Article: Markers of Muscle Damage and Performance Recovery following Exercise in the Heat.

ABSTRACT: PURPOSE: To determine whether competitive intermittent exercise in the heat affects recovery, aggravates markers of muscle fiber damage, and delay the recovery of performance and muscle glycogen stores. METHODS: Plasma creatine kinase, serum myoglobin, muscle glycogen and performance parameters (sprint, endurance and neuromuscular testing) were evaluated in 17 semiprofessional soccer players before, immediately after and during 48 h of recovery from a match played in 43°C (HOT) and compared to a control match (21°C with similar turf and set-up). RESULTS: Muscle temperature was ~ 1°C higher (P<0.001) following the game in HOT compared to control, and reached individual values between 39.9 and 41.1°C. Serum myoglobin levels increased by more than 3 fold following the matches (P<0.01), but values were not different in HOT compared to control and they were similar to baseline values following 24 h of recovery. Creatine kinase was significantly elevated both immediately and 24 h after the matches, but the response following HOT was reduced compared to control. Muscle glycogen responses were similar across trials and remained depressed for more than 48 h following both matches. Sprint performance and voluntary muscle activation were impaired to a similar extend following the matches (sprint by ~ 2% and voluntary activation by ~ 1.5%; P<0.05). Both of these performance parameters as well as intermittent endurance capacity (estimated by a Yo-Yo IR1 test) were fully recovered 48 h after both matches. CONCLUSIONS: Environmental heat stress does not aggravate the recovery response from competitive intermittent exercise associated with elevated muscle temperatures and markers of muscle damage, delayed resynthesis of muscle glycogen and impaired post-match performance.Medicine and science in sports and exercise 12/2012; · 3.71 Impact Factor -
Article: Individual responses to short-term heat acclimatisation as predictors of football performance in a hot, dry environment.

ABSTRACT: To identify the relationship between field performance in a hot environment and individual heat acclimatisation responses in football players. Nineteen semiprofessional football players completed a match in 21°C followed by 6 days of acclimatisation in dry heat (38-43°C, 12-30% relative humidity) and a match in ~43°C. A heat-response test (30 min walk+30 min seated; 44°C) was performed at the beginning and end of the acclimatisation period. The acclimatisation period increased sweat rate by 34% during a standard heat-exposure test and reduced sweat sodium concentration by 18% (both p≤0.005). Plasma volume changes showed large interindividual differences (-10 to +20%). Match-running performance was impaired in hot ambient condition and demonstrated marked interindividual differences (total distance -6.0±5.8%, high-intensity running -16.4±21.5%, both p≤0.002). Only haematological markers investigated during the heat-response test correlated with the ability of the player to cope with heat stress in a competitive situation; that is, changes in haematocrit between the heat-response tests were correlated to changes in total running during the game, r=-0.75; 90%CI [-0.88 to -0.51]. Heat acclimatisation responses and in turn, match-running performance in the heat, are highly individual. The players displaying the largest haematological adaptations were able to maintain the same activity when playing in the heat as when playing in temperate conditions. Article: Skeletal muscle glycogen content and particle size of distinct subcellular localizations in the recovery period after a high-level soccer match.

ABSTRACT: Whole muscle glycogen levels remain low for a prolonged period following a soccer match. The present study was conducted to investigate how this relates to glycogen content and particle size in distinct subcellular localizations. Seven high-level male soccer players had a vastus lateralis muscle biopsy collected immediately after and 24, 48, 72 and 120 h after a competitive soccer match. Transmission electron microscopy was used to estimate the subcellular distribution of glycogen and individual particle size. During the first day of recovery, glycogen content increased by ~60% in all subcellular localizations, but during the subsequent second day of recovery glycogen content located within the myofibrils (Intramyofibrillar glycogen, a minor deposition constituting 10-15% of total glycogen) did not increase further compared with an increase in subsarcolemmal glycogen (-7 vs. +25%, respectively, P = 0.047). Conversely, from the second to the fifth day of recovery, glycogen content increased (53%) within the myofibrils compared to no change in subsarcolemmal or intermyofibrillar glycogen (P < 0.005). Independent of location, increment in particle size preceded increment in number of particles. Intriguingly, average particle size decreased; however, in the period from 3 to 5 days after the match. These findings suggest that glycogen storage in skeletal muscle is influenced by subcellular localization-specific mechanisms, which account for an increase in number of glycogen particles located within the myofibrils in the period from 2 to 5 days after the soccer match.Arbeitsphysiologie 02/2012; 112(10):3559-67. · 2.15 Impact Factor -
Article: Physiological responses and physical performance during football in the heat.

