ABSTRACT: New synthetic pathways for novel C(3)-symmetric molecules featured with a thiophene-fused six-five-five carbon-bridged coplanar core structure have been established. Article: Facile synthesis of 5,10,15-hexaaryl truxenes: structure and properties.

ABSTRACT: A new synthesis of 5,10,15-hexaaryltruxene derivatives has been developed. Experimental observations and theoretical calculations confirmed that the presence of aryl substituents on the sp(3)-hybridized bridge carbon atoms had an effect on the photophysical properties of the truxene core. Interestingly, the cage compound 11 possessing a distorted truxene core was verified by X-ray diffraction analysis; the influence of the peripheral aryl substituents on the photophysical properties of the truxene core was diminished because of its molecular rigidity.Organic Letters 03/2011; 13(7):1714-7. · 5.86 Impact Factor -
Article: Near Total Regression of Diffuse Brain Metastases in Adenocarcinoma of the Lung with an EGFR Exon 19 Mutation: A Case Report and Review of the Literature.

ABSTRACT: We report the case of a woman with diffuse brain metastases from lung cancer who experienced total regression of the metastases under gefitinib treatment. The 58-year-old woman was referred to our hospital with a complaint of severe headache. A brain MRI revealed diffuse metastatic lesions in the cerebra and cerebellum. Adenocarcinoma of the lung with multiple brain metastases was diagnosed. The tumor was positive for an epidermal growth factor receptor (EGFR) exon 19 deletion mutation. She was treated with gefitinib 250 mg per day. One year later, the diffuse brain metastases had totally resolved. EGFR-tyrosine kinase inhibitor therapy could be a first-line treatment for patients with advanced adenocarcinoma of the lung with EGFR mutation, especially in those with brain metastases.Case Reports in Oncology 01/2011; 4(3):445-51. -

Article: Osteopontin expression is a valuable marker for prediction of short-term recurrence in WHO grade I benign meningiomas.

ABSTRACT: Prediction of recurrence remains a challenge in histopathological benign/grade I tumors. Osteopontin (OPN) plays important roles in tumorigenesis, invasion, and metastasis of several human cancers. In this study, we investigated OPN protein expression by evaluating the differences between recurrent and non-recurrent histologically benign meningiomas. Thirty-two patients were enrolled, and 23 benign non-recurrent meningiomas and 9 benign recurrent meningiomas were followed for a mean of 34 months after complete surgical resection (Simpson grades I and II). Cytoplasmic OPN staining was evaluated by means of immunohistochemistry (IHC) score and by use of the Allred-8-unit system. We examined clinical biological data, their relationship with tumor recurrence, and the expression of OPN. Our results showed that meningioma recurrence correlated significantly with OPN IHC score (P = 0.001). An OPN Allred score between 0 and 3 was associated with a recurrence-free time of more than 25 months. In comparison, an OPN Allred score from 4 to 8 was indicative of a shorter average recurrence-free time. We concluded that OPN IHC score may play a role in prediction of the recurrence of the grade I meningiomas. Article: Cortactin, fascin and survivin expression associated with clinicopathological parameters in brain gliosarcoma.

ABSTRACT: Gliosarcoma is a very rare primary neoplasm of the central nervous system classified as a variant of glioblastoma. Cortactin, fascin and survivin have been found in several human cancers to play important roles in tumor progression, but the expression pattern of these biomarkers in gliosarcoma is unclear. Immunostaining for cortactin, fascin and survivin was assessed in 6 surgical specimens of brain gliosarcoma, and the relationship between the expression of these biomarkers and tumor size or clinical parameters were examined. Five of our six patients with gliosarcoma survived 3-17 months. One patient is still alive for more than 24 months. The mean immunostaining scores for cortactin were significantly higher in the gliomatous (score 236.6 +/- 45.4) and sarcomatous (score 233.3 +/- 51.4) components than in normal brain tissues (score 21.6 +/- 6.6). The mean cytoplasmic immunostaining scores for fascin and survivin were also significantly higher in the gliomatous and sarcomatous components than in normal brain tissues. In addition, survivin was also stained in the nucleus of tumor cells. Linear regression analysis showed that fascin score in the gliomatous component was significantly associated with tumor size (R = 0.69) and the fascin score in the sarcomatous component was significantly associated with patient's age (R = 0.87). In addition, the survivin cytoplasmic scores in the gliomatous and sarcomatous components were inversely associated with tumor size. Our results demonstrated that over-expression of cortactin, fascin and survivin is associated with malignant transformation of brain gliosarcoma. Article: Carcinosarcoma of the pleura and lung metastasized from a poorly differentiated squamous cell carcinoma of the esophagus.

