ABSTRACT: To investigate the correlation between corneal biomechanical properties, applanation tonometry and direct intracameral tonometry.
Patients scheduled for phacoemulsification were enrolled in this prospective observer-masked study. Central corneal thickness (CCT) was obtained with ultrasound pachymetry. Corneal biomechanical properties including corneal hysteresis (CH) and corneal resistance factor (CRF) were measured with the ocular response analyser. Applanation intraocular pressure (IOP) (IOPappla) was measured in the supine position. Intracameral IOP (IOPintra) was measured with a pressure transducer connected to a cannula inserted into the anterior chamber.
Fifty-eight eyes of 58 patients were included. There was a significant difference between IOPappla and IOPintra (17.3±4.1 vs 13.8±4.7 mm Hg, p<0.001). CCT was significantly correlated with CH and CRF (p<0.01). Univariate regression analysis revealed IOPappla was significantly correlated with CCT and CRF (p<0.05). IOPintra had no significant association with the corneal properties (p>0.05). Multivariate regression analysis revealed IOPappla=10.43+2.69CRF-1.99CH (p<0.001) and the difference between IOPappla and IOPintra (ΔIOP)=-1.57+0.55CRF (p=0.032).
The mean difference between IOPappla and IOPintra was 3.5 mm Hg. While IOPappla was correlated with the corneal biomechanical properties of CH and CRF, IOPintra was not. IOPappla was correlated more with the corneal biomechanical properties assessed by the ocular response analyser than with CCT alone.

ABSTRACT: This study was designed to investigate the expression and significance of NET-1 in hepatocellular carcinoma (HCC) and analyze the relationship between NET-1 gene expression and clinicopathologic factors in HCC.
NET-1 gene protein expression was detected by Western blot, fluorescence immunocytochemistry, confocal laser scanning microscopy and immunohistochemistry in 8 cases of HCC tissues, human hepatoma cell line SMMC-7721, and paraffin-embeded sections from 130 cases of HCC.
NET-1 gene protein expressed in 8 cases of HCC tissues by Western blot. The NET-1 gene protein positively located in the cytoplasm as irregular granules near Golgi apparatus in SMMC-7721cells, detected by fluorescent immunocytochemistry and observed by confocal laser scanning microscopy. The positive rate of NET-1 protein expression revealed by immunohistochemistry was 96.9% in HCC (126/130). NET-1 Protein expression in HCC was clearly correlative with HCC cytological variants, there were pronounced higher expressions in clear cell type, pleomorphic cell type, and sarcomatous change than that in hepatocytic type (P < 0.05). NET-1 Protein expression in HCC was positively correlative with the histopathologic grading, clinical stages and HCC with hepatitis and cirrhosis (P < 0.05), respectively, and negatively correlated with the presence of patches of necrosis (P < 0.05). But NET-1 protein expression was not associated with AFP level, tumor size and growth patterns, respectively.
NET-1 protein is expressed in cytoplasm of HCC cells as irregular granules near Golgi apparatus. NET-1 gene expression may promote the uncontrolled proliferation and abnormal differentiation in HCC cells.

ABSTRACT: To investigate the distribution of anterior corneal 4th-order spherical aberration in age-related cataract eyes of Chinese people and analyze its related factors, in order to guide the selection of intraocular lenses with modified spherical aberration profiles.
After outputting anterior corneal curve and height data from the corneal topographic maps (Humphrey Carl Zeiss) of 188 eyes of 144 age-related cataract patients, corneal 4th-order spherical aberration was computed using ray-training calculation programs. Correlation analysis was performed between the spherical aberration and corneal curve, corneal astigmatism, corneal shape factor.
There was wide individual variability in corneal 4th-order spherical aberrations with ranges of individual Zernike terms from -0.096 microm to +0.469 microm. Three eyes had negative spherical aberration. The mean coefficient of the 4th-order spherical aberration was 0.231 +/- 0.092 microm (95% confidence interval [CI] = 0.217 - 0.244 microm). The spherical aberration was neither correlated with corneal curve(r = 0.134, P = 0.067) nor correlated with corneal astigmatism (r = -0.075, P = 0.304). High correlation was found between spherical aberration and shape factor (r = -0.725, P = 0.000).
Anterior corneal 4th-order spherical aberration varied greatly among cataract eyes and some eyes had negative spherical aberration. Spherical aberration was neither correlated with corneal curve nor correlated with corneal astigmatism. But high correlation was found between spherical aberration and shape factor. Customized aspheric intraocular lenses are needed and shape factor can give some advice to select customized intraocular lenses with modified spherical aberration profiles.

ABSTRACT: NET-1 is a new tumor-related gene. So far, its expression in hepatocellular carcinoma (HCC) hasn't been reported. This study was to screen and compare the expression of NET-1 among fetal liver tissue, adult normal liver tissue, HCC and adjacent noncancerous tissues; explore its expression feature in HCC.
The expression of NET-1 mRNA in 3 specimens of fetal liver tissue, 4 specimens of adult liver tissue, 4 specimens of test HCC tissue, and 28 specimens of HCC and adjacent noncancerous tissues was detected by reverse transcription-polymerase chain reaction (RT-PCR). The expression level of NET-1 gene was analyzed by Four-Star image analysis software. NET-1 gene polyclonal antibody was generated by gene biological engineering, and its expression in human hepatic cancer cell line SMMC-7721 was observed under laser confocal microscope. The expression of NET-1 protein in the 28 specimens of HCC and adjacent noncancerous tissues was detected by immunohistochemistry.
NET-1 mRNA was expressed in all of the 4 specimens of test HCC tissue, but didn't express in normal adult and fetal liver tissues; the positive rates of NET-1 mRNA were 85.71% (24/28) in both HCC and adjacent noncancerous tissues. The mRNA level of NET-1 was significantly higher in HCC than in adjacent noncancerous tissues (0.65 vs. 0.47, P<0.05). Observed under confocal microscope, NET-1 antibody was expressed in cytoplasm of SMMC-7721 cells. The positive rate of NET-1 protein was significantly higher in HCC than in adjacent noncancerous tissues (96.43% vs. 71.43%, P<0.05). In both HCC and adjacent noncancerous tissues, the positive rate of NET-1 mRNA was positively correlated to that of NET-1 protein (r=0.48, P<0.05; r=0.40, P<0.05).
NET-1 gene expression might be an early event in HCC development, and may have specific value in HCC diagnosis.