ABSTRACT: To examine the impact of hot ambient conditions on physical performance and physiological responses during football match-play. Two experimental games were completed in temperate (∼ 21°C; CON) and hot ambient conditions (∼ 43°C; HOT). Physical performance was assessed by match analysis in 17 male elite players during the games and a repeated sprint test was conducted after the two game trials. Core and muscle temperature were measured and blood samples were obtained, before and after the games. Muscle and core temperatures were ∼ 1°C higher (P<0.05) in HOT (40.3 ± 0.1 and 39.5 ± 0.1°C, respectively) compared to CON (39.2 ± 0.1 and 38.3 ± 0.1°C). Average heart rate, plasma lactate concentration, body weight loss as well as post-game sprint performance were similar between the two conditions. Total game distance declined (P<0.05) by 7% and high intensity running (>14 km ⋅ h(-1)) by 26% in HOT compared to CON), but peak sprint speed was 4% higher (P<0.05) in HOT than in CON, while there were no differences in the quantity or length of sprints (>24 km ⋅ h(-1)) between CON and HOT. In HOT, success rates for passes and crosses were 8 and 9% higher (P<0.05), respectively, compared to CON. Delta increase in core temperature and absolute core temperature in HOT were correlated to total game distance in the heat (r = 0.85 and r = 0.53, respectively; P<0.05), whereas, total and high intensity distance deficit between CON and HOT were not correlated to absolute or delta changes in muscle or core temperature. Total game distance and especially high intensity running were lower during a football game in the heat, but these changes were not directly related to the absolute or relative changes in core or muscle temperature. Article: Pro- and anti-angiogenic factors in human skeletal muscle in response to acute exercise and training.

ABSTRACT: This study examined the effect of acute exercise and 4 weeks of aerobic training on skeletal muscle gene and protein expression of pro- and anti-angiogenic factors in 14 young male subjects. Training consisted of 60 min of cycling (∼60% of ), 3 times/week. Biopsies were obtained from vastus lateralis muscle before and after training. Muscle interstitial fluid was collected during cycling at weeks 0 and 4. Training increased (P < 0.05) the capillary: fibre ratio and capillary density by 23% and 12%, respectively. The concentration of interstitial vascular endothelial growth factor (VEGF) in response to acute exercise increased similarly (>6-fold; P < 0.05) before and after training. Resting protein levels of soluble VEGF receptor-1 in interstitial fluid, and of VEGF, thrombospondin-1 (TSP-1) and tissue inhibitor of matrix metalloproteinase-1 (TIMP1) in muscle were unaffected by training, whereas endothelial nitric oxide synthase protein levels in muscle increased by 50% (P < 0.05). Before and after training, acute exercise induced a similar increase (P < 0.05) in the mRNA level of angiopoietin 2, matrix metalloproteinase 9 and TSP-1. After training, TIMP1 mRNA content increased with exercise (P < 0.05). In conclusion, acute exercise induced a similar increase in the gene-expression of both pro- and anti-angiogenic factors in untrained and trained muscle. We propose that the increase in anti-angiogenic factors with exercise is important for modulation of angiogenesis. Article: Brain temperature and exercise performance.

ABSTRACT: Events arising within the central nervous system seem to be a major factor in the aetiology of hyperthermia-induced fatigue. Thus, various studies with superimposed electrical nerve stimulation or transcranial magnetic stimulation have shown that both passive and exercise-induced hyperthermia will impair voluntary motor activation during sustained maximal contractions. In humans, the brain temperature increases in parallel with that of the body core, making it very difficult to evaluate the independent effect of the cerebral temperature. Experiments with separate manipulation of the brain temperature in exercising goats indicate that excessive brain hyperthermia will directly affect motor performance. However, several homeostatic changes arise in parallel with hyperthermia, including factors that may influence both peripheral and central fatigue, and it is likely that these changes interact with the inhibitory effect of an elevated brain temperature.Experimental physiology 11/2011; 97(3):333-9. · 3.17 Impact Factor -
Article: Maximal voluntary contraction force, SR function and glycogen resynthesis during the first 72 h after a high-level competitive soccer game.

