I am designing a substrate for a nuclease assay, and want to utilize the formation of a hairpin in an oligo to remove the tedious purification steps that come from annealing two strands. Currently, I have a TTTT loop that is flanked by complementary DNA that should form 16 base pairs, with about 60% GC content. Does anyone out there use a similar substrate and have some wisdom to share? I am mostly concerned with how high a concentration I can get away with during annealing, and the best salts & salt concentrations to use. I am currently annealing at 2uM in TE with 100 mM KCl.

Now, if I cultivate the S.pasteurii in an Acetate YE medium, is it possible to use the same equation to estimate the number of cells according to the OD readings?

I'm trying to dissolve the inclusion body that contains rPA but my results are not good. For instance I've used a buffer consisted of 4 M Urea, 50mM 2-mercaptoethanol , 200 mM NaCl , 1mM EDTA and 50mM Tris base. The pH of the buffer is 9 but the final concentration of protein is 0.7 mg/ml. I've read different papers and I think this buffer should be able to solubilize any protein. I saw a paper that argues that NaCl may lead to salting out of soluble rPA. Could it be NaCl that prevents solubilization?

I'm using this medium: (Chung et al. 2010)
(per liter): 50 g glucose, 1.5 g MgSO4·7H2O, 0.2 mg Biotin, 3 g KH2PO4, 15 g (NH4)2SO4, 2.0 mL of trace salts stock solution and 1.0 mL of a solution containing 2 g/L biotin in 1M NaOH. The trace element solution contained (per liter) 5 mL H2SO4, 4.0 g CuSO4·5H2O, 14.0 g MnSO4·H2O, 2.6 g Na2MoO4·2H2O, 4.0 g H3BO3, 4.0 g CoCl2·6H2O, 22.0 g ZnSO4·7H2O, 55.0 g CaCl2·2H2O and 37.5 g FeCl3·6H2O.
But there is a problem with trace solution. Addition of ZnSO4.7H2O(22 g/lit) and CaCl2.2H2O(55 g/lit) produce a solid precipitate. What should I do?

I have a control problem that involves two ellipsoids named as ROC and ROD, in my algorithm the condition of stability is as following: the ROC should be in ROD, we know that ROC and ROD in the worst case have overlap with each other. We need to enlarge ROD such that ROC is in that and this should be optimized. I have some questions, as follows:
1- How to detect the distance of ellipsoids when one ellipsoid is completely in the other or when they have overlap? (except contact points that distance is zero)
2- When two ellipsoids have overlap how can I enlarge one ellipsoid such that it touches the second ellipsoid?
3- When two ellipsoids have no contact (they are not separated) how can I change the bigger ellipsoid such that it touches the inner ellipsoid?

What is the total power use for 1-cycle, 3-cycle?
Cycle:
1. start from room temp. 25C to 100 C,
2. pressure start increase from 0 to 1.7kg/cm2 at 100C,
3. maintain sterilization at 121C, 1.7kg/cm2 for 15 min.
4. pressure released to 0 and temp drop to 100C
5. temp further drop to 50C (keep warm status)

I had performed cellulase activity checking based on Ghose, T.K. (1989) protocol. The enzyme activity was determined at 121.0 FPU/ml (at 0.0030 Ezy concentration).
1. What does it mean?
2. How many ml of enzyme (at 0.0030 enzyme concentration from 1:20 stock dilution) that need to add-in to degrade 5% (w/v) biomass at final volume of 50ml?
