  • Article: [Study on content of two sesquiterpenes in Alpinia oxyphylla].

    ABSTRACT: To develop an HPLC method for determining oxyphyllenodiol A (1) and teuhetenone A (2) contained in Alpinia oxyphylla and to compare the contents of the two components contained in medicinal materials and prepared herbal medicines in pieces sold in the market and different fractions. HPLC and Waters sunfire C18 column (4. 6 mm x 250 mm, 5 microm) were adopted for gradient elute with the mobile phase of acetonitrile and water. The flow rate was 1.0 mL x min(-1) and the detection wavelength was 250 nm. 1 and 2 showed a good linear relationship within the range of 0.1296 - 0.8640 microg and 0.1635 - 1.0900 microg respectively, with the average recoveries of 99.08% and 97.80%. Their content ranges were 0.0059% - 0.0149% and 0.0080% - 0.0164% in different samples. The mean value of 1 and 2 were 0.0085% and 0.0104% in the whole fruits, and 0.0137% and 0.0157% in the seeds. They were undetected in the nutshells. The method is so precise, accurate and highly reproducible that it can be used to determine the contents of oxyphyllenodiol A and teuhetenone A in A. oxyphylla. The contents of the two components are mainly extracted from the seeds, with certain difference among different samples. There are a higher contents and no significant difference in the salted and raw seeds.

  • Article: Triterpenoid saponins from Dipsacus asper and their activities in vitro.

    ABSTRACT: Two new triterpenoid saponins 1 and 2, along with six known saponins 3-8, were isolated from the roots of Dipsacus asper. The structures of new compounds were established as 3-O-β-d-glucopyranosyl-(1 → 4)-[α-l-rhamnopyranosyl-(1 → 3)]-β-d-glucopyranosyl-(1 → 3)-α-l-rhamnopyranosyl-(1 → 2)-α-l-arabinopyranosyl-hederagenin-28-O-β-d-glucopyranoside (dipsacus saponin J, 1) and 3-O-α-l-arabinopyranosyl-hederagenin-28-O-β-d-glucopyranosyl-(1 → 6)-β-d-glucopyranosyl-(1 → 6)-β-d-glucopyranoside (dipsacus saponin K, 2). The structures were determined by extensive analysis of their spectroscopic data. Compounds 6 and 7 could significantly stimulate UMR106 cell proliferation and increase alkaline phosphatase activities in UMR106 cell at the concentration of 4 μM.

  • Article: [Determination of ergosterol in Ganoderma lucidum from different varieties and cultured tree species by HPLC].

    ABSTRACT: HPLC method for determining ergosterol was established to evaluate the quality of Ganoderma lucidum and to analyze of the ergosterol in different varieties and cultrued tree species. Samples from 17 different varieties and 14 different tree species were quantified by HPLC. The RP-HPLC was conducted on Diamonsil C18 column with acetonitrile as the mobile phase at 40 degrees C. The flow rate was 1.0 mL/min; and the detection wavelength was 282 nm. The fluctuations of ergosterol content was 0.093%-0.243% among different varieties and 0.080%-0.227% among different tree species. Hanzhi-2, Yuanzhi, Ming-1, Yesheng-1 hao were excellent at both ergosterol content and conversion rate among all the varieties, and Bai Li, Yang Mei have comparatively more ergosterol and high conversion rate among all the tree species in this study. This method is simple, sensitive and accurate. It could be used to determine the contents of ergosterol and the evaluation of the quality in Ganoderma. It also provides references for choosing varieties and tree species when culture Ganoderma.

  • Article: Study of the relationship between genetics and geography in determining the quality of Astragali Radix.