ABSTRACT: The aim of this study was to examine maximal voluntary knee-extensor contraction force (MVC force), sarcoplasmic reticulum (SR) function and muscle glycogen levels in the days after a high-level soccer game when players ingested an optimised diet. Seven high-level male soccer players had a vastus lateralis muscle biopsy and a blood sample collected in a control situation and at 0, 24, 48 and 72 h after a competitive soccer game. MVC force, SR function, muscle glycogen, muscle soreness and plasma myoglobin were measured. MVC force sustained over 1 s was 11 and 10% lower (P < 0.05) after 0 and 24 h, respectively, compared with control. The rate of SR Ca(2+) uptake at 800 nM [Ca(2+)](free) was lower (P < 0.05) after 0 h (2.5 μmol Ca(2+) g prot(-1) min(-1)) than for all other time points (24 h: 5.1 μmol Ca(2+) g prot(-1) min(-1)). However, SR Ca(2+) release rate was not affected. Plasma myoglobin was sixfold higher (P < 0.05) immediately after the game, but normalised 24 h after the game. Quadriceps muscle soreness (0-10 VAS-scale) was higher (P < 0.05) after 0 h (3.6), 24 h (1.8), 48 h (1.1) and 72 h (1.4) compared with control (0.1). Muscle glycogen was 57 and 27% lower (P < 0.001) 0 and 24 h after the game compared with control (193 and 328 vs. 449 mmol kg d w(-1)). Article: VO2 kinetics and performance in soccer players after intense training and inactivity.

ABSTRACT: The study's purpose was to examine the effects of a short-term period with intensified training or training cessation of trained soccer players on VO(2) kinetics at 75% maximal aerobic speed, oxidative enzymes, and performance in repeated high-intensity exercise. After the last match of the season, 18 elite soccer players were, for a 2-wk period, assigned to a high-intensity training group (n = 7) performing 10 training sessions mainly consisting of aerobic high-intensity training (8 × 2 min) and speed endurance training (10-12 × 30-s sprints) or a training cessation group (n = 11) that refrained from training. For the training cessation group, VO(2) kinetics became slower (P < 0.05) with a larger time constant (τ = 21.5 ± 2.9 vs 23.8 ± 3.2 s (mean ± SD, before vs after)) and a larger mean response time (time delay + τ = 45.0 ± 1.8 vs 46.8 ± 2.2 s). The amount of muscle pyruvate dehydrogenase (17%, P < 0.01) and maximal activity of citrate synthase (12%) and 3-hydroxyacyl-CoA (18%, P < 0.05) were lowered. In addition, the fraction of slow twitch fibers (56% ± 18% vs 47% ± 15%, P < 0.05), Yo-Yo intermittent recovery level 2 test (845 ± 160 vs 654 ± 99 m), and the repeated sprint performance (33.41 ± 0.96 vs 34.11 ± 0.92 s, P < 0.01) were reduced. For the high-intensity training group, running economy was improved (P < 0.05), and the amount of pyruvate dehydrogenase (17%) and repeated sprint performance (33.44 ± 1.17 vs 32.81 ± 1.01 s) were enhanced (P < 0.05). Inactivity slows VO(2) kinetics in association with a reduction of muscle oxidative capacity and repeated high-intensity running performance. Article: Muscle adaptations and performance enhancements of soccer training for untrained men.

