  How to generate DA neurons from hES cells through a simple protocol?

  I am puzzled by the countless protocols out there in generating DA neurons from human ES cells... After trying some protocols for a year I still cannot get good results. I wonder if there is Is aI am puzzled by the countless protocols out there in generating DA neurons from human ES cells... After trying some protocols for a year I still cannot get good results. I wonder if there is Is a simple protocol of deriving DA neurons from hES cells. For example, EB stage followed by dissociation of EBs into single cells and plate as a monolayer for terminal differentiation? I feel attaching EBs is so unreliable, but I am not sure if cells will attach as single cells right after the EB stage.

  Mesenchymal Stem Cells CD

  I´m about to start working with Mesenchymal Stem Cells, and I´m going to perform a flux cytometry to determine CD and I would like to know the CDs expected to be found on Mesenchymal Stem Cells. MyI´m about to start working with Mesenchymal Stem Cells, and I´m going to perform a flux cytometry to determine CD and I would like to know the CDs expected to be found on Mesenchymal Stem Cells. My knowledge tells me that Mesenchymal Stem Cells express CD166, CD105, CD90, CD44, CD31, CD13. I hope you could help me if I´m correct with this CD or if I'm mistaken and should search for other specific CDs?

  I am working on epithelial cells right now, and I am facing some isolation problems. So I ask to anybody who works with primary epith. cells: which enzyme do you successfully (high vitality,fast attachment to the plate,acrtive growth and expansion) use to detach epith cells from the basal lamina they sit on? Trypsin? collagenase? dispase? combination of them? I need to get only the epith cells not the MSC cells of the stroma on the other face of the basal lamina.

    I don't mean keratinocytes , but are similar as epithelial cells, your protocol might be a good approach for my cells as well! thank you very much

    You can try actin. Its a good choice for studying it while doing RT-PCR or general qPCR for gene expression studies. Regards, Gaurab
