  • Article: Association between maternal exposure to perfluorooctanoic acid (PFOA) from electronic waste recycling and neonatal health outcomes.

    ABSTRACT: Perfluorooctanoic acid (PFOA) has applications in numerous industrial and consumer products. The widespread prevalence of PFOA in humans demonstrated in recent studies has drawn considerable interest from the public. We aimed to evaluate the exposure of mothers to PFOA and the potential hazards to neonates in a primitive electronic waste recycling area, Guiyu, China, and a control area, Chaonan, China. Our investigation included analyses of maternal serum samples, health effect examinations, and other relevant factors. Questionnaires were administered and maternal serum samples were collected for 167 pregnant women. Solid phase extraction method was used for all analytical sample preparation, and analyses were completed using high performance liquid chromatography tandem mass spectrometry method. The PFOA concentration was higher in maternal serum samples from Guiyu than in samples from Chaonan (median 16.95, range 5.5-58.5ngmL(-1); vs. 8.7, range 4.4-30.0ngmL(-1); P<0.001). Residence in Guiyu, involvement in e-waste recycling, husband's involvement in e-waste and use of the family residence as workshop were significant factors contributing to PFOA exposure. Maternal PFOA concentrations were significantly different between normal births and adverse birth outcomes including premature delivery, term low birth weight, and stillbirths. After adjusting for potential confounders, PFOA was negatively associated with gestational age [per lg-unit: β=-15.99days, 95% confidence interval (CI), -27.72 to -4.25], birth weight (per lg-unit: β=-267.3g, 95% CI, -573.27 to -37.18), birth length (per lg-unit: β=-1.91cm, 95% CI, -3.31 to -0.52), and Apgar scores (per lg-unit: β=-1.37, 95% CI, -2.42 to -0.32), but not associated with ponderal index. Mothers from Guiyu were exposed to higher levels of PFOA than those from control areas. Prenatal exposure to PFOA was associated with decreased neonatal physical development and adverse birth outcomes.

  • Article: Carcinogenic polycyclic aromatic hydrocarbons in umbilical cord blood of human neonates from Guiyu, China.

    ABSTRACT: Unregulated electronic-waste recycling results in serious environmental pollution of polycyclic aromatic hydrocarbons (PAHs) in Guiyu, China. We evaluated the body burden of seven carcinogenic PAHs and potential health risks for neonates. Umbilical cord blood (UCB) samples were collected from Guiyu (n=103), and the control area of Chaonan (n=80), China. PAHs in UCB were determined by gas chromatography/mass spectrometry. The median ∑7c-PAH concentration was 108.05 ppb in UCB samples from Guiyu, vs. 79.36 ppb in samples from Chaonan. Residence in Guiyu and longer cooking time of food during the gestation period were significant factors contributing to the ∑7c-PAH level. Benzo[a]anthracene (BaA), chrysene (Chr), and benzo[a]pyrene (BaP) were found to correlate with reduced neonatal height and gestational age. Infants experiencing adverse birth outcomes, on the whole, displayed higher BaA, Chr, and BaP levels compared to those with normal outcomes. We conclude that maternal PAH exposure results in fetal accumulation of toxic PAHs, and that such prenatal exposure correlates with adverse effects on neonatal health.

    Article: Sensitized chemiluminescence reaction between hydrogen peroxide and periodate of different types of Mn-doped ZnS quantum dots

    ABSTRACT: In this work, different types of Mn-doped ZnS QDs were prepared and applied to study of the chemiluminescence reaction of hydrogen peroxide and periodate. The effects of the size of nanoparticles, shell protection and the stabilizing agents on chemiluminescence intensity were studied. The results indicated that the size of quantum dots was not the only factor to enhance the intensity of chemiluminescence system. Different stabilizing agents and the silica film protection will also influence the performance of the quantum dots in the chemiluminescence reaction. A CL spectrum was examined by using a series of high-energy cut-off optical filters and a possible mechanism of the CL was also proposed.   • Article: Birth outcomes related to informal e-waste recycling in Guiyu, China.