    ABSTRACT: Astragali Radix (AR), prepared from the roots of Astragalus membranaceus (FISCH. ex LINK) BUNGE or its variey, A. membranaceus (FISCH. ex LINK) BUNGE var. mongholicus (BUNGE) HSIAO., is one of the most used and valuable traditional Chinese medicines (TCMs). Historically, Hunyuan, Shanxi Province in China is the geo-authentic producing area of AR and crude AR from here called "geo-authentic." According to tradition, geo-authentic TCMs define both authenticity and quality. However, no scientific investigation has ever determined whether the superior quality of Hunyuan AR is due to the genetic characteristics or to the local environment. In our study, seeds of 30 AR samples representing the two varieties from different regions were cultivated in Hunyuan under the same conditions. A method, using ultra-performance liquid chromatography coupled with photodiode array detector and evaporative light scattering detectors, was developed to evaluate the quality through a simultaneous determination of four major isoflavonoids and four major saponins. The two AR varieties were successfully distinguished by principal component analysis while samples of the same species with different seeds origins could not be distinguished. A genetic study demonstrated that the internal transcribed spacer sequences of the nuclear ribosomal DNA in A. membranaceus var. mongholicus samples from different geographical regions were highly conservative. These results indicate that the content of active components in AR depends on the interaction of genotype and environment. At the varietal level, genetic properties appear to be more important for pharmaceutical quality than environmental factors, while on the intraspecific level environmental factors might be more important than genetic properties.

  • Article: [Identifying the characteristics of FTIR spectra of herba epimedii icariin via wavelet analysis and RBF neural network].

    ABSTRACT: In the present paper, the authors extracted active components of herba epimedii and their important features using Fourier transform infrared spectroscopy (FTIR), correlation coefficient comparison, and multilevel wavelet analysis. The extracted features were then used to classify herba epimedii via radial basis function (RBF) neural network. There were altogether 250 samples of the medicine with various different types, including epimedium brevicornu Maxim., E. sagittatum (Sieb. et Zucc. ) Maxim, E. pubescens Maxim., E. koreanum Nakai and E. wushanense T. S. Ying. An important component of herba epimedii, herba epimedii icariin, has a special peak at 1 259 cm(-1) on the FTIR spectra obtained from the methanol extraction, which is consistent with the result obtained by traditional HPLC qualitative analysis. Therefore, this special peak can be used to determine if herba epimedii contains herba epimedii icariin. Furthermore, large variations in the spectrum caused by low content of icariin, weak absorption peaks and noise were successfully removed by applying correlation coefficient comparison and multilevel wavelet analysis, which significantly increased the quality of classification of RBF neural network. This paper creates a framework of fast identification of herba epimedii icariin in raw herba epimedii by FTIR spectra via wavelet analysis and RBF neural network.

  • Article: [Effects of storage time on magnolol and honokiol contents in bark of Magnolia officinalis].

    ABSTRACT: To reveal the relationship between the storage time of the bark of Magnolia officinalis and the content of phenols in it, and lay a theoretical foundation for the harvest, processing, management and storage. The contents of magnolol and honokoiol in 15 bark samples, collected from the main producing areas in China, were determined in the time of freshly harvest and 3 and 10 years after respectively by HPLC method. It showed that within a certain period of time, bark storage was favorable to conversion and accumulation of phenols, that the content of magnolol tended to increase from year 0 to year 3, then followed by slight decrease with years on account of volatilization of phenols, but was still higher when the bark was stored for 10 years than that that when the bark was freshly harvested, and the content of honokoiol still tended to increase when the bark had been stored for 10 years. The phenols in bark of M. officinalis is quite stable and the bark can be stored for 10 years or longer.

    ABSTRACT: To establish an effective method for the identification of Herba Epimedii. The HPLC fingerprints of 8 officinal species including 5 species recorded in 'Chinese Pharmacopoeia', which are Epimedium brevicornu, E. sagittatum, E. pubescens, E. koreanum and E. wushanense, and the other 3 species, E. acuminatum, E. myrianthum and E. leptorrhizum, recorded in 'Guizhou Quality Criteria for Traditional Chinese Medical Material and Nationality Medical Material', and of other 24 species in genus Epimedium in China as well had been analyzed and some fingerprinting features for the identification had been pointed out. The fingerprinting features of E. koreanum are rather different from others and can be used for the species identification. There are some common fingerprinting features in the most samples of E. brevicornu, E. acuminatum and E. myrianthum, respectively, and these features are seldom found in the samples of other species. The HPLC fingerprints of E. pubescens and E. leptorrhizum appear steadily among all samples of the species respectively, but it is difficult to distinguish the spcecies from other species with the HPLC fingerprints only. The HPLC fingerprints of E. sagittatum and E. wushanense vary greatly within the species. It is also showed that the quality of Herba Epimedii varied significantly according to the HPLC features.

