  • Article: Neural differentiation ability of mesenchymal stromal cells from bone marrow and adipose tissue: a comparative study.

    ABSTRACT: Background aims. The characteristics, such as morphologic and phenotypic characteristics and neural transdifferentiation ability, of mesenchymal stromal cells (MSC) derived from different origins have yet to be reported for cases isolated from the same individual. Methods. The proliferation capacity, secretion ability of neurotrophins (NT) and neural differentiation ability in rat MSC isolated from bone marrow (BMSC) and adipose tissue (ADSC) were compared from the same animal. Results. The ADSC had a significantly higher proliferation capacity than BMSC according to cell cycle and cumulative population doubling analyses. The proportion of cells expressing neural markers was greater in differentiated ADSC than in differentiated BMSC. Furthermore, the single neurosphere derived from ADSC showed stronger expansion and differentiation abilities than that derived from BMSC. The findings from Western blot lent further support to the immunocytochemical data. The mRNA and protein levels of nerve growth factor (NGF) and brain-derived growth factor (BDNF) expressed in ADSC were significantly higher than those in BMSC at different stages before and following induction. Conclusions. Our study suggests that the proliferation ability of ADSC is superior to that of BMSC. Furthermore, differentiated ADSC expressed higher percentages of neural markers. As one possible alternative source of BMSC, ADSC may have wide potential for treating central nervous system (CNS) diseases.

  • Article: Schwann-like cells can be induction from human nestin-positive amniotic fluid mesenchymal stem cells.

    ABSTRACT: We examined the morphological, phenotypic, and functional characteristics of human amniotic fluid mesenchymal stem cells (AF-MSCs) differentiated towards a Schwann cell lineage. Initially, we induced human AF-MSCs into nestin-positive AF-MSCs. And then, these nestin-positive AF-MSCs were induced into floating neurospheres. After that, neurospheres were induced to differentiate into Schwann-like cells using glia growth factors. In comparison with AF-MSCs, nestin-positive AF-MSCs significantly increased the ratio of neurosphere formation and the percentage of nestin expression in the neurosphere. Differentiated AF-MSCs showed morphological changes similar to those found in Schwann cells. Expression of the Schwann cell markers was determined by immunocytochemical staining and western blotting. Furthermore, differentiated AF-MSCs could promote neurite outgrowth in co-culture with dorsal root ganglia neurons. These results suggest that conversion of human nestin-positive AF-MSCs into cells with Schwann-like cell characteristics is possible and that these cells may have the potential for future cellular therapy for peripheral neurological disorders.

  • Article: Differentiation of cryopreserved human umbilical cord blood-derived stromal cells into cells with an oligodendrocyte phenotype.

    ABSTRACT: In this study, we examined the phenotypic characteristics of human umbilical cord blood-derived mesenchymal stromal cells (UCB-derived MSCs) differentiated along an oligodendrocyte pathway. We induced human UCB-derived MSCs to form floating neurospheres, and these neurospheres were then induced to differentiate into oligodendrocyte progenitor-like cells using multiple induction factors. Differentiated UCB-derived MSCs showed morphologic characteristics of an oligodendrocyte phenotype. The expression of cell surface markers characteristic of oligodendrocyte progenitor cells or oligodendrocytes was determined by immunocytochemical staining. These results suggest that human UCB-derived MSCs can be induced to differentiate into cells with an oligodendrocyte phenotype and that these cells may have potential in the future cellular therapy of central neurological disorders.

  • Article: [Raman and PL spectra studies of natural and HPHT synthetic diamonds].

