  • Article: X-ray and DFT Study of Glaucocalyxin A Compound with Cytotoxic Activity

    ABSTRACT: The title compound glaucocalyxin A (1) (7α, 14β-dihydroxy-ent-kaur-16-en-3,15-dione) isolated from the leaves of isodon excisoides was characterized by IR, 1H NMR, 13C NMR, 1H-1H COSY, HMQC, HMBC, and EIMS, and its crystal structure was determined by single-crystal X-ray diffraction. The X-ray crystal structure revealed that the molecular backbone of the chosen crystal is a tetracyclic system, including three six-membered rings and a five-membered ring, and the three six-membered rings are in a chair-like conformation. The five-membered ring adopts a twisted envelope-like conformation, and its geometrical parameters were compared with theoretical calculations at the B3LYP and HF level of theory. The molecules form extensive networks through the intra- and intermolecular hydrogen bonds. The experimental NMR data were interpreted with the aid of magnetic shielding constant calculations, by means of the GIAO (gauge-lncluding atomic orbitals) method. Calculated and experimental results were compared with a satisfactory level of agreement. Molecular electrostatic potential map was used in an attempt to identify key features of the diterpenoid glaucocalyxin A that is necessary for its activity. Calculations of molecular electrostatic potential and stabilization energies suggest that the protonation of glaucocalyxin A will be able to occur on carbonyl oxygen atoms.

    ABSTRACT: To investigate the chemical constituents of Ligularia xanthotricha. Silica gel column chromatography and preparative TLC were employed for the isolation and purification. The structures were identified on the basis of spectral data (IR, EI-MS, 1H-NMR, 13C-NMR, DEPT) and chemical evidence. Seven compounds were isolated and identified as follows: lupeol (1), lupeol palmitate (2), 3, 28-dihydroxyl-lupeol (3), betulinic acid (4), taraxasterol (5), taraxasteryl palmitat (6) and taraxasteryl acetate(7). All the compounds were isolated from this plant for the first time.

The structures were identified by IR, MS, 1D/2DNMR spectral data and X-ray single crystal diffraction and other methods1. Nine compound were isolated and identified as 8beta-hydroxyeremophil-7(11)-ene-12, 8alpha(4beta, 6alpha)-diolide (1), 8beta-methoxyeremophil-7(11)-ene-12, 8alpha(4beta, 6alpha)-diolide (2), petasin (3), isopetasin (4), liguhodgsonal (5), ligudentatol (6), ligujapone (7), lupeol (8) and lupeol palmitate (9). Compounds 2, 3, 4, 6, 7 and 9 were isolated from the plant for the first time.

The chemical constituents were isolated and repeatedly purified on silica gel column and the structures were elucidated by the NMR spectra and physicochemical properties. Eight flavones were obtained and they were identified as isosakuranetin, 7, 4'-dimethylaro madendrin, acacetin, cirsimaritin, rhamnetin, eupatolin, 5, 7, 2', 4'-tetrahydroxy-6, 5'-dimethoxyflavone, hyperoside. All the compounds were obtained from herbs of A. ordosica for the first time.

Various chromatographic techniques were employed for the isolation and purification of the chemical constituents. The structures were elucidated by chemical and spectral analysis. Seven flavonoid compounds were isolated and identified as apigenin, eupatilin, chrysoeriol, quercetin-3, 3'-dimethyl ether, quercetagetin-3, 6-dimethyl ether, quercetagetin-3, 6-dimethyl ether-7-O-beta-D-pyranoglucoside and quercetagetin-3, 6-dimethyl ether-4'-O-beta-D-pyranoglucoside, respectively. All of these flavonoid compounds were isolated from the plant for the first time.

Compounds I, II, IV, V and VI were isolated from R. japonica var. galaucocalyx for the first time.

  • Article: [Ion solvation and ion association in high-concentration of NaBF4/DMF solutions].

    ABSTRACT: Ion solvation and ion association in solutions of NaBF4/DMF were studied by infrared and Raman spectroscopy and ab initio quantum chemistry. It was found that the sodium cation interacts on oxygen atom of C=O group in DMF molecules. The solvation number of Na+, four, was obtained according to the change analysis of CH3 rocking band. The splitting of the nu1 mode of BF4(-) anion indicates the presence of contact ion pairs. Ionic association degree increases with increasing molality of the salt. The solvated structure of Na+ and the geometry of Li+ BF4(-) ion pair were also suggested based on the ab initio calculations.

Compound IV was found from genus Lamium for the first time and the rest of the compounds were found from Lamium maculatum var kansuense for the first time.

These constituents were obtained from E. songoricum for the first time.

    Article: Isomeric distribution of conjugated linoleic acids (CLA) in the tissues of layer hens fed a CLA diet.

