    ABSTRACT: To observe the effect of mesenteric lymph duct ligation on free radical and inflammatory mediator of myocardium with severe hemorrhagic shock in rats at different period, and explore the effect of intestinal lymphatic pathway on myocardium injury pathogenesis in shock rats. 78 male Wistar rats were divided into the sham group, shock group and ligation group. The model of serious hemorrhagic shock was established in shock group, ligation group, and mesenteric lymph was blocked by ligating mesenteric lymph duct in ligation group after resuscitate. All rats were executed and taken out heart making for homogenate of 10 percent to determine the MDA, SOD, tumor necrosis factor-alpha (TNFalpha), interleukin-6 (IL-6), myeloperoxidase (MPO), NO and NOS at after shock 90 min, after transfusion and resuscitate 0 h, 1 h, 3 h, 6 h, 12 h and 24 h etc. different times, and the expression of inducible nitric oxide synthase (iNOS) mRNA in myocardium was detected by RT-PCR. The contents of MDA, TNFalpha, IL-6, MPO, NO, NOS and iNOS expression in myocardium of shock group were rising after transfusion and resuscitate, and that was higher level at 3 h to 12 h, and that was significantly higher than sham group, the activity of SOD was significantly lower than sham group. The contents of MDA, TNFalpha, IL-6, MPO, NO, NOS and iNOS expression in myocardium of ligation group were significantly lower than that of shock group at sameness points, and the SOD activity was higher. The mesenteric lymph duct ligation and blocking mesenteric lymph could reduce the PMN detaining, decrease the discharging of TNFa and IL-6, reduce the NO and expression of iNOS mRNA, and reduce the releasing of free radical and consuming of SOD.

    ABSTRACT: To observe the effect of ligation of mesenteric lymph duct on release of oxygen free radical and inflammatory mediator in lung of rat with serious hemorrhagic shock at different periods, and explore the role of intestinal lymphatic pathway on acute lung injury (ALI). Seventy-eight male Wistar rats were randomly divided into the sham operation group, shock group, and ligation group. The model of serious hemorrhagic shock was reproduced in shock group, ligation group, and mesenteric lymph flow was occluded by ligating mesenteric lymph duct in ligation group after resuscitation. Eight rats were sacrificed and the lung was harvested and homogenized for the determination of malondialdehyde (MDA), superoxide dismutase (SOD), tumor necrosis factor-alpha (TNF-alpha), interleukin-6 (IL-6) and myeloperoxidase (MPO) activity at 90 minutes after shock, 0, 1, 3, 6, 12 and 24 hours after transfusion and resuscitation. The contents of MDA, TNF-alpha, IL-6 and MPO activity in lung homogenate of shock group were elevated after transfusion and resuscitation, and the levels were higher significantly at 3-12 hours, than those of the sham operation group, while the activity of SOD was significantly lower than that of sham operation group (P<0.05 or P<0.01). The contents of MDA, TNF-alpha, IL-6 and MPO in lung homogenate of ligation group at 3, 6, 12 and 24 hours after resuscitation were significantly lower than those of shock group at the same time points, and the SOD activity were higher (P<0.05 or P<0.01). The results demonstrate that the ligation of mesenteric lymph duct could ameliorate ALI in rats with serious hemorrhagic shock, and its mechanism might relate with reduction of sequestration of neutrophils, thus reduce the release of TNF-alpha and IL-6, and also reduce the release of oxygen free radical and consumption of SOD. Therefore mesenteric lymphatics might play an important role in pathogenesis of ALI.

    ABSTRACT: To explore the interferential effect of ligustrazine injection (LI) on lymph circulation in rats with acute microcirculation disturbance (AMD). Sixteen Wistar male rats were divided into experimental and control groups. AMD was done by Dextran 500 injection from jugular vein and effects of LI on lymph circulation were observed in these rats by lymphatology methods. During AMD phase, the contractility of mesenteric lymphatic (ML) , the intestinal lymph flux, lymphocytes out put were obviously decreased, and there was little monocytes in the lymph fluid, but the lymph fluid viscosity was increased. After treatment of LI, the contractility of ML, the intestinal lymph flux, lymphocytes out put was obvious increased. There was a lot of monocytes in the lymph fluid and the lymph fluid viscosity was decreased. There were signifcant difference compared with control group (P < 0.05). LI can influence the recovery of AMD though enhancing the contractility of ML and the transport function of lymph circulation and decreasing lymph fluid viscosity.

    ABSTRACT: To observe the effect of mesenteric lymph duct ligation on nitric oxide (NO) and its expression in lung in rats with serious hemorrhagic shock at different periods, and explore the role of intestinal lymphatic pathway in acute lung injury (ALI). Seventy-eight male Wistar rats were randomly divided into the sham operation group (n=6), shock group (n=42), and ligation group (n=30). The model of serious hemorrhagic shock was reproduced in shock group and ligation group. Mesenteric lymph was diverted by ligating mesenteric lymph duct in ligation group after resuscitation. All rats were executed and their lungs were harvested. NO and nitric oxide synthase (NOS) were determined in lung homogenate 90 minutes after shock, and 0, 1, 3, 6, 12 and 24 hours after resuscitation. The expression of inducible nitric oxide synthase (iNOS) mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR). The content of NO, activity of NOS and expression of iNOS mRNA in lung tissue in shock group were increased 3 hours after transfusion and resuscitation, and they became higher 6-12 hours after resuscitation. The values were significantly higher than those in sham operation group, 90 minutes after shock and 0 hour after transfusion and resuscitation (P<0.05 or P<0.01). The values were only increased 3 hours and 6 hours after transfusion and resuscitation, and all the values on 6, 12 and 24 hours after resuscitation were significantly lower in the ligation group compared with those in shock group at the same time points (P<0.01 or P<0.05). The results demonstrate that the ligation of mesenteric lymph duct could reduce the NO and expression of iNOS mRNA in the lung tissue, thus ameliorate ALI in rats with serious hemorrhagic shock. Mesenteric lymph might play an important role in the pathogenesis of ALI.

    ABSTRACT: To explore changes in lymph circulation in rats with experimental disseminated intravascular coagulation (DIC). Thirty-two male Wistar rats were randomly divided into two groups: DIC group and sham-operation group, with 16 rats in each group.In each group, 8 rats were used to observe the lymphatic circulation of intestinal mesentery, and the others were used to observe lymphatic dynamics. Experimental DIC model was replicated by injecting dextran 500 via jugular vein in Wistar rats. The changes in lymph circulation were observed in these rats with lymphology methods. During the phase of DIC, the contractility of mesenteric micro-lymphatic (ML), the intestinal lymph flux, lymphocytes output were obviously decreased, and there was a few monocytes in the lymph fluid, but the lymph fluid viscosity was increased. After treatment with normal saline (NS), the contractility of ML, the intestinal lymph flux, lymphocytes output were obviously increased, and there was a large amount of monocytes in the lymph fluid, with significant difference compared with sham-operation group (all P<0.05). Moreover, the lymph fluid viscosity was decreased compared with sham-operation group (P<0.05). The disturbance in lymph circulation mainly includes depression of contractility of lymphatics, the transport function of lymph circulation, and increased lymph fluid viscosity.
