    ABSTRACT: To investigate the expression of activating receptor NKG2D(natural killer group 2,member D) on peripheral blood mononuclear cells(PBMCs) in patients with oral squamous cell carcinoma(OSCC) and its clinicopathological significance. The expression of NKG2D on PBMCs in 54 OSCC patients and 20 healthy controls were assessed by flow cytometry. The data was analyzed with SPSS16.0 software package for independent-sample t test and one-way ANOVA test. The positive rate of assay for NKG2D on PBMCs in OSCC patients (12.49±5.26)% was lower than that of the controls (22.93±8.14)% (P<0.05). And which was significantly different among different clinicopathological parameters such as tumor size and disease stage(P<0.05), but no significant difference was found among different gender, age, tumor location, tumor differentiation and cervical lymph node status (P>0.05). The NKG2D expression on PBMCs in OSCC patients was lower than that of healthy controls and was correlated with tumor size and disease stage.     ABSTRACT: In the title compound, C(17)H(14)N(2)O(2), the central pyridazine ring forms dihedral angles of 47.29 (5) and 88.54 (5)° with the benzene rings, while the dihedral angle between the benzene rings is 62.68 (6)°. In the crystal, molecules are linked by two weak C-H⋯O hydrogen bonds and three weak C-H⋯π inter-actions.

    ABSTRACT: To examine the expression of soluble major histocompatibility complex class I-related chain A (sMICA) in the serum in patients with oral squamous cell carcinoma (OSCC) and to explore its clinicopathological significance. Seventy-eight OSCC patients were selected as an experiment group, and 19 healthy persons as a control group. The sMICA in the serum in the experiment group and the control group was detected by enzyme-linked immunosorbent assay. The detection rate of sMICA in the serum in the experiment group was 98.7% (77/78), with the 95% confidence interval 74.30-93.95 pg/mL and the median 82.17 pg/mL, The detection rate in the control group was 94.7% (18/19), with the 95% confidence interval 29.48-50.30 pg/mL and the median 37.54 pg/mL. The sMICA in the serum in the experiment group was higher than that in the control group (P<0.01). There was statistic difference in the serum sMICA in the experiment group among the different clinicopathological parameters such as tumor size, disease stage and regional lymph node status (P<0.05), but no difference was found in gender, age, and tumor differentiation (P>0.05). The sMICA in the serum in the OSCC patients increases, and is related with the tumor size, disease stage and regional lymph node status. Determination of sMICA in the serum may provide useful information to evaluate the immune state of OSCC patients.

    ABSTRACT: To investigate the vaccine potency of MHC class I chain-related gene A (MICA) modified oral squamous cell carcinoma cells. Oral squamous cell carcinoma Tb cells transfected with eukaryotic expression vector pEGFP-N1-MICA and overexpressing MICA protein were inactivated by 120Gy irradiation and vaccinated human peripheral blood leucocytes reconstituted SCID (Hu-PBL/SCID)mice via intra-peritoneal injection, and the non-transfected or blank vector transfected Tb cells were used as the controls. The inhibition effect on tumorigenicity of subcutaneously challenged Tb cells in vaccinated Hu-PBL/SCID mice was detected.The expression of NKG2D and the cytotoxicity in vitro to Tb cells of peripheral blood mononuclear cells (PBMCs) and spleen cells were measured by flow cytometry and lactate dehydrogenase (LDH) release assay. SPSS16.0 software package was used for statistical analysis. MICA gene modified Tb tumor vaccine resulted in remarkable loss of tumor size and tumor weight in vaccinated Hu-PBL/SCID mice. Flow cytometry and lactate dehydrogenase (LDH) release assay showed MICA gene modified Tb tumor vaccine up-regulated the expression of NKG2D on PBMC and spleen cells and enhanced the cytotoxicity to tumor cells. Significant difference was found between MICA-transfected vaccine and non-transfected and blank vector-transfected vaccine (P<0.05). MICA gene modified oral squamous cell carcinoma vaccine can enhance the ability of antitumor immune response,and MICA may be considered as a promising immunotherapy target of oral squamous cell carcinoma.

    ABSTRACT: To investigate the effect on natural killer (NK) and cytotoxic T lymphocyte (CTL)-mediated cytotoxicity by genetic overexpression of MHC class I chain-related protein A (MICA) in oral squamous cell carcinoma (OSCC). The OSCC cells by genetic overexpression of MICA were detected to identify the biological features including cell growth curve, cell cycle distribution, plate clone forming rate and tumorigenicity in nude mice. The expression of natural killer group 2, member D (NKG2D) receptor and the cytotoxicity to target tumor cells of NK92 and CTL cells, which co-cultured with the transfected OSCC cells or the non-transfected or blank vector-transfected controls, were measured by flow cytometry and lactate dehydrogenase (LDH) release assay. There was no difference in biological features before and after MICA gene transfection to OSCC cells. Flow cytometry and LDH release assay showed that MICA-overexpressed OSCC cells enhanced the cytotoxicity to target tumor cells and up-regulated the expression of NKG2D on NK92 and CTL (P<0.05). MICA may be considered as a promising immunotherapy target of OSCC.

