    ABSTRACT: Our current marine natural product program investigated the second metabolites of an actinomycete Streptomyces cavourensis YY01-17 originating from the Antarctic ecological niche to discover potential antitumor chemical entities. Two new compounds, along with a known compound, were isolated from the ethyl acetate extract of the fermentation broth of the marine-derived actinomycete, and their structures were elucidated, respectively, as 2(S)-3'-hydroxybutan-2'-yl 2-hydroxypropanoate (1), (E)-3-hydroxy-2,4-dimethylhept-4-enamide (2), and 2-hydroxy-3-methylbutanoic acid (3) on the basis of spectroscopic data interpretation.

  • Article: Secondary metabolites from fungus Nigrospora sp.

    ABSTRACT: Six compounds were purified from the extracts of fungus Z18-17 (Nigrospora sp.) isolated from Scyphiphora hydrophyllacea, a tree from the intertidal zone of Shankou, Guangxi Province of China, which was reported to have cytotoxic activity. Systematic study on the chemical constituents of Nigrospora sp. resulted in the isolation of one new 6-phenylhexanoic acid derivative, one new derivative of uridine, and four known compounds. Their structures were characterized, respectively, as methyl 5-acetamido-6-(4-hydroxyphenyl)-4-oxohexanoate (1), uridine-5'α-hydroxypropanoate (2), 4-hydroxyphenethyl 2-hydroxypropanoate (3), methyl pyroglutamate (4), cyclo (Pro-Val) (5), and cyclo (Phe-Hyp) (6). The cytotoxic activities of the two new compounds against HL-60 cell line were tested and the IC(50) values of the two new compounds were all over 100 μM.

    ABSTRACT: Continuous research on the ethyl acetate extract of the fermentation broth of the marine fungus Y26-02 (Penicillium sp.) led to the purification of one known and three new compounds. Their structures were elucidated, respectively, as butyl 2-(4-oxo-5,6-dihydro-2H-pyran-3-yl) acetate (1), 4-hydroxyphenethyl 2-(4-oxo-5,6-dihydro-2H-pyran-3-yl) acetate (2), 3-hydroxybenzyl 2-(4-oxo-5,6-dihydro-2H-pyran-3-yl) acetate (3), and desoxypatulinic acid (4) on the basis of their spectroscopic and physico-chemical properties.

    Article: Two new compounds from the broth of the marine fungus Penicillium griseofulvum Y19-07.

    ABSTRACT: Two new compounds, 4-hydroxyphenethyl methyl succinate (1) and 4-hydroxyphenethyl 2-(4-hydroxyphenyl)acetate (2), were isolated from the EtOAc extract of the broth of the marine fungus Penicillium griseofulvum Y19-07. Five known compounds were also obtained in this study. The structures of the new compounds were elucidated by 1D and 2D NMR spectroscopy and mass spectrometry. All of the isolates were evaluated for their scavenging properties toward the 2,2-diphenyl-1-picrylhydrazyl free radical by spectroscopic assays. Also, in the cytotoxicity assay of the two new compounds against HL-60 and PC-3 prostate cancer cell lines, compound 2 showed potential activity with an IC(50) value of 64.5 microM against human HL-60 cancer cells.

The structures of the new compounds were determined by spectroscopic and chemical analysis.

  • Article: Cytotoxic piperazine-2,5-dione derivatives from marine fungus Gliocladium sp.

    ABSTRACT: Two new piperazine-2,5-dione derivatives, gliocladride A (1) and B (2), along with deoxymycelianamide (3) were isolated from the mycelial mass of marine fungus Gliocladium sp. Their structures were established on the basis of spectral data, and the stereochemical assignments were made by chiral HPLC analysis of the hydrolyzed compounds and optical rotation comparison with known compound. Their cytotoxic activity was tested on three cancer cell lines, HL-60, U937 and T47D by MTT assay. As a result, gliocladride A (1) and B (2) showed moderate cytotoxic activity against the three cell lines with IC50 values from 11.60 microg/ml to 52.83 microg/ml, while deoxymycelianamide (3) showed strong cytotoxic activity against U937 cell line with IC50 values of 0.785 microg/ml.

Their structures were elucidated, respectively, as 5-(ethynyloxy)-3-hydroxy-3,6-dihydro-2H-pyran-2-one (1), 4-(butoxymethyl)benzene-1,2-diol (2), 4-(methoxymethyl)benzene-1,2-diol (3), 2-acetoxymethylphenol (4), N-(2-phenylethyl)acetamide (5), and 4-hydroxyphenethyl acetate (6) on the basis of their spectroscopic and physico-chemical properties.