    ABSTRACT: To assess the impact of exposure to informal e-waste recycling on birth outcomes. We compared record-based birth outcomes (n=24,493) and levels of cord blood lead (CBPb) (n=531) in Guiyu and a control area in Xiamen. Guiyu births showed significantly higher rates of adverse birth outcomes including stillbirth (4.72% vs. 1.03%), low birth weight (6.12% vs. 4.12%), term low birth weight (3.40% vs. 1.57%), and lower Apgar scores (9.6 vs. 9.9) and mean birth weight (3168 g vs. 3258 g) than did births from the control site, all P<0.01. Regression analysis showed Guiyu had about four times higher risk of stillbirth compared with Xiamen. The median level of CBPb was much higher in neonates of Guiyu than those in the control group (10.78 μg/dL vs. 2.25 μg/dL). Prenatal exposure to informal e-waste recycling related to high rate of adverse birth outcomes, lower Apgar scores and unsafe lead level in cord blood.

  • Article: In utero exposure to polychlorinated biphenyls and reduced neonatal physiological development from Guiyu, China.

    ABSTRACT: Primitive electronic-waste recycling resulted in serious pollution of polychlorinated biphenyls (PCBs) in the environment of Guiyu, China. We aimed to assess body burdens of PCBs and potential health risks for neonates. Umbilical cord blood (UCB) samples were collected from Guiyu, and the control area of Chaonan, China. PCBs in UCB were determined by gas chromatography coupled with mass spectrometry. The median ∑PCBs concentration was 338.56 ng g⁻¹ lipid in UCB samples from Guiyu, vs. 140.16 ng g⁻¹ lipid in samples from Chaonan. Mothers' involvement in electronic-waste recycling activities and house used as family workshop were the significant factors that contributed to ∑PCBs level. Some individual PCB congeners were found to correlate with reduced neonatal height, weight, Apgar scores, gestational age, and body mass index (BMI). Difference of ∑PCBs level was found between normal birth and adverse birth outcomes. Neonates from Guiyu are exposed to high levels of PCBs; prenatal exposure to PCBs appears to influence neonatal health, but still needs further confirmation.

  • Article: Association between lead exposure from electronic waste recycling and child temperament alterations.

    ABSTRACT: We aimed to evaluate the dose-dependent effects of lead exposure on temperament alterations in children from a primitive e-waste (obsolete electrical and electronic devices) recycling area in Guiyu of China and a control area (Chendian, China). Blood lead levels (BLL) might be correlated with temperament, health, and relevant factors that were evaluated through Parent Temperament Questionnaire (PTQ), physical examination, and residential questionnaires. We collected venipuncture blood samples from 303 children (aged 3-7 years old) between January and February 2008. Child BLL were higher in Guiyu than in Chendian (median 13.2 μg/dL, range 4.0-48.5 μg/dL vs. 8.2 μg/dL, 0-21.3 μg/dL) (P<0.01). Significant differences of mean scores in activity level (4.53±0.83 vs. 4.18±0.81), approach-withdrawal (4.62±0.85 vs. 4.31±0.89), and adaptability (4.96±0.73 vs. 4.67±0.83) were found between Guiyu and Chendian children (all P<0.01). High BLL (BLL≥10μg/dL) child had higher mean scores of approach-withdrawal when compared with those children with low BLL (BLL<10 μg/dL) (4.61±0.87 vs. 4.30±0.88, P<0.01). Location of child residence in Guiyu, and parents engagement in work related to e-waste were the risk factors related to child BLL, activity level, approach-withdrawal, adaptability, and mood. Child hand washing prior to food consumption was a protected factor for BLL and several dimensions. There are close relationships between BLL elevation, temperament alteration and the e-waste recycling activities in Guiyu. Primitive e-waste recycling may threaten the health of children by increasing BLL and altering children temperament, although the exposure to other toxicants needs to be examined in future studies.

  • Article: Assessment of cadmium exposure for neonates in Guiyu, an electronic waste pollution site of China.