ABSTRACT: We examined the physical demands of small-sided soccer games in untrained middle-age males and muscle adaptations and performance effects over 12 weeks of recreational soccer training in comparison with continuous running. Thirty-eight healthy subjects (20-43 years) were randomized into a soccer (SO), running (RU) and control (CO) group. Two-three weekly 1-h training sessions were performed. Muscle lactate (30.1 +/- 4.1 vs. 15.6 +/- 3.3 mmol/kg d.w.), blood lactate, blood glucose and time above 90% HR(max) (20 +/- 4% vs. 1 +/- 1%) were higher (p < 0.05) during training in SO than in RU. After 12 weeks of training, quadriceps muscle mass and mean muscle fibre area were 9 and 15% larger (p < 0.05) in SO, but unaltered in RU, and in SO, the fraction of FTx fibres was lowered (10.7 +/- 1.8 vs. 17.9 +/- 3.2%). In SO, citrate synthase activity was 10 and 14% higher (p < 0.05) after 4 and 12 weeks, but unaltered in RU. After 4 weeks VO(2max) and Yo-Yo IE2 performance were elevated (p < 0.05) to a similar extent in SO (7 and 37%) and RU (6 and 36%) but increased further (p < 0.05) from 4 to 12 weeks in SO (6 and 23%). In SO, 30-m sprint performance was improved (p < 0.05) by 0.11 +/- 0.02 s. Blood lactate during running at 11 km/h was lowered (p < 0.05) from 0 to 4 and 4 to 12 weeks (2.6 +/- 0.3 vs. 3.8 +/- 0.6 vs. 6.1 +/- 0.9 mM) and from 0 to 12 weeks in RU. No changes occurred for CO. Article: Effect of 2-wk intensified training and inactivity on muscle Na+-K+ pump expression, phospholemman (FXYD1) phosphorylation, and performance in soccer players.

ABSTRACT: The present study examined muscle adaptations and alterations in performance of highly trained soccer players with intensified training or training cessation. Eighteen elite soccer players were, for a 2-wk period, assigned to either a group that performed high-intensity training with a reduction in the amount of training (HI, n = 7), or an inactivity group without training (IN, n = 11). HI improved (P < 0.05) performance of the 4th, 6th, and 10th sprint in a repeated 20-m sprint test, and IN reduced (P < 0.05) performance in the 5th to the 10th sprints after the 2-wk intervention period. In addition, the Yo-Yo intermittent recovery level 2 test performance of IN was lowered from 845 +/- 48 to 654 +/- 30 m. In HI, the protein expression of the Na(+)-K(+) pump alpha(2)-isoform was 15% higher (P < 0.05) after the intervention period, whereas no changes were observed in alpha(1)- and beta(1)-isoform expression. In IN, Na(+)-K(+) pump expression was not changed. In HI, the FXYD1ser68-to-FXYD1 ratio was 27% higher (P < 0.01) after the intervention period, and, in IN, the AB_FXYD1ser68 signal was 18% lower (P < 0.05) after inactivity. The change in FXYD1ser68-to-FXYD1 ratio was correlated (r(2) = 0.35; P < 0.05) with change in performance in repeated sprint test. The present data suggest that short-term intensified training, even for trained soccer players, can increase muscle Na(+)-K(+) pump alpha(2)-isoform expression, and that cessation of training for 2 wk does not affect the expression of Na(+)-K(+) pump isoforms. Article: High-intensity training versus traditional exercise interventions for promoting health.

ABSTRACT: The purpose of this study was to determine the effectiveness of brief intense interval training as exercise intervention for promoting health and to evaluate potential benefits about common interventions, that is, prolonged exercise and strength training. Thirty-six untrained men were divided into groups that completed 12 wk of intense interval running (INT; total training time 40 min wk(-1)), prolonged running (approximately 150 min wk(-1)), and strength training (approximately 150 min wk(-1)) or continued their habitual lifestyle without participation in physical training. The improvement in cardiorespiratory fitness was superior in the INT (14% +/- 2% increase in V˙O2max) compared with the other two exercise interventions (7% +/- 2% and 3% +/- 2% increases). The blood glucose concentration 2 h after oral ingestion of 75 g of glucose was lowered to a similar extent after training in the INT (from 6.1 +/- 0.6 to 5.1 +/- 0.4 mM, P < 0.05) and the prolonged running group (from 5.6 +/- 1.5 to 4.9 +/- 1.1 mM, P < 0.05). In contrast, INT was less efficient than prolonged running for lowering the subjects' resting HR, fat percentage, and reducing the ratio between total and HDL plasma cholesterol. Furthermore, total bone mass and lean body mass remained unchanged in the INT group, whereas both these parameters were increased by the strength-training intervention. INT for 12 wk is an effective training stimulus for improvement of cardiorespiratory fitness and glucose tolerance, but in relation to the treatment of hyperlipidemia and obesity, it is less effective than prolonged training. Article: CNS fatigue provoked by prolonged exercise in the heat.

