ABSTRACT: To assess the value of fluorescence in situ hybridization (FISH) in the diagnosis of bladder cancer.
Urine samples from 100 patients suspected of having bladder cancer were collected before cystoscopy for immediate urine cytology and FISH analysis. The criteria for FISH abnormality were determined by evaluating the urine specimens from 20 subjects without urogenital neoplasm.
The overall sensitivity of cytology and FISH was 43.2% and 82.4%, and their specificity was 92.3% and 88.5%, with diagnostic concordance rate of 56.0% and 84.0%, respectively. The differences between FISH and cytology showed statistical significance in the sensitivity, diagnostic concordance rate, non-muscle-invasive cancer and primary cancer.
The sensitivity and efficiency of FISH in the detection of bladder cancer are superior to those of cytology, especially for prophase cancer.

ABSTRACT: To study the association of S100A4 mRNA expression level with the differentiation and metastasis of renal cell carcinoma (RCC).
Tissue specimens were obtained from 31 patients undergoing surgery for renal cell carcinomas between May 2001 and May 2004. Reverse transcriptional PCR was performed for amplification of S100A4 mRNA (212 bp) from 31 RCC tissues and matched normal kidney tissues besides the tumor, and the differential S100A4 mRNA expression was analyzed for its association with the clinical manifestations of the patients.
S100A4 mRNA expression was detected in all the RCC tissues and normal tissues adjacent to the carcinoma using fluorescence method, and in 29 cases, S100A4 mRNA expression was significantly higher in the RCC tissues than in the matched adjacent tissues. One case had high expressions in both tissues. Patients with poorly differentiated RCC had significantly higher S100A4 expression index than those with well differentiated RCC (7.94 vs 5.06, P<0.001), and patients with metastasis also had higher expression index (9.61 vs 5.53, P<0.001). No obvious difference was noted in the expression index between granular cell tumor and clear cell carcinoma (6.98 vs 6.02, P>0.05).
S100A4 may serve as a marker for assessing the progression of RCC and provide assistance for clinical therapeutic decisions. S100A4 mRNA expression is correlated to RCC differentiation and may facilitate prognostic evaluation of RCC. Differential expression of S100A4 mRNA is correlated to the metastasis of RCC.

ABSTRACT: To evaluate the effects of preoperative chemotherapy for Wilms' tumor.
Thirty-five surgically treated cases of Wilms tumor were retrospectively analyzed, 27 of which received preoperative chemotherapy.
After chemotherapy, the tumor volume reduced obviously in 17 cases and adverse effects of the chemotherapeutic agents occurred in 15 cases. Subsequent operation was performed in all the patients, of whom 1 died during the operation, 4 had delayed healing of the incisions and 5 still had decreased leucocytes.
Chemotherapy before the operation for Wilms tumor can effectively reduce the complications of the operation. Appropriate chemotherapy protocol and vigorous conservative drug therapy help prevent the occurrence of adverse effects and complications of the chemotherapy.

ABSTRACT: To explore the diagnostic approaches and values of the imaging modalities for traumatic renal injuries.
The clinical records of 74 cases of renal trauma treated in Nanfang Hospital were retrospectively reviewed to assess the diagnostic value of intravenous urography (IVU), type B ultrasonography and computed tomography (CT).
The positivity rates by IVU, type B ultrasonography, and CT were 89% (43/49), 80% (55/68) and 100% (51/51) respectively for the diagnosis of renal trauma.
IVU is rapid and convenient, ultrasonography less costly and invasive, and CT accurate for diagnosis of renal traumas.

ABSTRACT: To investigate the effect of portal venous inoculation of donor splenocytes combined with cyclosporin A (CsA) administration on cardiac allograft survival in mice.
Heterotopic cardiac transplantation between fully allogenic NIH/q and BALB/C strain mice was performed. A modified procedure of neonatal heart-in-ear transplantation, as originally described by Fulmer et al, was adopted. We prepared donor splenocytes from NIH/q or third-party C57BL/6 spleens for BALB/C recipients, which were injected preoperatively via the recipient portal vein or the systemic vein 1 week before the heart-in-ear transplantation. The recipients were subsequently treated with a short course of the immunosuppressive agent, CsA (4 mg/kg starting from 7 d before the operation till 5 d after it).
Portal venous inoculation of donor splenocytes combined with CsA significantly prolonged cardiac graft survival (n=6, P<0.05) that reached 31.00+/-3.23 d, and 2 of the 6 allografts survived for more than 35 d. Donor splenocytes injected via the systemic vein or third-party C57BL/6 mice splenocytes injected via the portal vein did not prolong graft survival (P>0.05). CsA alone or portal venous inoculation of donor-specific splenocytes alone also prolonged graft survival (P<0.05), with mean graft survival time of 18.50+/-2.59 d and 16.11+/-1.97 d respectively.
Combination of portal venous inoculation of donor-specific splenocytes and CsA can prolong murine cardiac allograft survival, which is donor antigen-specific.

ABSTRACT: To assess the fertilizing capacity of human sperm preserved in electrolyte-free (EF) solution at 4 degrees Celsius.
The motility, acrosome status and fertility index of human sperm were respectively assessed before and after preservation in cold EF solution.
The motility of human sperm so preserved for one week was significantly higher than that of the sperm preserved in modified human tubal fluid (43.4+/-7.9% vs 9.5+/-2.5%, P<0.01). Although acrosome status of human sperm in EF solution before reinitiation differed little from that of fresh sperm (capacitated sperm: 7.6+/-1.8% vs 6.4+/-1.8%; acrosome-reacted sperm: 3.0+/-1.7% vs 2.4+/-1.1%, P>0.05), the percentage of capacitated and acrosome-reacted sperm in the EF solution significantly increased after reinitiation (16.0+/-2.3% vs 7.6+/-1.8% and 9.4+/-2.1% vs 3.0+/-1.7% respectively, P<0.01). The penetration rate and fertility index of the sperm in EF solution were comparable with those of fresh sperm (48.1% vs 50.9%; 1.38+/-0.16 vs 1.29+/-0.13, respectively).
Preservation in cold EF solution does not induce human sperm to undergo capacitation and acrosome reaction, and the sperm so preserved for one week possesses comparable fertilizing capacity as fresh sperm does.