    ABSTRACT: This study aimed to determine the levels of placental cadmium (PCd) and cord blood cadmium (CBCd) and the resulting expression of placental metallothionein (MT) in neonates and to investigate cadmium (Cd) exposure levels in neonates and mothers who live in Guiyu, China, an electronic waste (e-waste) pollution site. Among the 423 mothers included in the study from 2004/2005 to 2007, 289 lived in Guiyu (exposed group) and 134 lived in Chaonan, located 10 km away from Guiyu (controls) and had never been exposed to e-waste pollution. CBCd and PCd levels were measured by atomic absorption spectrometry. Placental MT was examined by immunohistochemistry. Information on maternal and neonatal characteristics and exposure conditions was obtained from hospital records and by personal interviews. For the 3 years, the median CBCd was higher for Guiyu neonates than for controls (3.61 vs. 1.25 μg/L), with 25.61% of Guiyu subjects exhibiting a median CBCd that exceeded the safety limit defined by the World Health Organization (5 μg/L), as compared with 14.18% of control neonates (p < 0.01). In Guiyu, the mean PCd was higher than that for controls (0.17 ± 0.48 vs. 0.10 ± 0.11 μg/g, p ≤ 0.01). The high levels of CBCd and PCd were significantly associated with parents' occupational and environmental exposure to e-waste recycling pollutants. Staining for MT was positive and dense for 67.00% (67/100) of Guiyu neonates as compared with 32.69% (17/52) of controls (p < 0.01). Exposure to e-waste recycling pollutants increased Cd exposure in neonates, which was accompanied by increased placental MT expression.

    ABSTRACT: In this paper, Mn-doped ZnS quantum dots (QDs) capped by a molecularly imprinted polymer (MIP) were synthesized. The results showed a high selectivity of the MIP-capped Mn-doped ZnS QDs toward the template molecule (4-nitrophenol) by QD fluorescence quenching. The application of MIP-capped Mn-doped ZnS QDs to the chemiluminescence (CL) system was also studied using a KIO(4)-H(2)O(2) system. This application combines the good selectivity of MIP with the high sensitivity of CL. The linear range of this CL system is from 0.1 to 40 microM, and the detection limit (DL) for 4-nitrophenol in the water can reach 76 nM. The method was also used in the real water samples, and the recoveries can fall in the range of 91-96%.

  • Article: Effect of simultaneous silencing of HPV-18 E6 and E7 on inducing apoptosis in HeLa cells.

    ABSTRACT: The objective of this investigation was to determine if simultaneous silencing of the human papillomavirus type 18 (HPV-18) E6 and E7 oncogenes using RNA interference (RNAi) would be a potential therapeutic approach against the carcinogenic activity of this virus. Two synthetic double-stranded oligonucleotides, encoding short hairpin transcripts corresponding to HPV-18 E6 and E7 genes, were cloned into pGenesilence (pGS) 1.0 vectors to produce pGS-E6, pGS-E7, and pGS-(E6+E7), respectively. Our results showed that the expression of HPV-18 E6 class 1 and HPV-18 E7 in HeLa cells was markedly decreased after being transfected with pGS-E6, pGS-E7, and pGS-(E6+E7) vectors. Of the three vectors, pGS-(E6+E7) had a greater ability to decrease the growth rate of HeLa cells, inhibit colony formation in soft agar, and significantly reduce tumor growth in nude mice. We also found that depletion of HPV-18 E6 and E7 in this manner promoted apoptosis of HeLa cells. Our data showed that simultaneously decreasing HPV-18 E6 and E7 gene expression in HeLa cells by RNAi could significantly inhibit tumor growth under in vitro conditions and in nude mice. These data suggest that gene therapy may be a possible therapeutic approach for HPV-positive cervical cancers.