    ABSTRACT: The isomeric distribution of conjugated linoleic acids (CLA) in the tissue lipids of hens in relation to that in the diet was examined. Silver-ion high-performance liquid chromatography was used to quantify individual CLA isomers in total tissue lipids, phospholipids, and triacylglycerols. It was found that the deposition of CLA isomers in hen tissues was selective. All tissues including serum, liver, heart, kidney, abdominal fat, and leg and breast muscles had lesser amounts of total cis/trans isomers ranging from 75.87 to 89.13% of total CLA, which was in contrast to the value of 92% of total CLA in the dietary lipids. Total trans/trans isomers in all tissue lipids ranging from 6.11 to 18.02% of total CLA were greater than that in the diet (4.19%). Among the individual trans/trans isomers, all tissues except for adipose tissue and brain incorporated greater amounts of t-12,t-14-18:2, t-11,t-13-18:2,t-10,t-12-18:2, t-9,t-11-18:2, and t-18,t-10-18:2 compared with the values of the diet. Within the cis/trans group, lesser amounts of c-10,t-12/t-10,c-12-18:2 were found to incorporate into all tissues compared with the value of the diet. Serum and liver had higher percentages of c-9,t-11/t-9,c-11, whereas the other tissues had similar levels of this isomer compared with that of the diet. It was also observed that supplementation of CLA in the diet of layer hens decreased the concentration of docosahexaenoic acid (22:6n-3) in all of the tissue lipids. It is concluded that dietary CLA can transfer to the tissue but that incorporation of CLA isomers into the tissue is selective in hens.

    ABSTRACT: To provide the foundation for reasonable utilization by analyzing the essential oils of Ligularia virgaurea. The essential oils were extracted by using steam distillation and separated with GC capillary columns. The components were quantitatively determined with normalization method, and were identified with GC-MS. 41 components were identified, which took up 72.73% of the essential oils. The main components of essential oils were 4-methyl-1-(1-methylethyl)-3-cyclohexen-1-ol(14.369%), crotonic acid, 2,2-dimethyl-butanoic acid, 1-methyl-3-(1-methylethyl)-benzene, (1s-endo)-1,7,7-trimethyl-bicyclo[2,2,1]heptan-2-ol, trans-1-methyl-4-(1-methylethyl)-2-cyclohexen-1-ol, alpha-cadinol and alpha,alpha,4-trimethyl-3-cyclohexene-1-methanol.

Six flavonoid compounds were isolated and identified as kaempferol, tiliroside, quercetin, quercetin -3-O-beta-D-glucosides, scutellarin, 5,7-dihydroxy-8-methoxyflavone and one pyrone were isolated and identified as helipyrone. All of these compounds was isolated from the plant for the first time.

  • Article: Production of conjugated linoleic acids through KOH-catalyzed dehydration of ricinoleic acid.

    ABSTRACT: Production of conjugated linoleic acids (CLA) using castor oil as starting material involves conversion of ricinoleic acid to methyl 12-mesyloxy-octadec-9-enoate (MMOE) followed by dehydration. This process usually uses 1,8-diazabicyclo-(5.4.0)-undec-7-ene (DBU) as an expensive dehydrating reagent. The present study reports that potassium hydroxide (KOH) can serve as a dehydrating reagent in replacement of DBU. The results showed that conversion of MMOE to CLA catalyzed by KOH was an efficient reaction, with a 77% conversion efficiency at 80 degrees C. The CLA isomeric profile produced in KOH-catalyzed dehydration reaction was similar to that catalyzed by DBU. The CLA mixture produced in KOH-catalyzed dehydration of MMOE at 80 degrees C contained 72% 9c,11t-18:2 and 26% 9c,11c-18:2 while in that catalyzed by DBU, 9c,11t-18:2 and 9c,11c-18:2 accounted for 78 and 16%, respectively. It was found that the temperature of dehydration was an important factor in the determination of CLA isomer composition and yield of conversion. Elevating the temperature from 78 to 180 degrees C decreased not only the conversion efficiency but also production of total c,t-18:2 and c,c-18:2 isomers regardless of dehydration catalyzed by either DBU or KOH. It is concluded that KOH may replace DBU as a dehydrating reagent in conversion of MMOE to CLA when the reaction conditions are optimized.

  • Article: Preferential incorporation of trans, trans-conjugated linoleic acid isomers into the liver of suckling rats.

    ABSTRACT: The present study was designed to compare the conjugated linoleic acid (CLA) isomeric distribution pattern in the liver of suckling rats in relation to those in the milk and maternal diet. Silver-ion HPLC was used to separate individual CLA isomers. It was found that the isomeric distribution pattern in the milk was very similar to that in the maternal dietary fat. However, the CLA isomeric distribution patterns in the liver phospholipids (PL) and triacylglycerols were different from those in the diet and milk. In the liver PL, total cisltrans isomers accounted for 63.6-63.9% of total CLA, which was in contrast to the values of 88.1-89.1% in the milk and diet. In the liver PL, total transltrans isomers were 20.6-20.8% of the total CLA isomers whereas they were only 2.6-3.7% in the milk and diet. It is concluded that trans/trans-CLA were preferentially incorporated into the liver whereas for the incorporation of cis/trans-CLA there was partial discrimination.
