  • Article: Sequence analysis of PAX9, MSX1 and AXIN2 genes in a Chinese oligodontia family.

    ABSTRACT: The goal of our research was to look into the clinical traits and genetic mutations in nonsyndromic oligodontia in a Chinese family and to gain insight into the role of mutations of PAX9, MSX1 and AXIN2 in oligodontia phenotypes. 6 subjects from a family underwent complete oral examination, including panoramic radiographs. Retrospective data were reviewed and blood samples were collected. PCR primers for PAX9, MSX1, and AXIN2 were designed through the Oligo Primer Analysis Software. PCR products were purified and sequenced using the BigDye Terminator Kit and analysed by the 3730 DNA Analyzer. The proband missed 4 permanent canines, 2 permanent maxillary lateral incisors, 2 permanent mandibular lateral incisors, and 2 permanent mandibular central incisors, whilst his maternal grandfather lacked only 2 permanent mandibular central incisors. Moreover, the size of some permanent teeth appeared smaller than normal values of crown width of Chinese people. Oligodontia and abnormalities of teeth were not present in other family members. Radiographic examination showed that the proband and the rest of family members retained all germs of the third molars. There was one known mutation A240P (rs4904210) of PAX9 in the coding region in the proband and the maternal family members (II-2, II-3, and II-4), which possibly contributed to structural and functional changes of proteins. No mutations were identified in MSX1 and AXIN2. Our findings may imply that the PAX9 A240P mutation is a risk factor for oligodontia in the Chinese population. A240P is likely to be a genetic cause of oligodontia though previous literature suggested it as a polymorphism only.

    Article: [Morphology of the soft palate in normal individuals when pronouncing: a digital cephalometric study].

    ABSTRACT: To investigate the morphology of the soft palate in normal individuals with digital radiography, when they pronounced the high vowel of "i", and to provide the references for therapy of the cleft palate. In this study, the sample comprised 27 normal subjects. With the digital cephalometry, the morphology of the soft palate when pronouncing the high vowel of "i" was observed. And the dimensional difference of the soft palate when pronouncing between different gender was studied. When pronouncing the high vowel of "i", the morphology of the soft palate was like the shape of the knee. And it could be divided into two parts: horizontal and vertical. The length of the vertical part in male group was (24.92 +/- 2.03) mm, the length of the vertical part in the female group was (20.66 +/- 2.77) mm. The length of the vertical part was different between male and female group (P < 0.001). The morphology of the palate when pronouncing the high vowel of "i" is similar. And the velar length of the vertical part of the male is longer than the female.

  • Article: [Effect of nano-granule titanium films on synthesis of osteoblasts].

    ABSTRACT: To compare the synthetic ability of osteoblasts on the surface of different nano-granule titanium films and investigate the correlation between nanophase titanium films and cellular biocompatibility. Four different nano-granule titanium films were produced by direct current magnetron sputtering, at ambient, 100 degrees C, 250 degrees C, 380 degrees C substrate temperature, respectively. Rat osteoblasts were seeded on the surface of four treated groups of titanium film samples and non-treated Ti sample(control group). The production of osteocalcin (OC) in all five groups were detected by using double antibody sandwich enzyme-linked immunosorbent assay. The production of OC increased gradually from day 7 to day 14 in all groups. In the control group, it showed significant differences with other five groups on day 7. On day 14, the production of OC in 100 degrees C group was the highest, and it showed significant differences with 380 degrees C, control group and blank group. In 250 degrees C group, the production of OC also showed significant differences with 380 degrees C, control group and blank group (P < 0.05). Titanium with nano-modified surface had good biocompatibility and different nano-granule titanium films could affect the synthesis of osteoblasts.

  • Article: [Analysis of clinical features and mutation associated with oligodontia].

    ABSTRACT: To investigate the mutation characteristics of paired box homeotic gene 9 (PAX9) and muscle segment homeobox gene 1 (MSX1) of patients with congenital oligodontia. Clinical manifestations were recorded by taking complete oral examinations in patients with congenital nonsyndromic oligodontia and some of his normal family members. Pedigree information was confirmed by extended interviews and a pedigree was constructed. Inheritance mode and clinical features were analyzed. Assessment of crown width compared to normal value of crown width in Chinese people was based on the registrations and measurements of study cast. Comparison of craniofacial form, malocclusion types and characteristics were conducted via cephalometric analysis by taking lateral cephalometric radiographics. Venous blood samples were collected and DNA was extracted from leukocytes. DNA sequencing and mutation analysis were analyzed in exon 1, 2, 3, 4 of PAX9 and exon 1, 2 of MSX1 coding region by polymerase chain reaction (PCR). The teeth shape abnormality of the patient was noticed by a measured smaller crown width compared to normal values of crown width in Chinese people. The result of cephalometric analysis indicated no obvious inherited tendency in the proband in terms of facial osseo type and jaw bone pattern. One mutation was found in the proband and his mother in exon 3 of PAX9, the missense mutation G718C causing a conservative change A240P was present. Mutation was not found in MSX1. These findings suggest that the missense mutation G718C in exon 3 of PAX9 is likely the cause of oligodontia.

  • Article: [Construction of nano-granule titanium film and study of its surface topography].