Deux oligonucléotides synthétiques double brin codant les courts transcrits en épingle correspondant aux gènes E6 et E7 du VPH-18 ont été clonés dans des vecteurs pGenesilence (pGS) 1.0 pour produire pGS-E6, pGS-E7 et pGS-(E6+E7), respectivement. Nos résultats ont montré que l'expression de E6 VPH-18 de classe 1 et de E7 VPH-18 chez les cellules HeLa était diminuée de façon marquée à la suite d'une transfection avec les vecteurs pGS-E6, pGS-E7 et pGS-(E6+E7). Des trois vecteurs, pGS-(E6+E7) possédait la meilleure capacité à diminuer le taux de croissance des cellules HeLa, à inhiber la formation de colonies en agar mou et à réduire significativement la croissance tumorale chez les souris nues. Nous avons aussi trouvé que la déplétion de E6 et E7 du VPH-18 à l'aide de cette approche favorisait l'apoptose des cellules HeLa. Nos résultats montrent que la diminution simultanée de l'expression de E6 et E7 du VPH-18 chez les cellules HeLa par ARNi pouvait inhiber significativement la croissance tumorale in vitro et chez les souris nues. Ces résultats suggèrent que la thérapie génique pourrait constituer une approche thérapeutique des cancers du col positifs au VPH.

  • Article: Monitoring of lead, cadmium, chromium and nickel in placenta from an e-waste recycling town in China.

    ABSTRACT: Toxic heavy metals are released to the environment constantly from unregulated electronic waste (e-waste) recycling in Guiyu, China, and thus may contribute to the elevation of lead and other heavy metals levels in placenta. We aimed to investigate concentrations of heavy metals, including lead (Pb), cadmium (Cd), chromium (Cr), and nickel (Ni) in placenta from Guiyu and compared them with those from a control area where no e-waste processing occurs. Two hundred and twenty human placentas were collected from Guiyu (n=101) and the control area (n=119). The placenta concentrations of Pb, Cd, Cr, and Ni (PCPb, PCCd, PCCr, and PCNi) were determined by graphite furnace atomic absorption spectrometry (GFAAS). Risk factors of high exposure and correlation with adverse pregnancy outcomes were analyzed using Spearman correlation analyses. PCPb from Guiyu ranged from 6.51 to 3465.16ng/g with a median of 301.43ng/g, whereas PCPb from the control area ranged from 4.53 to 3176.12ng/g with a median of 165.82ng/g (P=0.010). We also observed that in Guiyu, 41.6% of women (42/101) had PCPb>500ng/gwt (wet weight), compared with 24.4% of women (29/119) in the control area (P=0.006). No significant differences of PCCd and PCCr were found between the two groups. In contrast, PCNi was higher in samples from the control area (median 14.30, range 1.76-593.70ng/g) than in Guiyu samples (median 7.64, range 1.19-1108.99ng/g) (P=0.000), and a negative correlation between PCNi and gestational age was found in this study (P=0.017). Spearman correlation analyses showed that there was correlation between PCPb and residence in e-waste recycling area. Environmental pollution, resulted from unregulated e-waste recycling activities, may contribute to elevated PCPb in neonates born in Guiyu and threaten their health.

  • Article: Polybrominated diphenyl ethers in umbilical cord blood and relevant factors in neonates from Guiyu, China.

    ABSTRACT: We aimed to evaluate the exposure of neonates to polybrominated diphenyl ethers (PBDEs) from a primitive e-waste (obsolete electrical and electronic devices) recycling area, in Guiyu, China, and a control area, Chaonan, China, through umbilical cord blood (UCB), the health effects, and relevant factors. Questionnaires were addressed, and UCB was collected shortly after birth from 153 pregnant women between May and July 2007. Blood samples were prepared by liquid-liquid extracting methods. PBDE concentration was determined by gas chromatography/mass spectrometry in the electron capture negative ionization mode. The total PBDE concentration was higher in UCB samples from Guiyu than in Chaonan samples (median 13.84, range 1.14-504.97 ng g(-1) lipid, vs 5.23, range 0.29-363.70 ng g(-1) lipid) (p < 0.05). BDE-209 was the dominant PBDE congener, followed by BDE-47, -153, and -99. Residence in Guiyu, which is a site for e-waste recycling, involvement in e-waste recycling, and the residence also being used as a family workshop were significant factors contributing to PBDE exposure. PBDE levels significantly differed in neonates by normal birth and adverse birth outcomes including stillbirth, low birth weight, and premature delivery (p < 0.05). The neonates from Guiyu are exposed to high levels of PBDEs. Prenatal exposure to PBDEs may affect neonates' health in Guiyu, which still needs to be evaluated in larger epidemiological studies.