  • Article: Fast quality control of Herba Epimedii by using Fourier transform infrared spectroscopy.

    ABSTRACT: Herba Epimedii is a well-known traditional Chinese medicine (TCM) having the effect of nourishing the kidney and strengthening the 'Yang'. Its primary effective constituents are considered to be the 8-prenyl flavonols, which can be assorted into 4'-methoxyl-prenylflavonols (MPFs) and 4'-hydroxyl-prenylflavonols (HPFs), according to the group (methoxyl or hydroxyl) located at 4' in their structures. The Fourier transform infrared spectroscopy (FT-IR) has been widely used in the researches of TCMs. In the present study, the FT-IR was attempted to be applied in the quality control of Herba Epimedii. We compared the IR spectra of 17 pure flavonoids, of which eight were derived from Herba Epimedii, and found a characteristic absorption peak at 1259+/-1 cm(-1), corresponding to the MPFs, the major 8-prenyl flavonols in the aerial parts of the Epimedium species. This peak could also be found in the IR spectra of both the herbal samples and their 70% ethanol extracts. Moreover, the intensity of this peak was in the direct correlation with the total content of MPFs. The correlation values, representing the semblance of two spectra, of the IR spectrum of herbal sample and icariin, in the range of 1280-1200 cm(-1), had been established to be a good index for the quality control of the herbs. Accordingly, a correlation value of not less than 0.50 could be used as the essential screening criteria for the herbs. The FT-IR could be used for the fast and effective quality control of Herba Epimedii.

  • Article: [Study on total alkaloids content and its influential factors in medicinal materials of Tripterygium].

    ABSTRACT: To investigate the total alkaloids content profile and diversity of medicinal materials of Tripterygium. The total alkaloids were extracted by organic solvent and purified by column chromatography (aluminium oxide), and determined by UV spectrometry. The total alkaloids content in phloem was 2 to 5 times richer than that in xylem, and their changes had shown concordance. For the different species/types, the content from high to low in order was T. hypoglaucum, Tripterygium F, T. wilfordii and T. regellii, but in each type, the disparity among different populations content was very small. The populations with higher content were in Suichuan, Jiangxi Province, Huangshan, Anhui Province, Jingning, Zhejiang Province and Kunming, Yuannan Province; which came from T. hypoglaucum and Tripterygium F. The degree of difference of total alkaloids content among individuals in each population was different. The populations, which had a rich content diversity, were in Jingning, Zhejiang Province, Taining, Fujian Province and Kunming, Yunnan Province. They will be used for good germplasm selecting. The thickness of medicinal materials did not affect the total alkaloids content in the same plant. The total alkaloids content in xylem changed with seasons: the highest in March, then declining from June to December, the lowest in December, While this did not happen in phloem.

    ABSTRACT: Herba Epimedii, the dried aerial parts of Epimedium brevicornum Maxim., E. sagittatum Maxim., E. pubescens Maxim., E. wushanense T. S. Ying, and E. koreanum Nakai, has been widely used in the field of nourishing the kidney and reinforcing 'Yang' for centuries. The descriptions of external appearance of Herba Epimedii are rather simple, so it is difficult to identify the raw materials of Herba Epimedii only with the traditional methods. In the present paper, the methods of FTIR and the second derivative IR spectra were used for the identification of Herba Epimedii, by which, E. brevicornum Maxim. could be identified from E. koreanum Nakai, and so E. sagittatum (Sieb. et Zucc.) Maxim. from E. acuminatum Franch., as well as E. pubescens Maxim. from E. stellulatum Stearn, E. baojingensis Q. L. Chen et B. M. Yang, E. zhushanense K. F. Wu & S X. Qian and E. myrianthum Stearn. According to the features of the IR spectra of each species, a method based on FTIR for the identification of Herba Epimedii was established, and it conduced to solve the confusion of species of Herba Epimedii.

  • Article: [Systematic studies on quality of main species of Herba epimedii].

