with 26 kDa size that is expressed as a GST fusion protein. When i have done on column GST cleavage in the flow through both GST and my protein of interest coming together. Even i tried Superdex 200 SEC there also proteins are co-eluting. My protein pI is 4.54 and GST pI is 6.09 so i did ion exchange (mono q) with buffer pH 6.5 i eluted by gradient elution with 1M NaCl but here also both the protein coming together. Please suggest some idea to purify the protein. I need this protein for crystallization and structural studies.

I have used an in-house DNA extraction protocol to isolate DNA from young leaf tissues and even though i can see the DNA on the Agarose without much degradation, the bands are not intact (slightly spread). Can any one point to me how i can troubleshooting this to get intact DNA bands?

I have been researching Japan's high dependency on food imports. Japan imports 60% of his foodstuffs which has become an issue of concern among Japanese citizens and for the Japanese government. In this context, the low food self-sufficiency rate of 40% has been discussed as a problem, in particular in the context of the expansion of free trade negotiations Japan has been involved in. I have been asked several times by students whether a low food self-sufficiency rate is really a problem, or if it is portrayed as a problem in order to promote domestic foodstuffs more strongly? Is it mandatory to have the value of Cronbach's alpha above .70?
In my study using questionnaire survey, I have four variables and job satisfaction is one independent variable. I measured job satisfaction with a 3-item scale. Responses were captured in a 5-point LIkert scale. The value of Cronbach's alpha is .65. However, the value of Cronbach's alpha for other variables (captured with 5-point Likert scale are above .70. Can I use the data (with job satisfaction having alpha value below .70) to test my hypotheses?

I made a 10-fold serial dilution of malaria parasite and tested it using PCR. I did a 10 dilutions from 1:10 until 1:10e-10. with a start up parasitemia of 2%. dilutions number 1:10e-7 and 1:10e-8 amplified at the same cycle! any explanations why?

Hi, I have to find out six proteins change due aging in muscle tissue. I am performing western blot right now. Is there another technique available to get accurate protein concentration? I don't have purified proteins to use as standers so can't perform ELISA.
If you do not have a pure protein of interest, you cannot obtain its "accurate" concentration.
It is not clear if you wanted "six techniques to study protein changes" or "changes to six proteins". If it is the former, there are many techniques that address many properties of proteins; some require pure preps while others don't. Over aging, many changes can occur to a protein: the level of expression, post-translational modifications (these can be simply numerous), folding, proteolysis etc. If you know what exactly you are looking for, it will be easier for you to find a way.Following