    ABSTRACT: Investigations of crystal habit, quality and defects analysis of natural diamond and HPHT synthetic diamonds from Fe-C(H) and Ni-C system by Raman and PL spectra indicate that most of the diamonds from Fe-C(H) system have an octahedral habit which is the main habit in natural diamond, while most of the diamonds from Ni-C system have a hexakisoctahedral habit. Because there are more photoluminescence centers in synthetic diamond from Ni-C system than that from Fe-C(H) system, and in synthetic diamond than in natural diamond, the quality of natural diamond is best though it forms in more polygenic environment, which suggested that during the process of diamond formation clearing "impurity" must occur. Therefore, the authors believe that not only the substance system variance but also time and space conditions for diamond formation should be considered when the authors try to probe into the relations of formation process between natural and HPHT synthetic diamonds.

  • Article: In vivo magnetic resonance tracking of Feridex-labeled bone marrow-derived neural stem cells after autologous transplantation in rhesus monkey.

    ABSTRACT: Bone marrow stroma cells-derived neural stem cells (BMSCs-D-NSCs) transplantation is a promising strategy for the treatment of nervous system disorders. The development of a non-invasive method to follow the fate of BMSCs-D-NSCs in vivo is very important for the future application of this treatment. In this paper, we show for the first time, that BMSCs-D-NSCs from rhesus monkeys can be labeled in vitro with the superparamagnetic iron oxide (SPIO) contrast agent Feridex and Poly-L-lysine (PLL) without affecting morphology, cell cycle, telomerase activity, proliferation and differentiation ability of the labeled cells. Furthermore, when autografted into the striatum, these cells survived, differentiated and were incorporated into the brain, and could be reliably tracked using MRI, as confirmed by histological examination of the grafting sites with PKH(67) fluorescence. These results suggest that Feridex labeling of BMSCs-D-NSCs is feasible, efficient and safe for MRI tracing following autografting into the rhesus monkey nervous system.

  • Article: [Distribution of nitrogen and hydrogen in diamond and its significance to nucleation and growth of diamond].

    ABSTRACT: From the center to the rim of natural diamond there are differences in nitrogen and C-H bond contents, suggesting that diamond was formed under varying substance conditions. Both the nitrogen and C-H bond contents tend to decrease from the center to the rim as a whole, indicaing that nitrogen and hydrogen are expended little by little during the formation of diamond. But in the middle area of the sample both the nitrogen and C-H bond contents have a tendency to increase. This implies that nitrogen and hydrogen are added into the environment of diamond formation, meanwhile nitrogen is added earlier. So the procedure of diamond formation may be divided into three stages, namely nucleation and growth in the early stage, growth in the middle stage and in the late stage. In the early and late stages nitrogen and hydrogen are consumed in the environment of diamond formation. In the middle stage nitrogen and hydrogen must be replenished for the growth of diamond, and nitrogen should be replenished earlier than hydrogen. Hydrogen is useful to the nucleation and growth of diamond. During the formation of diamond the compounds of nitrogen and hydrogen do not exist. So we must avoid the formation of compounds of nitrogen and hydrogen for the nucleation and growth of diamond if we plan to introduce hydrogen to the synthesis of diamond at high temperature and high pressure (HPHT). Implanted hydrogen in graphite for the HPHT synthesis of diamond is a good choice.

  • Article: [Sinerem labeling and MRI tracking of neural stem cells in vivo and in vitro].

    ABSTRACT: To label rat neural stem cells (NSCs) with the complex of Sinerem, the ultrasmall superparamagnetic iron oxide (USPIO), and poly-L-lysine (PLL), and evaluate the feasibility of tracking the labeled cells with magnetic resonance imaging (MRI) in vitro and in vivo. Sinerem was incubated with PLL to obtain the complex of Sinerem-PLL. The mesenchymal stem cells (MSCs) isolated from the bone marrow of SD rats were cultured and induced to differentiate into the neural stem cells. The second-passage cells were cultured overnight with the Sinerem-PLL complex, after which Prussian blue staining and transmission electron microscopy were performed to observe the nanoparticles in the cytoplasm. Cell apoptosis assay was performed to assess the cell viability 1 day, 1 week, and 2 weeks after the labeling. Cell tracking with 4.7 MR system was carried out in vivo and in vitro using T(2)WI and T(2)*WI sequences. The NSCs could be effectively labeled with Sinerem-PLL complex with the labeling efficiency exceeding 95%. Prussian blue staining showed numerous blue iron particles in the cytoplasm, and under transmission electron microscope, these particles accumulated in the endosomes/lysosomes. The labeling did not significantly affect the cell viability and proliferation. Remarkable low signal density changes of the labeled cells was seen on T(2)WI and T(2)*WI in vivo and in vitro. NSCs can be effectively labeled with Sinerem-PLL complex, and MRI can be used to track the labeled cells in vivo and in vitro.