    ABSTRACT: To achieve the quality control of Herba Epimedii, and to evaluate the quality of medical materials of the 8 official species including the 5 species recorded in 'Chinese Pharmacopoeia' including Epimedium brevicornum, E. sagittatum, E. pubescens, E. koreanum and E. wushanense, and the 3 other species (E. acuminatum, E. myrianthum, E. leptorrhizum) recorded in 'Guizhou Quality Criteria for Traditional Chinese Medical Material and Nationality Medical Material', and E. pseudowushanuse (new species) which is used as E. wushanense for a long time. The contents of icariin and total flavonoids of 102 samples of 9 officinal species of Herba Epimedii were determined by HPLC and UV, respectively. The contents of icariin in about 30% of the samples of the 5 species recorded in 'Chinese Pharmacopoeia' were lower than 0.5%, which is acceptable quality recorded in 'Chinese Pharmacopoeia'. Refering the literatures, we suggested the total contents of epimedin A, B, C and icariin (epi-medium multi-glycosides, ABCI) should be established as a new standard instead of the content of icariin. The content of total flavonoids, not less than 5.0%, and ABCI, not less than 1.3%, could be used to evaluate the quality of the above medical materials efficiently.

    ABSTRACT: To review the quality of Epimedium extract in the market. The contents of icariin, epimedin C, sagittatoside B and total flavonoids in Epimedium extracts sold in the market were assayed by the methods of HPLC and UV respectively. HPLC fingerprintings were obtained at the same time. The contents of icariin in most of the extracts are closely similar with the ones labeled by the companies. 3 type chromatograms were classified in all the HPLC fingerprintings, and were corresponded with their raw materials. The contents of epimedin C, sagittatoside B and total flavonoids were different in the samples with the same content of icariin. We can primarily confirm the origin of raw materials by comparing the HPLC fingerprinting of extracts with the ones of materials. The difference of extracts quality mainly comes from the difference of materials. So we suggest that Epimedium extract product should be labeled the origin of materials, and assayed with more compound contents, to ensure the quality stabilization.

  • Article: [RAPD analysis for genetic relationship and diversity of three species of genus Tripterygium].

    ABSTRACT: To study the genetic relationship and diversity of 3 species which belong to Tripterygium. Samples were collected and divided into 4 types: typical T. wilfordii, typical T. hypoglaucum and their middle type according to morphological characters, and T. regelii. RAPD markers were used to measure the genetic relationship and diversity of 110 individuals from 22 natural populations in China. 10 primers were selected from 100 ones screened. A total of 128 bands were scored and 123 of them were polymorphic. Cluster analysis indicated that all the samples could be divided into 3 parts: 5 individuals from T. regelii gathered closely and were separated from other population and formed single branch. All 4 populations from T. hypoglaucum gathered directly. The middle type showed a nearer relationship with T. wilfordii than with T. hypoglaucum, although there was genetic differentiation between populations from middle type and those from T. wilfordii. The middle type has more diversity than T. wilfordii. The distance between different middle type populations and T. wilfordii was different. The existence of middle type populations between T. wilfordii and T. hypoglaucum suggested that the 2 species should be combined into one species. Considering all populations except T. regeli, the generic differentiation among populations was significant and the genetic diversity existed mainly among different populations. So samples of Tripterygii should be collected from different ppopulations for the reservation of the genetic diversity.

    ABSTRACT: Researches on taxonomy, resources, cultivation, quality, chemicals and processing of Herba Epimedii in last ten years were reviewed. The taxonomy of Chinese species in "Flora of China (Vo1.29)" (published in 2001) and "The genus Epimedium and other herbaceous Berberidaceae" (published in 2002) were mainly introduced and commented. The quality evaluation of raw material of Herba Epimedii, and effects of distribution, growing time and medicinal part on the quality were summarized.

    ABSTRACT: To study the content variation of triptolide in medicinal material of Tripterygium and provide theoretical basis for the hereditary improvement, the gathering and process, the quality evaluation and the provenance division in medicinal material of Tripterygium. HPLC method was used to determine the content of triptolide. The relations between triptolide and germplasm, growth year, gathering season were found out basically. The triptolide contents in xylem are affected by hereditary factors remarkably. While the triptolide contents in phloem are not affected obviously. The accumulation of triptolide needs the certain growth years. However when growth is beyond certain years, the triptolide content decreases with the disintegration of secondary metabolism in xylem. The triptolide in xylem is highest in winter and decreasing in growing season. The triptolide in phloem is less affected by the season.