  • Article: Lead affects apoptosis and related gene XIAP and Smac expression in the hippocampus of developing rats.

    ABSTRACT: Lead (Pb) exposure poses devastating effects on central nervous system development of children. To replicate aspects of this neurotoxicity, we examined the effect of lead on the expression of apoptosis and of apoptosis-related genes, XIAP (X chromosome-linked inhibitor of apoptosis protein) and Smac (second mitochondrial activator of caspase), in the hippocampus of developing rats. A total of 48 rats (30-day old) were randomly divided into four groups for intragastrical perfusion of lead acetate [Pb(Ac)2]: untreated, low (2 mg/kg/d), medium (20 mg/kg/d), and high (200 mg/kg/d) dose groups. Pb content was determined in blood, and the apoptosis indexes and XIAP and Smac gene expression were analyzed in the hippocampus. There was a significant difference in apoptosis indexes (AI) between the exposed and control groups (p < 0.01). AI was highest in the high exposure group. XIAP gene expression was reduced in the exposed groups and the expression was negatively correlated with blood lead levels (BLLs) (p < 0.05). But the four groups did not differ in the expression of Smac (p > 0.05). Our data indicate that exposure to Pb(Ac)2 caused a dose-dependent and significant increase of apoptosis in the hippocampus of developing rats through depressing the expression of the XIAP but not the Smac genes.

  • Article: Lead and cadmium synergistically enhance the expression of divalent metal transporter 1 protein in central nervous system of developing rats.

    ABSTRACT: Divalent metal transporter 1 (DMT1) can transport a large range of ions, including toxic lead (Pb) and cadmium (Cd), across membranes. In this study, a total of 24 rats were divided into four groups for intragastrical perfusion treatment: control, Pb alone, Cd alone, and Pb + Cd. Pb and Cd contents in blood were detected, and the mRNA and protein levels of DMT1 were analyzed in the cerebellum, cortex, and hippocampus. Both Pb and Cd levels were elevated in all groups perfused with Pb and/or Cd, except for Pb level in the Cd-alone group (P < 0.05). The mRNA level of DMT1 did not differ among the four groups (P > 0.05). However, the DMT1 protein expression was significantly increased by 0.9-, 1.0-, and 1.1-fold in cerebellum, cortex, and hippocampus of the Pb + Cd group than in controls, respectively. Pb and Cd exposure can synergistically induce DMT1 protein synthesis and has implications for transportation of toxic ions in the developing rat's brain.

  • Article: Monitoring of lead load and its effect on neonatal behavioral neurological assessment scores in Guiyu, an electronic waste recycling town in China.

    ABSTRACT: Guiyu is the major electronic waste (e-waste) recycling town in China. The primary purpose of this study was to measure the lead levels in neonates and examine the correlation between lead levels and neurobehavioral development. One hundred full-term neonates from Guiyu and fifty-two neonates from neighboring towns (control group) in the late summer of 2006 were selected for study. The lead levels in the umbilical cord blood (CBPb) and lead levels in meconium (MPb) of neonates were determined with atomic absorption spectrophotometry. The neonatal behavioral neurological assessment (NBNA) was conducted on all neonates. A questionnaire related to the exposure to lead of pregnant women was used as a survey of the neonates' mothers. Compared with the control group, neonates in Guiyu had significantly higher levels of lead (P < 0.01), and the mean CBPb and MPb were 113.28 microg L(-1) and 2.50 microg g(-1), respectively. The relatively high lead levels in the neonates of the Guiyu group were found to correlate with their maternal occupation in relation to e-waste recycling. Neonates with high levels of lead load have lower NBNA scores (P < 0.01). There was a statistically significant difference in NBNA scores between the Guiyu group and the control group by t test (P < 0.05). No correlation was found between CBPb and NBNA scores; however, a negative correlation was found between MPb and NBNA scores (P < 0.01). There is a correlation between relatively high lead levels in the umbilical cord blood and meconium in neonates and the local e-waste recycling activities related to lead contamination. This study suggests that environmental lead contamination due to e-waste recycling have an impact on neurobehavioral development of neonates in Guiyu.