    ABSTRACT: To construct four different micro- and nano-phase titanium film models and investigate the characteristics of their surface micro-topography. Four different titanium films were prepared on commercial titanium discs, by direct current magnetron sputtering, at ambient, 100, 250, 380 degrees C substrate temperature, respectively. Their surface topography and crystal sizes were investigated using atomic force microscope (AFM) and X-ray diffraction (XRD). The size of granule and surface roughness in different group was calculated and compared. All samples were covered by a thin film consisting of dense round or ovaloid granules. The granules and crystals was growing as the substrate temperature increasing. The Ti substrate had greater effect on the surface topography of film compared with Si substrate. This kind of complex topography caused the surface roughness of Ti substrate group decreased as the granules growing. In our study, four different micro- and nano-phase titanium film models were constructed for our coming investigation of their topographical influence on biological reaction of proteins and cells. Basic data on surface features was obtained for next in vitro and in vivo experiment.

  • Article: [Comparative research of craniofacial structural characteristics of syndromic and non-syndromic patients with cleft palate].

    ABSTRACT: To compare craniofacial structural characteristic of syndromic and non-syndromic patients with cleft palate and to probe into the reason of their maxillary hypoplasia and to lay a foundation for better treatment protocol for syndromic patients with cleft palate. The sample consisted of 8 individuals with Treacher Collin syndrome, 9 patients with Pierre Robin syndrome, and 40 patients with non-syndromic cleft palate which met certain criterions. Lateral cephalometric radiographs were obtained from each study subject. A total of 18 variables, comprising 9 angular, 7 linear, and 2 ratio measurements, were studied. SPSS 12.0 software was used to describe the statistics data, consisting of means, SD, and t test. The tendency for smaller value in SNB and larger value in ANB indicate a severer anteroposterior mandibular hypoplasia in the syndromic type cleft. There was no difference between syndromic and non-syndromic cleft palate patients with regards to maxillary anteroposterior development. No significant difference on the development in cranial base area was found either. This interplay of cranio-facial bones is very important in the development of this complex, which will not only affect the growth but also the defect development. In this study, the patients with the two syndromes showed the similar growth potential of maxilla with the non-syndromic cleft patients.

    ABSTRACT: To study the morphology of the soft palate in normal humans with digital radiography and to provide the references for therapy of the cleft. 106 normal people were involved. The morphology of the soft palate was observed with digital cephalometry. All static images of soft palate could be divided into six types: Shuttle-shaped, crescent-shaped, strip-shaped, S-shaped, hamulus-shaped and anomalous shaped. The dynamic image was knee-shaped. The morphology of the soft palate is varied.

  • Article: [Association of estrogen receptor gene polymorphisms and primary trigeminal neuralgia].

    ABSTRACT: To investigate the association of estrogen receptor (ER) gene polymorphism and primary trigeminal neuralgia. By polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), ER gene polymorphism was analyzed in 20 trigeminal neuralgia (TR) patients and 20 control individuals, and the distribution of ER genotype was compared in TR group and control group. There was no significant difference in frequencies of allele and genotype in XbaI or PvuII polymorphism or XbaI with PvuII polymorphisms together between TR group and control group (P > 0.05). The genotypic distribution of Xx or PpXx in TR group was higher than control group, and it was contary to xx, ppxx or Ppxx in TR group and control group. XbaI or PvuII polymorphism may be related to TR. Women with PpXx genotype may be a dangerous factor to primary trigeminal neuralgia.

  • Article: [Expression of Ki-67 antigen in ameloblastoma and its clinical significance].

    ABSTRACT: The expression of Ki-67 antigen of ameloblastoma was examined in order to investigate the different proliferation activity of histological variants of ameloblastoma and its clinical significance. 70 cases of different histological specimen of ameloblastoma were analyzed by immunohistochemical method using Ki-67 antibody. The Label Index was calculated in percentage of Ki-67 positive cells after examined with an image analysis system. The results showed that the Labeled Index in malignant ameloblastoma was the highest 14.72% +/- 2.87%. The Labeled Index in solid ameloblastoma was in the middle, among which the follicular 4.42% +/- 1.05% was higher than the plexiform 3.64% +/- 1.23%. The Labeled Index in mono-cystic ameloblastoma was the lowest 2.21% +/- 1.09%. The results demonstrated that the proliferation activity varied according to the histological pattern of ameloblastoma. The prognosis with different proliferation activity was also varied accordingly.

  • Article: [Mode of new bone formation following distraction osteogenesis in mandibular lengthening].

    ABSTRACT: To investigate the mode and influential factor of new bone formation following distraction osteogenesis in mandibular lengthening. Corticotomy was performed on bilateral mandibles in twelve adult male goats. A custom-made distractor was used to lengthen the mandible at a rate of 1 mm/day for 10 days (total 10 mm elongation). Four goats were sampled respectively at 2, 4 and 8 weeks after completion of distraction. The lengthening mandibles were examined by roentgenography and histology. Newly formed callus was observed in the distraction gap after mandibular lengthening. The new bone exhibited intramembranous ossification generally, but cartilage islands could be found in the specimen that distractor loosed. The above findings indicate that the mode of new bone formation in mandibular lengthening following distraction osteogenesis appears to be intramembranous ossification and that endochondral ossification takes place in case distractor has loosened.

  • Article: Radiographic analysis of a calcifying odontogenic cyst

    ABSTRACT: The Radiographic appearance of 33 cases of calcifying odontogenic cyst (COC) from 1971 to 1998 were analyzed. COC has a variety of radiographic appearances. According to the WHO classification1), such cysts can be divided into five subtypes in order to be considered as central COC and one peripheral type. The subtypes are as follows: I: Unilocular, II: COC associated with odontoma, III: alveolar bone type, IV: compound type, V: recurrences that have become malignant. Ghost cells are an important feature in the diagnosis of COC. However, ghost cells may occur in other tumors as well. Differentiation of the types of COC according to radiographic appearance will aid the clinical diagnosis and treatment of odontogenic cysts.