    ABSTRACT: The current research concerns the clinicopathological significance of MHC class I chain-related protein A (MICA) expression in oral squamous cell carcinomas (OSCCs). The expression and location of MICA protein in 14 normal oral mucous and 45 cancerous and para-cancerous tissues were assessed by immunohistochemistry and levels of MICA mRNA expression in 29 cancerous and para-cancerous tissues were determined by the real-time polymerase chain reaction. Data were analyzed with the SPSS16.0 software package. MICA was found to be located in the cytoplasm and plasma membrane. Expression was higher in para-cancerous than in cancerous tissues (P<0.05). However, no statistical difference was found between the following: 1) para-cancerous tissue with normal mucosa; 2) normal mucosa with cancerous tissue;and 3) among different clinicopathological parameters in OSCC (P>0.05). The level of MICA mRNA was higher in OSCCs than in para-cancerous tissues, and was correlated with the regional lymph node status and disease stage (P<0.05). The levels of MICA protein and mRNA expression differ among normal oral mucosa, para-cancerous tissue, and cancerous tissue. MICA may contribute to the tumorigenesis and progression of OSCC.

    ABSTRACT: To construct the eukaryotic expression vector, encoding major histocompatibility complex class I-related chain A gene (MICA), for the further research of transfecting Tca8113-Tb cell line(a metastatic cell line of brain metastasis from human tongue cancer Tca8113 cells in nude mouse), and to establish a stable MICA overexpression oral squamous cell line. cDNA of MICA gene from pCMV-SPORT6-MICA was amplified by PCR, and subcloned into eukaryotic expression vector pEGFP-N1 marked with green fluorescent protein (GFP). The recombinant plasmid was sequenced and transfected into Tca8113-Tb cell line by lipofectamine 2000. After screen culture by G418, stable tranfected Tca8113-Tb cell line was established using definite dilution method. The expressions of GFP protein was viewed directly with fluorescence microscopy and the overexpression of MICA was identified by RT-PCR, real time PCR and immunocytochemistry. The MICA gene was amplified by PCR and then cloned into the vector, whose sequence was identical to that in the GenBank. The transfected cells showed the expression of GFP. And the overexpression of MICA gene in transfected cells was detected by RT-PCR, real time PCR and immunocytochemistry. The recombinant eukaryotic expression vector pEGFP-N1-MICA has been constructed successfully and stably expressed in Tca8113-Tb cell line, providing a foundation for further studies on the function of MICA in vitro.

    ABSTRACT: The aim of this study was to evaluate the clinical effect of heterogeneous acellular dermal matrix in the surgical treatment of advanced oral submucous fibrosis (OSF). There were eight patients who had undergone surgical treatment of trismus caused by OSF. Surgery was performed under general anaesthesia given through a nasoendotracheal tube using a fibreoptic bronchoscope. All the fibrous bands on the buccal mucosa were incised and bluntly dissected to stretch the mouth opening. Based on the defect, heterogeneous acellular dermal matrix graft was applied directly on the bilateral wounds. A tie-over dressing technique was used to hold the graft firmly to its bed.The sutures and dressing material were removed on the tenth to fourteenth day after the operation. All the patients were asked to stretch the mouth opening 3 times daily and were followed up for at least 6 months. A modality of medical treatment including multiple micronutrient supplements and steroid injection therapy were carried out if necessary. The changes in the interincisal distances (IDs) were analyzed in the preoperative, intraoperative, and postoperative stages. The data was analyzed with SPSS16.0 software package for one-way ANOVA. The overall success rate was 100% without any or only partial graft loss. There were no immunologic reactions or significant complications.The mean preoperative, intraoperative, and final follow-up interincisal distances were 12.04, 35.46, and 29.33 mm, respectively. Evaluation of the changes in the IDs revealed statistically significant differences between the values recorded at the different stages.(P<0.05). Heterogeneous acellular dermal matrix may be a valuable biomaterial for repairing oral mucosal defects after surgery on the fibrous bands in patients with advanced OSF through covering and protecting wound surface early,which shortens the healing time of wound, decreases relapse of fibrosis and improve the restricted mouth opening.

    ABSTRACT: Cervical neurilemmoma originating from cervical spine could extend through foramen intervertebrale and displayed a radiographic features as a cervical intra- and extra-spinal dumbbell-shaped mass.Because of its early compressive spine cord, patients usually visited neurosurgery at first. In this paper, two patients of cervical intra- and extra-spinal dumbbell-shaped neurilemmoma visiting oral and maxillofacial surgery complained of upper cervical mass were reported, aiming to improve the diagnostic and therapeutic skill for oral and maxillofacial surgeons. Two case of cervical dumbbell tumors who complained of a neck mass underwent surgery and were followed up by oral and maxillofacial surgeons .The pathogenesis,operative approach and prognosis were discussed by reviewing relevant literatures retrospectively. Both of the two patients' presenting symptoms were upper-cervical masses. Preoperative imaging suggested trans-cervical vertebra dumbbell masses.The tumors were removed through combining the posterior midline and lateral cervical approach, and a gross total resection was achieved.No evidence of recurrence was noted after one-year of follow-up. The initial symptom of cervical intra- and extra-spinal dumbbell-shaped neurilemmoma may be a cervical mass and inconspicuous compressive spine cord. Analysis is essentially devoted to the radicality of tumor resection, nerve root preservation, relation to the vertebral artery, and compromise of spinal stability.The surgical resection of dumbbell-shaped cervical spinal neurilemmoma can pose a formidable challenge.
