  • Article: Bigelovii A induces apoptosis of HL‑60 human acute promyelocytic leukaemia cells.

    ABSTRACT: In the present study, the antitumor effects of the nor‑Oleanane type triterpene saponin, Bigelovii A, isolated from Salicornia bigelovii Torr, were examined. Bigelovii A was demonstrated to inhibit HL‑60 human acute promyelocytic leukaemia cell growth with an IC50 value of 2.15 µg/ml. In addition, Bigelovii A promoted apoptosis in HL‑60 cells, as shown by apoptotic morphological changes and the hypodiploid cell assay. Apoptotic induction by Bigelovii A was associated with the downregulation of Bcl‑2, the upregulation of Bax and the activation of caspase‑3, as demonstrated by RT‑PCR and western blot analysis. In addition, a lactate dehydrogenase release test indicated that Bigelovii A may exhibit cytotoxic activity by the induction of cell membrane impairment. This study is the first to identify that Bigelovii A exhibits potential antitumor activity and induces marked apoptosis and membrane permeabilisation in HL‑60 cells. Bigelovii A may be a novel candidate for future cancer therapy.

  • Article: Another two novel ceramides from Zephyranthes candida

    ABSTRACT: From the the bulbs of Zephyranthes candida (Amaryllidaceae), another two novel ceramides have been isolated and identified. The structures of the two novel compounds were established as (2S,3S,4R,21E,2′R)2-[N-(2′-hydroxynonadecanoyl)-N-(1′′,2′′-dihydroxyethyl)amino]-21-hexacosene-1,3,4-triol, named zephyranamide C (1), and 1,3,4,5,6-pentahydroxy-2-(2′-hydroxyhexacosanoyl-amino)-18-(E)-tetracosene, named zephyranamide D (2). Their structures and stereochemistries were elucidated by spectral techniques including 1H NMR, 13C NMR, as well as HSQC, HMBC, DEPT, and COSY.     ABSTRACT: Two new coumarin biosides, tert-O-β-D-apiofuranosyl-(1→6)-O-β-D-glucopyranosyl-byakangelicin (1) and 2′-O-β-D-apiofuranosyl-(1→6)-β-D-glucopyranosyl-peucedanol (2), were isolated from the fresh roots of Angelica dahurica. The structures of the new compounds were elucidated on the basis of spectral analysis.

  • Article: Two new nortriterpenoid saponins from Salicornia bigelovii Torr. and their cytotoxic activity.

    ABSTRACT: Investigation of characteristic constituents of Salicornia bigelovii Torr. led to isolation of two new 30-nortriterpenoid glycosides, Bigelovii A (1), Bigelovii B (2), together with two known 30-nortriterpenoid glycosides 3-4 and three known oleanane-type triterpenoid glycosides 5-7. The structures of new compounds were elucidated by extensive 1D and 2D NMR, and MS spectroscopic analysis, and chemical evidences. All compounds were isolated for the first time from Chenopodiaceae. Thus compounds 1-4 were evaluated for their cytotoxicity and compouds 1, 3 showed moderate activity against four cell lines, HL-60 (promyelocytic leukemia), MCF-7 (breast carcinoma), HepG2 (liver carcinoma) and A549 (lung carcinoma), with IC(50) values of 6.18, 78.08, 13.64 and >100μM for 1; 31.87, >100, ~100, >100μM for 3, respectively.

    ABSTRACT: To study the water-soluble chemical constituents of the flower buds of Lonicera macranthoides. The chemical constituents were isolated and purified by means of chromatographic techniques and their structures were elucidated by spectroscopic methods. Six compounds were isolated and identified as trans-linalool-3,7-oxide-6-O-beta-D-glucopyranoside(1), secol oganoside(2), secoxyloganin(3),chlorogenic acid(4), caffeic acid(5), sucrose(6). Compound 1 is isolated from nature as a single compound for the first time, compounds 2-3 are isolated from this plant for the first time.

    ABSTRACT: To research the chemical constituents from stems and leaves of Lonicera macranthoides. Various column chromatographies were employed to isolate and purify the constituents. Their structures were elucidated by spectral analysis (IR, MS, 1H-NMR, 13 C-NMR) and chemical evidence. Nine constituents were obtained and identified as loganin (I), loganic acid (II), morroniside (III),7-O-ethyl-morroniside (IV), scopoletin (V), caffeic acid (VI), chlorogenic acid (VII), beta-sitosterol (VIII), daucosterol (IX). Compounds I-VI are isolated from the plant for the first time. All the compounds are found for the first time from the stems and leaves of Lonicera macranthoides.

Dried aerial parts of Bupleurum chinense were extracted with EtOH, the chemical constituents in the extract were isolated and their structures were identified by physical and chemical properties and spectral analysis. Five compounds were obtained and identified as 1-O-caffeoylglycerol (1), esculetin (2), scopoletin (3), kaempferol-3-O-beta-D-rutinoside (4) and alpha-spinastero (5). Compounds 1-3 are obtained from Bupleurum chinense for the first time.

    ABSTRACT: Two novel ceramides, Candidamide A (1) with a phytosphingolipid structure, and Candidamide B (2) with a tertiary amide structure, together with 12 known compounds (3-14) have been isolated from the bulbs of Zephyranthes candida, The structures of 1 and 2 have been elucidated to be 1,3,5,6-tetrahydroxy-2-(2'-hydroxytetracosanoyl amino)-8-(E)-octadecadiene (1) and (2S,3S,4R,8E,2'R)-2-[N-(2'-hydroxyoctadecanoyl)-N-(1'',2''-dihydroxyethyl)-amino]-8-hexacosene-1,3,4-triol (2) on the basis of spectroscopic evidence including IR, MS, NMR ((1)H-NMR, (13)C-NMR, DEPT, (1)H-(1)H COSY, HSQC, HMBC). The known compounds were identified as (2S)-3',7-dihydroxy-4'-methoxyflavan (3), (2S)-4'-hydroxy-7-methoxyflavan (4), (2S)-4',7-dihydroxyflavan (5), 7-hydroxy-3', 4'-methylenedioxyflavan (6), ambrettolide (7), beta-sitostero1 (8), beta-daucosterin (9), rutin (10), pancratistatin (11), lycorine (12), haemanthidine (13), and haemanthamine (14). In the antimicrobial assay, candidamide A (1) and candidamide B (2) displayed moderate activities against bacteria Staphylococcus aureus and Escherichia coli, and fungi Aspergillus niger, Candida albicans and Trichophyton rubrum.

Their structures were elucidated by chemical and spectral methods. Eight compounds were isolated from the whole plant of Zephyranthes candida, and identified as (2S)-3',7-dihydroxy-4'-methoxyflavan (I), beta-sitosterol (II), stigmasta-5,22-dien-3beta-ol (III), stigmasterol (IV), beta-daucosterin (V), Butylisobutyl phthalat (VI), Heptacosane (VII) and Ambrettolide (VIII). I-XIII are all isolated from this plant for the first time.

Fresh roots of Angelica pubescens were extracted with EtOH,the chemical constituents in the extract were isolated by column chromatography as macroporous absorption resin,and ODS etc. Their structures were confirmed by spectral analysis (NMR, MS). Seven compounds were isolated and their structures were identified as nodakenetin (I), umbelliferone (II), columbianetin (III), beta-D-glucosyl-columbianetin (IV), columbianin (V), tert. -O-beta-D- glucopyranosyl -(R)-by-akangelicin (VI) and sec. -O-beta-D- glucopyranosyl-(R) -byakangelicin (VII). Compound IV, VI and VII are isolated from Angelica pubescens for the first time.