  • Article: [Study on heavy metals in soils contaminated by acid mine drainage from Dabaoshan mine, Guangdong].

    ABSTRACT: Mining activities in the Dabaoshan area in the upper reach of the Hengshihe River have caused severe environmental changes, the waste water of milling and refining drained directly into the Hengshihe River, which contaminated the soils along the river severely. It is shown that Pb, Zn, Cd and Cu have contaminated the soil, the Cd contamination was more severe, and the contaminated level of Pb, Zn reached moderately to strongly polluted. The pH value of river and soil affected directly the heavy metals concentration of total and exchangeable ions, and presented negative pertinences. The levels of Pb, Zn, Cu and Cd in the surface soil of Shangbacun village in the lower reach of the river were found as high as 257.762, 350.235, 5.083 and 186.901 mg x kg(-1) respectively, which were relatively higher than those of the background values of soil 1.03, 1.75, 16.9 and 3.7 times respectively, and the result on the soil profiles showed that the contaminations have infiltrated into lower layer soil, ecological environment was harmed severely.

  • Article: [Three-dimensional structural reconstruction of the neurons and astrocytes in rat hippocampal CA3 area].

    ABSTRACT: To observe the distribution of the pyramidal neurons and the surrounding astrocytes in rat hippocampal CA3 area and reconstruct their three-dimension structure. Whole-cell patch-clamp recording of the brain slices, intracellular staining with Lucifer yellow, glial fibrillary acidic protein (GFAP) immunofluorescence, and confocal laser scanning microscopy were utilized in this study. According to the firing patterns, the hippocampal pyramidal neurons were classified into phasic or non-phasic firing neurons. Monolayer scanning and three-dimensional reconstruction showed that many astrocytes aggregated in tight clusters around the pyramidal neurons positive for Lucifer yellow staining, forming close contact. The two types of neurons contacted the astrocytes in different manners. Surrounding the cell body and dendrites of the non-phasic firing neurons, many astrocytes were seen to form contact with the neurons, whereas the phasic firing neurons formed contact with the astrocytes through the dendrites. The spatial distribution of the astrocytes surrounding different types of the hippocampal neurons may differ.

  • Article: [Responses of neurons and astrocytes in rat hippocampus to kainic acid-induced seizures].

    ABSTRACT: To investigate the time course of the responses of neurons and astrocytes in rat hippocampus (HI) to kainic acid (KA)-induced seizures in various regions. By means immunohistochemical staining for anti-Fos protein and anti-glial fibrillary acidic protein (GFAP), the regional distribution of reactive neurons and astrocytes in the HI was observed at different time points after a unilateral stereotaxic microinjection of KA into the lateral ventricle of rats to cause limbic and generalized convulsive seizures. The injection of KA triggered limbic motor seizures including immobilization, staring, facial and jaw clonus ect. followed by recurrent generalized convulsive seizures. After KA-induced seizures, the GFAP-positive astrocytes and Fos-positive neurons were markedly increased in the HI. The increase of GFAP immunoreactivity was observed 30 min after the seizure onset, reaching the maximum at 1 h; the increase of Fos immunoreactivity was detected at 1 h after the onset, peaking at 2 h. The neurons and astrocytes in rat HI are highly active during seizures and the reactive astrocytes might play an important role in epileptogenesis.