ABSTRACT: Exercise-induced hyperthermia is associated with central fatigue as indicated by an impaired ability to sustain maximal motor activation during prolonged voluntary efforts. Therefore, exercise in hot environments challenges not only to the cardiorespiratory and locomotive systems but also to the brain. However, exercise with superimposed hyperthermia is not only a challenge to the brain it also provides an excellent model for studying factors of importance for central fatigue. Excessive heat storage within the brain appears to be the primary cause for the central fatigue during exercise in the heat, but pharmacological manipulations provide evidence for involvement of the dopaminergic system and other monoamines. Thus, enhanced dopaminergic activity may counteract hyperthermia mediated central fatigue and improve performance in the heat, while noradrenaline re-uptake inhibition appears to aggravate central fatigue and degrade exercise performance. Hyperthermia mediated central fatigue may include other cerebral perturbations such as reduced perfusion of the brain, accumulation of ammonia or depletion of neuronal energy stores, but further research is needed to elucidate their possible contributions.Frontiers in bioscience (Elite edition) 01/2010; 2:779-92. -
Article: Reduced volume and increased training intensity elevate muscle Na+-K+ pump alpha2-subunit expression as well as short- and long-term work capacity in humans.

ABSTRACT: The present study examined muscle adaptations and alterations in work capacity in endurance-trained runners as a result of a reduced amount of training combined with speed endurance training. For a 6- to 9-wk period, 17 runners were assigned to either a speed endurance group with a 25% reduction in the amount of training but including speed endurance training consisting of six to twelve 30-s sprint runs 3-4 times/wk (SET group n = 12) or a control group (n = 5), which continued the endurance training ( approximately 55 km/wk). For the SET group, the expression of the muscle Na(+)-K(+) pump alpha(2)-subunit was 68% higher (P < 0.05) and the plasma K(+) level was reduced (P < 0.05) during repeated intense running after 9 wk. Performance in a 30-s sprint test and the first of the supramaximal exhaustive runs was improved (P < 0.05) by 7% and 36%, respectively, after the speed endurance training period. In the SET group, maximal O(2) uptake was unaltered, but the 3-km (3,000-m) time was reduced (P < 0.05) from 10.4 +/- 0.1 to 10.1 +/- 0.1 min and the 10-km (10,000-m) time was improved from 37.3 +/- 0.4 to 36.3 +/- 0.4 min (means +/- SE). Muscle protein expression and performance remained unaltered in the control group. Article: Impact of carbohydrate supplementation during endurance training on glycogen storage and performance.

ABSTRACT: Glucose ingestion may improve exercise endurance, but it apparently also influences the transcription rate of several metabolic genes and it alters muscle metabolism during an acute exercise bout. Therefore, we investigated how chronic training responses are affected by glucose ingestion. In previously untrained males performance and various muscular adaptations were evaluated before and after 8 weeks of supervised endurance training conducted either with (n = 8; CHO group) or without (n = 7; placebo) glucose supplementation. The two groups achieved similar improvements in maximal oxygen uptake and peak power output during incremental cycling (both parameters elevated by 17% on average) and both groups lost approximately 3 kg of fat mass during the 8 weeks of training. An equal reduction in respiratory exchange ratio (0.02 units) during submaximal exercise was observed in both groups. Beta-hydroxyacyl-CoA-dehydrogenase activity was increased in both groups, however, to a larger extent in the placebo group (45 +/- 11%) than CHO (23 +/- 9%, P < 0.05). GLUT-4 protein expression increased by 74 +/- 14% in the placebo group and 45 +/- 14% in CHO (both P < 0.05), while resting muscle glycogen increased (P < 0.05) to a larger extent in the placebo group (96 +/- 4%) than CHO (33 +/- 2%). Article: Adipose triglyceride lipase in human skeletal muscle is upregulated by exercise training.

