ABSTRACT: To construct an adenovirus vector (Ad-mTSLPR-Ig) expressing the fusion protein of the extracellular domain of mouse TSLPR and the Fc fragment of mouse Ig, further purify and assess the soluble fusion protein mTSLPR-Ig.
The gene of mTSLPR extracellular domain was amplified from cDNA library of mouse thymus by PCR. Then an adenovirus vector (Ad-mTSLPR-Ig) expressing the fusion protein of mTSLPR and the Fc fragment of mouse Ig was constructed. After Ad-mTSLPR-Ig was transfected into COS-7 cells, the supernatants were collected. The fusion proteins in the supernatants were detected by double antibody sandwich ELISA. The soluble fusion proteins mTSLPR-Ig were purified by protein A affinity chromatography.
The gene sequence of Ad-mTSLPR-Ig was confirmed by DNA sequencing, and the recombinat adenoviruses harboring mTSLPR-Ig were successfully obtained after the infection of Ad-mTSLPR-Ig. Fusion proteins mTSLPR-Ig in the supernatants were detected by ELISA assay. The purified fusion protein were obtained by affinity chromatography, and identified by Western blot.
The soluble fusion proteins mTSLPR-Ig have been obtained successfully, which enables further study of its biological activity.

ABSTRACT: Allergen-specific immunotherapy is a potential treatment for allergic diseases. We constructed an allergen-cytotoxic T lymphocyte-associated antigen 4 (CTLA-4)-encoding DNA vaccine, administered it directly to antigen-presenting cells (APCs), and investigated its ability and mechanisms to ameliorate allergic airway inflammation in an asthmatic mouse model. An allergen-CTLA-4 DNA plasmid (OVA-CTLA-4-pcDNA₃.₁) encoding an ovalbumin (OVA) and the mouse CTLA-4 extracellular domain was constructed and transfected into COS-7 cells to obtain the fusion protein OVA-CTLA-4, which was able to bind the B7 ligand on dendritic cells (DCs), and induced CD25⁺ Foxp3⁺ regulatory T (Treg) cells by the coculture of naive CD4⁺ T cells with DCs in vitro. In an animal study, BALB/c mice were sensitized and challenged with OVA to establish the asthmatic model. Vaccination with a high dose of OVA-CTLA-4-pcDNA₃.₁ significantly decreased interleukin-4 (IL-4) and IL-5 levels and eosinophil counts and prevented OVA-induced reduction of the gamma interferon level in the bronchoalveolar lavage fluid. In addition, these mice suffered less severe airway inflammation and had lower levels of OVA-specific IgE and IgG1 titers in serum. Also, high-dose OVA-CTLA-4-pcDNA₃.₁ vaccination inhibited the development of airway hyperreactivity and prevented OVA-induced reduction of the percentages of Foxp3⁺ Treg cells in the spleen. Our results indicate that a high dose of allergen-CTLA-4-encoding DNA vaccine was more effective in preventing an allergen-induced Th2-skewed immune response through the induction of Treg cells and may be a new alternative therapy for asthma.

ABSTRACT: The contribution of emphysema to lung cancer risk has been recognized, but the effect size needs to be further defined. In this study, 565 primary lung cancer cases were enrolled though a prospective lung cancer cohort at Mayo Clinic, and 450 controls were smokers participating in a lung cancer screening study in the same institution using spiral computed tomography (CT). Cases and controls were frequency matched on age, gender, race, smoking status, and residential region. CT imaging using standard protocol at the time of lung cancer diagnosis (case) or during the study (control) was assessed for emphysema by visual scoring CT analysis as a percentage of lung tissue destroyed. The clinical definition of emphysema was the diagnosis recorded in the medical documentation. Using multiple logistic regression models, emphysema (≥ 5% on CT) was found to be associated with a 3.8-fold increased lung cancer risk in Caucasians, with higher risk in subgroups of younger (<65 years old, OR = 4.64), heavy smokers (≥ 40 pack-years, OR = 4.46), and small-cell lung cancer (OR = 5.62). When using >0% or ≥ 10% emphysema on CT, lung cancer risk was 2.79-fold or 3.33-fold higher than controls. Compared with CT evaluation (using criterion ≥ 5%), the sensitivity, specificity, positive and negative predictive values, and the accuracy of the clinical diagnosis for emphysema in controls were 19%, 98%, 73%, 84%, and 83%, respectively. These results imply that an accurate evaluation of emphysema could help reliably identify individuals at greater risk of lung cancer among smokers.