    Article: The hazard of chromium exposure to neonates in Guiyu of China.

    ABSTRACT: Guiyu is one of the most heavily chromium-polluted areas in China due to the presence of numerous electronic waste (e-waste) recycling sites in the region. In this study, we investigate the effect of umbilical cord blood chromium levels (UCBCLs) on neonates from Guiyu and discuss chromium-induced DNA damage of cord blood lymphocyte. Umbilical cord blood samples were collected from neonates of Guiyu (in 2006, n=100; in 2007, n=100) and the neighboring town of Chaonan (in 2006, n=52; in 2007, n=50) that is associated with the fishery. UCBCLs of the neonates were determined by graphite atomizer absorption spectrophotometer. Comet experiment was used to examine lymphocyte DNA damage. Questionnaires to gauge chromium exposure were administered to the mothers of the neonates. The mean UCBCLs of neonates in the Guiyu group in 2006 and 2007 were 303.38 microg/L and 99.90 microg/L with median 93.89 microg/L and 70.60 microg/L, respectively. We observed significant differences between the results in UCBCLs of neonates in Guiyu and the control group (P<0.01). There was no significant difference of UCBCLs in neonates between 2006 and 2007 in Guiyu (P>0.05). Higher levels of chromium in neonates were found to correlate with their mothers' exposure to e-waste recycling. There were significant differences in terms of DNA damage between the Guiyu group and the control group (P<0.05). There was a correlation between DNA damage and the UCBCLs of neonates (P<0.05). There is conclusive evidence that high UCBCLs in neonates exists in e-waste recycling areas in Guiyu and that e-waste recycling activity poses serious environmental problems. Chromium pollution is threatening the health of neonates around the recycling sites.

    Article: Blood lead and cadmium levels and relevant factors among children from an e-waste recycling town in China.

    ABSTRACT: Primitive electronic waste (e-waste) recycling is ongoing in Guiyu, and thus toxic heavy metals may keep on threatening to the health of local children. Some related factors may contribute to the elevation of blood lead levels (BLLs) or blood cadmium levels (BCLs). To investigate the children's BLLs and BCLs in Guiyu and Chendian as compare to discuss the effects of primitive e-waste recycling activities on children's health. Two hundred and seventy-eight children less than 8 years who lived in Guiyu and Chendian were observed, and their BLLs and BCLs were determined by graphite atomizer absorption spectrophotometer. Questionnaire survey for risk factors was also performed and data were analyzed using spearman correlation analyses and logistic regression analyses. Children living in Guiyu had significantly higher BLLs and BCLs as compared with those living in Chendian (p<0.01). In Guiyu, 70.8% of children (109/154) had BLLs>10 microg/dL, and 20.1% of children (31/154) had BCLs>2 microg/L, compared with 38.7% of children (48/124) had BLLs>10 microg/dL and 7.3% of children (9/124) had BCLs>2 microg/L in Chendian (p<0.01, respectively). We also observed a significant increasing trend in BLLs with increasing age in Guiyu (p<0.01). Mean height of children in Guiyu was significantly lower than that in Chendian (p<0.01). The risk factors related to children's BLLs and BCLs mainly included father's engagement in the work related to e-waste, children's residence in Guiyu and the amount of time that children played outside near the road everyday. There are close relationships between the BLLs, BCLs in children and the primitive e-waste recycling activities in Guiyu. Environmental pollution, especially lead pollution, has threatened the health of children living around e-waste recycling site.