ABSTRACT: Transformation of a poorly differentiated squamous cell carcinoma (SCC) of the esophagus into a âcarcinosarcomaâ of the pleura and lung has never been reported and its histogenetic origin is still debated. A 48-year-old man was admitted due to progressive dysphagia and a weight loss of 5 kilograms within 2 months. Upper gastrointestinal panendoscopic biopsy revealed poorly differentiated SCC of thoracic esophagus, upper third, T4N1M1a, stage IVa. He received concurrent chemoradiotherapy (CCRT). About 9 months later, rapid progression of lung metastases and pleural effusion were found. According to the histopathological and immunohistochemical stain results, carcinosarcoma was diagnosed. Palliative therapy was given and the patient eventually died of the disease 9 months after SCC of the thoracic esophagus was diagnosed and one month after carcinosarcoma of the pleura and lung were confirmed.Hematology/ Oncology and Stem Cell Therapy 01/2010; 3(1):47-50. -
Article: Retroperitoneal metastatic adenocarcinoma complicated with necrotizing fasciitis of the thigh in a patient with advanced rectal colon cancer.

ABSTRACT: Necrotizing fasciitis of the thigh due to colon cancer has not been previously reported, especially during radiotherapy. A 73-year-old woman admitted to our hospital was diagnosed with sigmoid colon cancer that had spread to the left psoas muscle; radiotherapy was performed. Three months after the initiation of radiotherapy, the patient developed gait disturbance, poor appetite and high fever and was therefore admitted to the emergency department of our hospital. Blood examination revealed generalized inflammation with a high white blood cell count and C-reactive protein level. Computed tomography of the abdomen revealed fluid and gas tracking from the retroperitoneum into the intramuscular plane of the grossly enlarged right thigh. Consequently, emergent debridement was not performed and conservative therapy was done. The patient died. Necrotizing fasciitis of the thigh due to the spread of rectal colon cancer is unusual, but this fatal complication should be considered during radiotherapy in patients with unresectable colorectal cancer.Case Reports in Oncology 01/2010; 3(2):304-9. -
Article: Acute myelogenous leukemia and acute leukemic appendicitis: a case report.

ABSTRACT: Acute myelogenous leukemia (AML) can involve the gastrointestinal tract but rarely involves the appendix. We report a male patient who had 1 year partial remission from AML and who presented with apparent acute appendicitis as the initial manifestation of leukemia relapse. Pathological findings of the appendix revealed transmural infiltrates of myeloblasts, which indicated a diagnosis of leukemia. Unfortunately, the patient died from progression of the disease on the 19th d after admission. Article: Gemcitabine plus vinorelbine as an effective salvage chemotherapeutic regimen in cisplatin-refractory germ cell tumor.

ABSTRACT: Germ cell tumors (GCT) are the most common malignancies among male adolescents. Approximately 5% of GCTs are of extragonadal origin. Cisplatin-based chemotherapy is the standard first-line treatment for GCT. Patients who fail to respond to first-line treatment usually have poor outcomes. High-dose chemotherapy with stem cell support, paclitaxel, gemcitabine, and oxaliplatin are reported as effective salvage treatment. We report two patients with cisplatin-refractory, metastatic, nonseminomatous, and extragonadal GCT treated successfully with gemcitabine plus vinorelbine (GV). One patient achieved good partial remission and had a five-month progression-free period. Another patient is still alive with stable disease after 4 cycles of treatment. Article: Radiofrequency ablation as a palliative treatment of primary liver sarcoma in an inoperable patient with hepatitis C virus-related cirrhosis.

ABSTRACT: Radiofrequency ablation (RFA) can be performed percutaneously, by laparotomy, or by laparoscopy. Recent advances in laparoscopic ultrasound (LUS) have improved the accuracy in detecting intrahepatic nodules. Primary liver sarcomas in chronic hepatitis C virus (HCV) infection with or without cirrhosis are extremely rare and the prognosis is poor. A 58-year-old man with a 5-year history of HCV-related Child-Pugh B cirrhosis was admitted with abdominal fullness, weight loss and lower-extremity edema lasting for 1 month. One mass lesion was measured as 4 cm in the segments 2 and 3 of the liver by abdominal computed tomography. As the patient's condition did not permit operation, he received LUS-guided RFA. After the treatment, the survival of this patient exceeded 48 months. Treatment of primary liver sarcomas arising in HCV infection with or without cirrhosis is primarily limited to surgery, chemotherapy and radiotherapy. Liver transplantation is not suggested for either primary or metastatic liver sarcoma. The described inoperable case is the only case report about a successful RFA treatment of a solitary liver sarcoma. We suggest that RFA can be considered as a choice of palliative treatment for primary liver sarcoma in inoperable patients with HCV-related cirrhosis.Onkologie 02/2009; 32(8-9):509-12. · 0.87 Impact Factor -

Article: Convergent/divergent synthesis and photophysical studies of alternating benzene-furan oligoaryls from substituted propargylic dithioacetals.

ABSTRACT: A range of oligoaryls that contain alternating benzene-furan rings is synthesized by a rapid convergent/divergent method from the annulation of a propargylic dithioacetal and an aldehyde with a propargylic dithioacetal moiety as a substituent. These oligomers are fairly soluble in a range of organic solvents and can be easily purified by reprecipitation. The substituents on the furan rings can be varied according to the substituents in the starting propargylic dithioacetals. This route provides a useful procedure for the synthesis of alternating benzene-furan oligomers without repeated units. These furan-containing oligoaryls are highly fluorescent in the visible region and are electrochemically active.