ABSTRACT: Multiple recent genome-wide studies of single nucleotide polymorphisms (SNP) reported associations between candidate chromosome loci and lung cancer susceptibility. We evaluated five of the top candidate SNPs (rs402710, rs2736100, rs4324798, rs16969968, and rs8034191) for their effects on lung cancer risk and overall survival.
Over 1,700 cases and 2,200 controls were included in this study. Seven independent, complementary case-control data sets were tested for risk assessment encompassing cigarette smokers and never smokers, using unrelated controls and unaffected full-sibling controls. Five patient groups were tested for survival prediction stratified by smoking status, histology subtype, and treatment.
After considering a history of chronic obstructive pulmonary disease as a risk factor altering lung cancer risk and comparing to sibling controls, none of the five SNPs remained significant. However, the variant rs4324798 was significant in predicting overall survival (hazard ratio, 0.46; 95% confidence interval, 0.30-0.73; P = 0.001) in small cell lung cancer.
None of the five candidate SNPs in lung cancer risk can be confirmed in our study. The previously reported association could be explained by disparity in tobacco smoke exposure and chronic obstructive pulmonary disease history between cases and controls. Instead, we found rs4324798 to be an independent predictor in small cell lung cancer survival, warranting further elucidation of the underlying mechanisms.

ABSTRACT: To describe the distinctive histopathological changes of nontuberculous mycobacteria lymphadenitis.
An experimental animal model of nontuberculous mycobacteria lymphadenitis was established and the histopathological changes were observed by light microscope. The paraffin imbedded tissue samples from patients suspected of having lymphoid tuberculosis were also detected by triplex polymerase chain reaction and studied by light microscope.
The distinctive histopathological changes of nontuberculous mycobacteria lymphadenitis in the animal model were as follows: (1) Tubercular granuloma formation in lymph nodes which were infected with nontuberculous mycobacteria. Coagulation necrosis was located at the center of the granuloma, and the necrosis looked different from the caseation necrosis caused by mycobacterium tuberculosis. Many neutrophils and their nuclear debris were distributed over the necrosis area. Surrounding the central necrosis area, many epithelioid cells, lymph cells and mononuclear cells could be found. The periphery of the granuloma was surrounded by fibrous tissues. Langhans giant cells could be found in the granuloma and outside the granuloma, while these cells were usually found only in the granuloma of tuberculosis. (2) Serpiginous necrosis was found in the lymph nodes infected with nontuberculous mycobacteria. Many neutrophils and their nuclear debris were distributed over the necrosis area. Around the central necrosis area, many epithelioid cells, lymph cells and mononuclear cells could be found. The fibrous tissues were in the borderline. (3) Star-like necrosis and aristiform necrosis were also found in lymph nodes infected with nontuberculous mycobacteria. A paraffin imbedded tissue sample was detected by triplex polymerase chain reaction and the diagnosis of nontuberculous mycobacteria lymphadenitis was made. In this sample, epithelioid granuloma, serpiginous necrosis and star-like necrosis were found. Neutrophils and their nuclear debris were found distributed over the necrosis area, while epithelioid cells, lymph cells, mononuclear cells and Langhans giant cells were found around the central necrosis area. Polar arrangement of the nuclei of epithelioid cells was evident. The lesion was surrounded by fibrous and collagen tissues.
Serpiginous, star-like and aristiform necrosis were the distinctive histopathological changes of nontuberculous mycobacteria lymphadenitis. Neutrophils and their nuclear debris were abundant over the necrosis area. Polar arrangement of the nuclei of epithelioid cells was also a distinctive histopathological manifestation.

ABSTRACT: we proposed a morphological pyramid multimodal medical image registration algorithm based on Quantum Particle Swarm Optimization (QPSO), this approach utilizes the global affine transformation model between images. With the non-linear and multi-resolution properties of the morphological pyramid structure, and bilinear interpolation, this algorithm can be implemented hierarchically and obtain the optimal parameters with QPSO. The experiment shows that this optimizing algorithm can do more effectively and more accurately work than Levenberg-Marquardt (LM) optimizing algorithm.

ABSTRACT: In this paper, we proposed multi-modal medical image registration method, which introduces mathematical morphology into image processing, and makes full use of the non-linear multi-resolution of the morphology Haar wavelet; the image pyramid is constructed according to the different resolution, which can enhance the speed of the registration. Genetic algorithm (GA) is used to accurately obtain the optimal parameters. In addition, we use the gradient of mutual information to measure the similarity between the two images. Introducing the gradient information into the mutual information makes the registration algorithm more robust and more accurate. The experiment shows that this algorithm can do more effectively and more accurately work than the algorithm based on Haar wavelet.