ABSTRACT: Mobilization of fatty acids from stored triacylglycerol (TG) in adipose tissue and skeletal muscle [intramyocellular triacylglycerol (IMTG)] requires activity of lipases. Although exercise training increases the lipolytic capacity of skeletal muscle, the expression of hormone-sensitive lipase (HSL) is not changed. Recently, adipose triglyceride lipase (ATGL) was identified as a TG-specific lipase in various rodent tissues. To investigate whether human skeletal muscle ATGL protein is regulated by endurance exercise training, 10 healthy young men completed 8 wk of supervised endurance exercise training. Western blotting analysis on lysates of skeletal muscle biopsy samples revealed that exercise training induced a twofold increase in skeletal muscle ATGL protein content. In contrast to ATGL, expression of comparative gene identification 58 (CGI-58), the activating protein of ATGL, and HSL protein was not significantly changed after the training period. The IMTG concentration was significantly decreased by 28% at termination of the training program compared with before. HSL-phoshorylation at Ser(660) was increased, HSL-Ser(659) phosporylation was unchanged, and HSL-phoshorylation at Ser(565) was decreased altogether, indicating an enhanced basal activity of this lipase. No change was found in the expression of diacylglycerol acyl transferase 1 (DGAT1) after training. Article: Recreational soccer is an effective health-promoting activity for untrained men.

ABSTRACT: To examine the effects of regular participation in recreational soccer on health profile, 36 healthy untrained Danish men aged 20-43 years were randomised into a soccer group (SO; n = 13), a running group (RU; n = 12) and a control group (CO; n = 11). Training was performed for 1 h two or three times per week for 12 weeks; at an average heart rate of 82% (SEM 2%) and 82% (1%) of HR(max) for SO and RU, respectively. During the 12 week period, maximal oxygen uptake increased (p<0.05) by 13% (3%) and 8% (3%) in SO and RU, respectively. In SO, systolic and diastolic blood pressure were reduced (p<0.05) from 130 (2) to 122 (2) mm Hg and from 77 (2) to 72 (2) mm Hg, respectively, after 12 weeks, with similar decreases observed for RU. After the 12 weeks of training, fat mass was 3.0% (2.7 (0.6) kg) and 1.8% (1.8 (0.4) kg) lower (p<0.05) for SO and RU, respectively. Only SO had an increase in lean body mass (1.7 (0.4) kg, p<0.05), an increase in lower extremity bone mass (41 (8) g, p<0.05), a decrease in LDL-cholesterol (2.7 (0.2) to 2.3 (0.2) mmol/l; p<0.05) and an increase (p<0.05) in fat oxidation during running at 9.5 km/h. The number of capillaries per muscle fibre was 23% (4%) and 16% (7%) higher (p<0.05) in SO and RU, respectively, after 12 weeks. No changes in any of the measured variables were observed for CO. ABSTRACT: The present review addresses mechanisms of importance for hyperthermia-induced fatigue during short intense activities and prolonged exercise in the heat. Inferior performance during physical activities with intensities that elicit maximal oxygen uptake is to a large extent related to perturbation of the cardiovascular function, which eventually reduces arterial oxygen delivery to the exercising muscles. Accordingly, aerobic energy turnover is impaired and anaerobic metabolism provokes peripheral fatigue. In contrast, metabolic disturbances of muscle homeostasis are less important during prolonged exercise in the heat, because increased oxygen extraction compensates for the reduction in systemic blood flow. The decrease in endurance seems to involve changes in the function of the central nervous system (CNS) that lead to fatigue. The CNS fatigue appears to be influenced by neurotransmitter activity of the dopaminergic system, but may primarily relate to inhibitory signals from the hypothalamus arising secondary to an increase in brain temperature. ABSTRACT: The present study examined muscle adaptations and alterations in work capacity in endurance-trained runners after a change from endurance to sprint training. Fifteen runners were assigned to either a sprint training (ST, n = 8) or a control (CON, n = 7) group. ST replaced their normal training by 30-s sprint runs three to four times a week, whereas CON continued the endurance training (approximately 45 km/wk). After the 4-wk sprint period, the expression of the muscle Na+-K+ pump alpha1-subunit and Na+/H+-exchanger isoform 1 was 29 and 30% higher (P < 0.05), respectively. Furthermore, plasma K+ concentration was reduced (P < 0.05) during repeated intense running. In ST, performance in a 30-s sprint test, Yo-Yo intermittent recovery test, and two supramaximal exhaustive runs was improved (P < 0.05) by 7, 19, 27, and 19%, respectively, after the sprint training period, whereas pulmonary maximum oxygen uptake and 10-k time were unchanged. No changes in CON were observed. The present data suggest a role of the Na+-K+ pump in the control of K+ homeostasis and in the development of fatigue during repeated high-intensity exercise.