  • Article: Piezoelectric diffuser/nozzle micropump with double pump chambers

    ABSTRACT: To eliminate check valve fatigue and valve clogging, diffuser/nozzle elements are used for flow rectification in a valveless diffuser/nozzle micropump instead of valves. However, the application of this type of micropump is restricted because of its pulsating or periodic flow and low pump flux. In this paper, a diffuser/nozzle Si/Glass micropump with two pump chambers by IC and MEMS technology is designed. The fabrication process requires only one mask and one etch step, so that the fabrication has the advantages of low cost, short processing period, and facilitation of miniaturization. The pump is equipped with a glass cover board so as to conveniently observe the flow status. Pump-chambers and diffuser elements are fabricated by the anisotropic KOH-etch technique on the silicone substrate, and the convex corner is designed to compensate for an anisotropic etch. The driving force of the micropump is produced by the PZT piezoelectric actuator. The pump performance with both actuators actuated in anti-or same-phase mode is also researched. The result indicates that the micropump achieves great performance with the actuators working at anti-phase. This may be because the liquid flows steadily, pulse phenomenon is very weak, and the optimal working frequency, pump back pressure, and flow rate are both double that of the pump driven in same-phase.

    ABSTRACT: Four new polyketide derivatives, Trichodermatides A-D (1-4) were isolated from the marine-derived fungus Trichoderma reesei. Trichodermatide A (1) is an unprecedented example of a polyketide with a ketal-containing pentacyclic skeleton. The chemical structures and absolute configurations of compounds 1-4 were elucidated by extensive spectroscopic methods, especially 2D NMR and CD spectral analysis, and supported by their proposed biosynthesis pathway. The cytotoxicity of 1-4 was evaluated against A375-S2 human melanoma cell line.

    ABSTRACT: A new piperazine-2,5-dione, named gliocladride, was isolated from marine fungus Gliocladium sp. The structure was established on the basis of spectral data, and the stereochemical assignments were made by 1H NMR spectrum and chiral HPLC of the hydrolyzed compound. Gliocladride showed a cytotoxic effect with an IC50 value of 3.86 microg/ml against human A375-S2 melanoma cell line.

    ABSTRACT: The uptake and transepithelial transport of the three main constituents macrocarpal A (M-A), macrocarpal B (M-B), and cypellocarpa C (Cy-C) from the fruits of Eucalyptus globulus Labill. were investigated. Monolayers of the human intestinal epithelial cancer cell line Caco-2 were incubated with M-A, M-B, and Cy-C to model its intestinal absorption and transport, respectively. The determination of compounds was performed by HPLC. The apparent permeability coefficients (P(app)) for M-A, M-B, and Cy-C in the apical-to-basolateral direction of a Caco-2 monolayer were (1.70+/-0.06)x10(-6), (1.99+/-0.10)x10(-6), and (6.08+/-0.41)x10(-6)cm/s, respectively. In the presence of iodoacetamide, the P(app) of Cy-C were both reducted in apical-to-basolateral and basolateral-to-apical directions. M-A and M-B appear to accumulate in the epithelial cells. The intestinal absorption of M-A, M-B, and Cy-C was passive diffusion as the dominating process and Cy-C was partly ATP-dependent.

    ABSTRACT: The characteristics of uptake, transepithelial transport and efflux of Z- and E-ajoenes isolated from the bulbs of Allium sativum were studied. A human colon cell model Caco-2 cell monolayers in vitro cultured had been applied to study the characteristics of uptake, transepithelial transport and efflux of Z- and E-ajoenes. The quantitative determination of Z- and E-ajoenes was performed by high-performance liquid chromatography. Z- and E-Ajoenes can be detected only in the apical side and can be metabolized, but both compounds can not be transported from apical-to-basolateral and basolateral-to-apical directions in cultured Caco-2 cell monolayers. The metabolism of Z- and E-ajoenes in Caco-2 cell monolayers can be partially inhibited by vitamin C as an anti-oxidant, metyrapone as an inhibitor to subtype CYP3A of cytochrome P450 drug metabolism enzymes, and sodium azide as an inhibitor to ATP production. It is shown that neither Z-ajoene nor E-ajoene can pass through Caco-2 cell monolayers, and that they can be metabolized by the cells. The metabolism might be in correlation with cytochrome P450 drugs metabolism enzymes in Caco-2 cell monolayers.

Their structures were determined based on chemical analysis and spectral methods (IR, 1D and 2D NMR, HR-FAB-MS). Compounds 1-5 were evaluated for cytotoxicity against human liver cancer cell (Hep G), and most of them exhibited potent activity. Compound 1 display the highest potency with IC50 values 10.4 microg mL-1.

Compounds were separated by column chromatography and their structures were elucidated by means of chemical and spectral analysis. Six compounds were isolated from the ethyl acetate and n-butanol extracts of the fermentation of marine fungus Alternalia sp. Their structures were elucidated as p-benzyloxy-phenol ( I ), p-hydroxy phenyl ethylamine( II ), 3-hydroxymethyl-8-hydroxyl-pyrrolopiperazine-2, 5-dione ( III ), 3-isobutyl-6-secbutyl-piperazine-2, 5-dione (IV), 5alpha, 8alpha-epidioxy-ergosta-6, 22-diene-3beta-ol (V), 3beta-hydroxxy-cholesta-5-ene (VI). Compounds I , II, III, IV have the activity of inducing morphological deformation of mycelia germinated from conidia of Pyricularia oryzae. Compounds I , II , III were isolated from the genus Alternalia for the first time.