This paper put forward the concept, main tasks and characteristics on the sustainable development of Chinese medicine resources, and analyses the current situation and problems in China. It has explained the basic principle and the sustainable development framework of Chinese medicines resources. The framework consisted of 4 parts: the research system, protection and control system, evaluation index system and development system. It provided external scientific standard for the research of the sustainable development on Chinese medicine resources.

    ABSTRACT: To study the chemical characteristic, to identify the different forms and to establish the new standard for the quality control of Cortex Magnoliae Officinalis. HPLC method was used with acetonitrile-water (63:37) as the mobile phase at room temperature. The chromatographic column was Lichrospher 100 RP-18e (4.6 mm x 250 mm, 5 microm). The flow rate was 1 mL x min(-1), and the detection wavelength was 294 nm. The chromatograms of 45 individuals from 13 seed resources of Cortex Magnolia Officinalis were recorded. The chemical characteristics analysis and comparability' s calculation of seed resources were made. It was proposed that the area ratio of peak 5 to 6 (characteristic I) and the area ratio of peak 5 and 6 to the total peak areas (characteristic II) are the identification characteristics for different seed resources of Cortex Magnoliae Officinalis. This method can be used effectively to identify the high quality seed resource of Cortex Magnoliae Officinalis.

  • Article: [Research on the technology of soaking under pressure and cutting process of Radix Curcumae].

    ABSTRACT: To determine the best processing technology parameters. The changes of the weights, volatile constituents, total extract quantities by MeOH or CHCl3 and curcumenol contents of the roots of Curcuma wenyujin and C. kwangsiensis roots before and after water soaking were determined, so as to determine the quality control index for the process of preparing Yujin slices. The shortest soaking and moistening time and the appropriate drying temperature were studied. (1) The weight was decreased and the quantities of volatile constituents were changed during water soaking process. But the total extracts quantities and the curcumenol contents didn't change obviously. So the quality control index was decided as soaking as short as possible. (2) The soaking time could be decreased by using high pressure. The soaking time was related with the short diameter of the cross section of the root. (3) The soaking under pressure technology is that soaking on -0.095 MPa for 0.5 h, then on 0.14 MPa for 10-16 h, and moistening for 36-48 h, cutting drying in room temperature or less than 40 degrees C.

    ABSTRACT: To comment on main resource species of Herba Epimedii. Taxonomy, resource investigation and quality evaluation of flavonoid contents were made. 7 species of Herba Epimedii having broad distribution were summarized. According the area of their distribution, they were named Epimedium sagittatum, E. brevicornu, E. acuminatum, E. koreanum, E. pubescens, E. leptorrhizum and E. wushanense. Each distribution in detail and icariin and likewise compounds contents of these 7 species were provided. Other 8 Epimedium species much used were proposed. Medicinal characters 15 species were provided in this paper. On the basis of distribution character, resource quantity and growing with other species mingled, it is suggested that 4 main production or purchasing districts in China be divided. The most important district is northeastern district, where only E. koreanum grows there, with the best guality. Northwestern and northern district is the second one, where E. brevicornu, mainly grows with much stable guality. Then it is eastern and southern district, where E. sagittatum grows with the largest quantity but unstable quality. Southwestern district is the last one, mingle species complex and identification important. In Sichuan province, E. acuminatum and E. pubescens are main species, with other 12 species growing there. In Guizhou province, E. acuminatum, E. leptorrhizum and E. wushanense are main species, other 7 species growing there and E. leptorrhizum with poor quality. It is suggested that E. acuminatum be recorded in Chinese Pharmacopoeia.

HPLC was used. The contents of paeoniflorin and paeonol in 9 collected samples, 5 from P. ostii and 4 from P. suffruticosa, and 6 herb samples bought from different areas were analyzed. The rough contents of 3 paeonol similar compounds and 2 paeoniflorin similar compounds were investigated. 1. There is no obvious difference in the contents of paeonol and paeonoflorin and the constitution of other main compounds between P. ostii and P. suffruticosa. Since they belong to different botanic species, it is suggested that both P. ostii and P. suffruticosa should be recorded in Chinese Pharmacopoeia as the origin of Cortex Paeoniae. 2. The glucosides of paeonol might change to paeonol in the course of the collecting and processing of the herb. Therefore when the contents of paeonol is used as the standard to evaluate the quality of the material and patent medicine, the attention should be paid to the transformation mentioned above.
