Article: Characterization and quantification of major constituents of Xue Fu Zhu Yu by UPLC-DAD-MS/MS.

ABSTRACT: Xue Fu Zhu Yu (XFZY), a classic recipe in traditional Chinese medicine (TCM), has been demonstrated to show protective effects on cardiovascular system. For quality control of XFZY products, qualitative analysis using ultra high performance liquid chromatography with diode-array detector-tandem mass spectrometry (UPLC-DAD-MS) was undertaken. Twenty-eight compounds from XFZY were simultaneously detected; among them, seventeen compounds were unequivocally identified, and another eight compounds were tentatively characterized. According to qualitative results, a new method for quantitative analysis of XFZY has been established by ultra high performance liquid chromatography coupled with diode array detector (UPLC-DAD). Twelve representative compounds unequivocally identified were used as chemical markers in quantitative analysis, including 5-hydroxymethyl-2-furaldehyde (5-HMF), hydroxysafflor yellow A (HSYA), amygdalin, albiflorin, paeoniflorin, liquiritin, ferulic acid (FA), naringin, hesperidin, neohesperidin (NH), isoliquiritigenin (IL) and glycyrrhizic acid (GA), which were derived from nine of eleven herbs of XFZY except Platycodon grandiflorum (Jacq.) A. DC. (Jiegeng) and Bupleurum chinense DC. (Chaihu). This UPLC method was validated in terms of linearity, LOD and LOQ, precision, repeatability, stability, and recovery tests. Article: [Variation of 4 kinds of compounds in Psoralea corylifolia processed by different methods].

ABSTRACT: OBJECITVE: To established an efficient method for simultaneous quantification of 7 compounds belonging to 4 chemical types in Psoralea corylifolia processed by different methods, and to elucidate variations of 4 kinds of compounds in different processed P. corylifolia. The chromatographic separation was performed on an ACUITY C18 column using acetonitrile and 0.1% formic acid aqueous solution in the gradient elution at 0.4 mL x min(-1). Detection wavelength was set at 246 nm. Column temperature was fixed at 50 degrees C. The 4 kinds of compounds including psoralenoside, isopsoralenoside (benzofuran glycosides), psoralen, isopsoralen, psoralidin (coumarins), bavachin (flavonoids), and bakuchiol (meroterpenes) were separated in 25 min. The correlation coefficients of those compounds were over 0.9993 in the tested range. The intra- and inter-day precisions were below 1.5%. The average recoveries ranged from 99.2% to 106%. This method is simple, rapid and accurate, which can be used for the determination of the 4 kinds of compounds in different processed P. corylifolia. In P. corylifolia processed by different methods, the contents of benzofuran glycosides, flavonoids, and meroterpenes in P. corylifolia processed by Leigong's method decrease obviously, while the contents of psoralen and isopsoralen increase significantly.Zhongguo Zhong yao za zhi = Zhongguo zhongyao zazhi = China journal of Chinese materia medica 08/2011; 36(15):2071-5. -
Article: [Study on excretion of stilbene glycoside (THSG) and its beta-cyclodextrin inclusion].

ABSTRACT: The excretion characteristics of stilbene glycoside (THSG) and its beta-cyclodextrin inclusion in bile, urine and feces after oral administration to rats were studied. Bile for 24 h, urine and feces for 72 h were collected. The content of THSG was determined by HPLC-UV. The established HPLC-UV method was available for the analysis of THSG in excreta and corresponded to the requirement of biological sample analysis. After given THSG and its beta-cyclodextrin inclusion, the amount of prototype THSG in feces were 3.27% and 0.61%, meanwhile THSG in bile were 0.20% and 0.18%, respectively. Only a little THSG was found in urine. The result showed that beta-cyclodextrin inclusion reduced the fecal excretion of THSG.
