ABSTRACT: The present study aims to clarify whether hyaluronan binding protein 1 (HABP1/p32/C1QBP) is an indicator of peritoneal and lymph node metastasis in epithelial ovarian cancer (EOC), which to the authors' knowledge is not previously reported by others. Western blot analysis demonstrated that HABP1 was highly overexpressed in most metastatic lesions. Of 89 patients whose primary tumors showed high HABP1 expression on immunohistochemical staining, 85 (95.5 %) presented peritoneal metastases and 43 (48.3 %) had lymph node metastases. Univariate and multivariate logistic regression analyses revealed that HABP1 overexpression correlated with peritoneal dissemination and lymph node metastasis in EOC. The specificity and positive predictive value of HABP1 staining were shown to be better for peritoneal metastasis, while the negative and sensitivity predictive value of HABP1 staining were better for lymph node metastasis. The odds ratio of high versus low staining for peritoneal spread was 9.236 (95 % confidence interval (CI), 2.705, 19.316), and that for lymph node metastasis was 8.614 (95 % CI, 2.507, 21.039). Furthermore, HABP1 protein may potentially be used alone or in combination with other markers as a predictive marker of EOC patients with lymph node metastasis and/or peritoneal dissemination.

ABSTRACT: Immune thrombocytopenia (ITP) is a chronic disease resulting from increased platelet destruction and impaired platelet production. Secondary ITP can be a manifestation of chronic graft-versus-host disease (GVHD) and represent a lymphoproliferative disorder. A boy with chronic graft-versus-host disease after cord blood stem cell transplantation who had severe refractory immune-mediated thrombocytopenia received infusion of rituximab weekly, 375 mg/m2, for 4 weeks. Platelets count of the patient was recovered, and rituximab was well tolerated with no severe toxicity observed during treatment.

ABSTRACT: One polysaccharide PTP was isolated and purified from the roots of Polygala tenuifolia. It consisted of galactose, glucose and galactose in the ratio of 3.1:3.7:2.5, and a small amount of rhamnose, mannose and xylose. 17 general amino acids were identified to be components of the protein-bound polysaccharide analyzed by automatic amino acid analyzer. In order to test the anti-cancer activity of PTP, we investigated its effect against the growth of human ovarian cancer cells SKOV3 in vitro and in ovarian cancer rats. The intracellular reactive oxygen species (ROS) and glutathione (GSH) in SKOV3 cells following PTP treatment were also quantified to explore the possible mechanism underlying the antitumor activity of the polysaccharide. The result showed that PTP is effective on inhibiting the proliferation of SKOV3 cells in a concentration-dependent manner. Furthermore, treatment with PTP caused a rapid depletion of intracellular GSH content and accumulation of intracellular ROS, thus resulting in the apoptosis, which may prove to be a pivotal mechanism for its cancer protection action. In addition, a significant tumor growth inhibition effect was observed in nude mice after PTP administration for 7 weeks. All above indicated PTP could be beneficial towards ovarian cancer therapy.

ABSTRACT: In this study, we prepared an acidic polysaccharide (CPPA) from the roots of Codonopsis pilosula. The effects of CPPA on tumor cell growth, invasion, and migration were examined in vitro. The CPPA not only induced a potent inhibitory effect on the invasion and migration potential of human epithelial ovarian cancer HO-8910 cells in vitro, evaluated by wound healing, transwell and cell adhesion assays, but also had an efficient anti-proliferation effect on tumor cells. Moreover, the CD44 expression on the HO-8910 cells was also attenuated by CPPA treatment. Therefore, our results indicate that CPPA may be a potential candidate compound for the prevention of tumor metastasis, presumably by inhibiting invasion, migration and adhesion of tumor cells, as well as the CD44 expression on the tumor cells.

ABSTRACT: A model was developed that permits computation of CCT diagrams for surface oxidation catalyzed heterogeneous nucleation of alloy droplets from a few experimental data. The model addresses the progress of droplet surface oxidation during continuous cooling in terms of oxide nucleation and growth kinetics. Avrami-corrected surface oxidation kinetics are adopted to account for the impingement of oxide islands growing on the droplet surface during continuous cooling. The model predicts droplet nucleation kinetics in relation to the oxidation potential in the atmosphere, droplet diameter, cooling schedule and four temperature-independent constants. CCT diagrams were computed for Sn-5 mass%Pb droplets cooling in nitrogen atmospheres with various oxidation potentials using nucleation data and cooling simulation of traveling mono-size droplets. Calculated CCT diagrams show that droplet surface oxidation sharply increases the droplet nucleation temperature particularly at high cooling rates. Computation can be extended to predict CCT curves for droplet nucleation in industrial thermal spraying processes.
